The nucleoside analogue gemcitabine (2', 2-difluorideoxycytide) is potential against a wide variety of solid tumors and considered to be one of the most active drugs in the treatment of non-small cell lung cancer (NSCLC). In this study, we investigated the signals of gemcitabine-induced apoptosis, especially in point of caspase pathway in A549. We exposed A549 cells to gemcitabine for dose/time dependent manner and the results showed that gemcitabine induced apoptotic cell death in a time/dose-dependent manner. We also treated to gemcitabine and Z-VAD-fmk as a pan-caspase inhibitor for 24 hours. Gemcitabine alone induced 35.3% cell death, and co-treatment with gemcitabine and Z-VAD-fmk induced 15.1% apoptotic cell death. Our results demonstrated that Z-VAD-fmk as a pan-caspase did not completely block the gemcitabine-induced apoptosis. Western blotting analysis showed that gemcitabine increased caspase-3, active caspase-8, p21 and p53 protein expressions in A549. Co-treatment with Z-VAD-fmk completely blocked caspase-3 and active caspase-8 protein expressions, but did not change the level of p21 and p53 protein expressions. Our data indicate that gemcitabine induced apoptosis through caspase-dependent and -independent pathways in A549.

Chondrocytes and synovial fluid derived markers are used to monitor for osteoarthritis (OA). Specific inhibitors, known as tissue inhibitors of metalloproteinases (TIMP), regulate the proteolytic activity of matrix metalloproteinases (MMP). This study investigated whether MMP and TIMP levels were altered in synovial fluid and cartilage following the experimental induction of OA in canines. Twenty mature beagle dogs underwent a unilateral surgical transection of the cranial cruciate ligament and the medial collateral ligament as well as a medial meniscectomy. Matrix metalloproteinase-2 and MMP-9 levels were assayed using Western blot and TIMP-2 levels were measured with enzyme-linked immunosorbent assays four weeks after OA induction. Increased MMP-2 expression was observed in chondrocytes isolated from cartilage following OA induction, but MMP-9 expression decreased. Matrix metalloproteinase-2 and MMP-9 levels in synovial fluid from the OA induced joint significantly increased compared to those of the sham group. Tissue inhibitors of metalloproteinase-2 concentrations were higher in chondrocytes from the OA cartilage, yet TIMP-2 remained lower in the synovial fluid of OA. This suggests the elevated release of MMP- 9 over MMP-2 into the synovial fluid following the cartilage degradation-related death of chondrocytes after OA. Osteoarthritis can be further deteriorated by increased MMP activity in the synovial fluid because TIMP-2 exist low concentration into the extracellular matrix. As a result, MMP activity, particularly MMP-9 activity, can be useful as a biomarker in diagnosing and monitoring the early stages of canine OA.

Proteomics is a useful approach to know protein expression, post-translational modification and protein function. We investigated the protein expression pattern and identity in chickens fed with the tissue culture medium waste after harvest of Korean wild ginseng (TCM-KWG) (Panax ginseng). Two groups (n=60/group) of day old broiler chickens were administered with 0 (control) and 0.8% (treatment) TCM-KWG through drinking water. After 5 weeks, we examined the protein expression pattern of fibularis longus and superficial pectoral muscle by Two-dimensional electrophoresis analysis. Interestingly, TCM-KWG treatment significantly increased five spot's density, and markedly reduced five spot's density in the muscles. We identified 10 proteins (desmin, myosin light chain 1, heat shock 25 kDa protein, collapsin response mediator protein-2A, alpha enolase, vimentin, actin alpha 1, my023 protein, pyruvate kinase and troponin T) by the matrix-assisted laser desorption ionization time of flight (MALDI-TOF).
Evaluation of Bactericidal Capacity of Silver Ion against Bovine Mastitis Pathogens

Bovine mastitis, defined as an inflammation of the mammary gland, is usually associated with bacterial infection. Thus, treatment and control of mastitis relies primarily on antimicrobial therapy. This study investigated the bactericidal actions of silver ion against causing various bovine mastitis pathogens. Minimal inhibitory concentrations (MICs), minimum bactericidal concentrations (MBCs) and bactericidal activity times and concentrations of silver ion against pathogens were determined. The effect of silver ion on bacterial morphology was studied by scanning electron microscopy (SEM). The MICs and MBCs of silver ion for various bacteria strains ranged from 1.9-15.6 μg/ml. SEM images indicated formation of a pit, distortion and disruption of cell walls in silver treated Staphylococcus aureus and Escherichia coli. The results demonstrate that silver ion has a bactericidal capacity against causing various pathogens of bovine mastitis and suggest that silver ions may be exploitable as a therapeutic/preventative tool of bovine mastitis.
산삼배양액 급여에 따른 육계의 생산성 및 질병 저항성 효과

The large amount of tissue culture medium (TCM), which contains some of the active secretory components of Korean wild ginseng (KWG; Panax ginseng) such as saponins, is usually discarded after harvest of KWG. The present study was aimed to investigate the efficacy of oral administration of the TCM-KWG on growth performance and diseases resistance in broiler chickens. A day old broiler chickens randomized in 6 groups (n = 60/groups) were administered orally with 0, 2, 4, 8, 16 and 32 mL/L TCMKWG through drinking water for 5 weeks and examined the change of weight gain, feed intake and blood components. Also, five weeks old broiler chickens (n = 15/groups) were challenged orally with Salmonella (S.) gallinarum and investigated the mortality in broiler chickens. An average weight gain and feed intake significantly didn't change in TCM-KWG administration groups as compared to control group. The concentration of calcium (Ca), phosphate (Pi) and potassium (K) in serum were increase by TCM-KWG administration in broiler chickens. We also found that oral administration of TCM-KWG through drinking water significantly reduced the mortality in broiler chickens experimentally infected with virulent S. gallinarum. The results of this study indicated that TCM-KWG administration may elevate the resistance on disease and improved the skeleton formation and body homeostasis of chickens, and TCM-KWG can be used as a cost-effective and environmentally alternative additives to control of the disease and growth.

TCM-KWG treatment significantly increased two spot's density, and markedly reduced one spot's density in the muscles. We identified 3 proteins (heat shock protein 90-alpha, myosin binding protein and cofilin 2) by the ESI-MS/MS (Q-TOF2, Micromass). These results demonstrate that TCM-KWG treatment may play a protection role against physiological stress in pigs,like as increased heat shock protein 90-alpha.

The purpose of this study is to determine the antimicrobial effect of tea tree oil a against pathogens relate to acne. Tea tree oil extracted from Australian Melaleuca alterifolia has been studied extensively for their antimicrobial properties against different type of bacteria and fungi. Tea tree oil has been reported to have antibacterial, antiinflammatory and antitumor activities, among others. By evaluating the antibacterial effect of Tea Tree Oil of Australia, the MIC value of Tea Tree oil against P. acnes was 0.05% and 0.5% against aerobic bacteria. This study showed that Tea Tree Oil has antibacterial effect against bacteria which induce acne. Tea Tree Oil had better antibacterial effect against P. acnes compared to aerobic bacteria. It is promising to develop a nature-based functional material for makeup product, which can cure and prevent acne.
Application of Silver Ion for Clinical Mastitis in Holstein Cows

Mastitis is an inflammation of the mammary gland that develops as a response to pathogenic microorganisms. Somatic cell count (SCC) and bacteriological analysis are the most accepted tools to determine udder health. This study evaluated the effects of intra-mammary gland infusion of silver ion against clinical mastitis in Holstein cows. Silver ion (20 μg/ml) was infused through the intra-mammary gland in quarters having clinical mastitis and milk was collected to determine SCC, and levels of lactoferrin and bovine serum albumin (BSA). Silver ion infusion decreased udder inflammation, firmness and swelling, and reduced clots, BSA, lactoferrin and SCC in milk. However, milk yield and circulating blood cells remained unchanged. The silver ion-mediated reductions of BSA and SCC indicate reduced inflammation and bacterial activity in silver ion-treated mammary glands in Holsteins with mastitis, which may be exploited in a curative strategy.
