Analysis of gene expression during odontogenic differentiation of cultured human dental pulp cells

Objectives: We analyzed gene-expression profiles after 14 day odontogenic induction of human dental pulp cells (DPCs) using a DNA microarray and sought candidate genes possibly associated with mineralization. Materials and Methods: Induced human dental pulp cells were obtained by culturing DPCs in odontogenic induction medium (OM) for 14 day. Cells exposed to normal culture medium were used as controls. Total RNA was extracted from cells and analyzed by microarray analysis and the key results were confirmed selectively by reverse-transcriptase polymerase chain reaction (RTPCR). We also performed a gene set enrichment analysis (GSEA) of the microarray data. Results: Six hundred and five genes among the 47,320 probes on the BeadChip differed by a factor of more than two-fold in the induced cells. Of these, 217 genes were upregulated, and 388 were down-regulated. GSEA revealed that in the induced cells, genes implicated in Apoptosis and Signaling by wingless MMTV integration (Wnt) were significantly upregulated. Conclusions: Genes implicated in Apoptosis and Signaling by Wnt are highly connected to the differentiation of dental pulp cells into odontoblast.
Physician’s Attitude toward Treating Breakthrough Cancer Pain in Korea

Purpose: Adequate control of breakthrough pain is essential for patients with cancer. Managing breakthrough pain mainly depends on understanding the concept of breakthrough pain and the proper usage of rescue medication by physicians. This study aims to assess the attitudes and practice patterns of palliative physicians in managing breakthrough pain for patients in Korea. Methods: This study was based on data from the 2014 breakthrough cancer pain survey conducted by the Korean Society for Hospice and Palliative Care. One hundred physicians participated in the online survey. Among total 33 self-reported questionnaires, twelve items were selected in this analysis. Results: Rapid onset of action is the main influencing factor in selecting rescue opioids. Oral oxycodone (65%) and parenteral morphine (27%) are commonly used. A few physicians (3%) prefer to use transmucosal fentanyl. The percentage of physicians prescribing oral oxycodone due to its rapid onset of action is just 21.5%, whereas the percentage of physicians using parenteral morphine is 81.5%. Two thirds of respondents (66%) answered that breakthrough pain is not well controlled with rescue medications. Conclusion: There is a gap between the needs of physicians in terms of the perceived difficulties of managing breakthrough cancer pain and their practice patterns selecting rescue medications.

In this study human bones and teeth, excavated from the Myeongam-risite in Asan, Chungcheongnam-do Province, have been analysed by nuclear DNA typing and mitochondrial DNA sequencing methods. Twenty-one samples of long bones and twenty-seven samples of teeth from twenty-one individuals were collected and analysed. Among these thirteenteeth were successfully subjected to nuclear DNA extraction, quantification, and PCR(Polymerase Chain Reaction) amplification. Silver STR III (D16S539, D7S820, D13S317) multiplex PCR method was used in this study for a short tandem repeat (STR) analysis. Mitochondrial DNAs of tooth samples were also amplified and sequenced by a DNA sequencer. These analyses show that a sample from Burial no. 29 and one from Burial no. 38(right) possessed the same maternal inheritance. This may suggest that the Myeongam-ri cemetery was used by a kin group for a relatively long period of time.
Usefulness of Additional SPECT/CT Identifying Lymphatico-renal Shunt in a Patient with Chyluria

Lymphoscintigraphy is known to be a usefuland non-invasive modality for the evaluation of lymphaticabnormality. However, lymphoscintigraphy has limitationsin evaluating chyluria because of its lack of anatomicalinformation. Additional single-photon emission computedtomography (SPECT) combined with computed tomography(CT) was considered to be potentially helpful indetecting the abnormal lymphatico-renal communication. A 20-year-old male patient was referred to our hospitalfor evaluation of recurrent chyluria. During the third recurrenceof chyluria, additional SPECT/CT along withlymphoscintigraphy was performed for evaluation. Fromthe combined SPECT/CT images, lymphatic drainage ofradiotracers to the kidney was well visualized, helpingdiagnosis of a patient with chyluria.
