        In order to understand the relationship between lipid peroxide in blood serum and various diseases supposed to be related to abnormal lipid metabolism, lipid peroxides in human patient serums were measured by fluorometry. The results obtained were folllowing: In normal human, the serum lipid peroxides showed a slight increase by age, but not much difference by sex. Lipid peroxides in blood serum of various patients with hypertention, diabetes, hepatitis, arteriosclerosis, stomach cancer were higher than those of the normal human. From these results obtained, the autors concluded that lipid peroxides in blood serum appeared to be correlated to various diseases supposed to be realted to lipid peroxidation.

        자동산화 Methyl Linoleate 가 Mouse 혈청의 효소활성에 미치는 영향 ( 급성 독성 )

        In order to investigate the acute toxicity of autoxidized methyl linoleate(AOML) on the activity of serum enzymes in the mouse, we administered once 0.45ml of AOML to ICR strain mouse by using stomach tube. The following results were obtained: The total lactic dehydrogenase(LDH) activities in the serum of AOML group were generally increased than those of mormal group. According to electrophoresis, the activities of LDH, were increased while those of LDH, were decreased. The activities of glutamic oxaloacetic transaminase(GOT), glutamic-pyruvic transaminase(GPT) and α-amylase in the serum of AOML group were increased more than those of mormal group. The activities of alkaline phosphatase in the serum of AOML group were increased but those of isozyme were not confirmend in the normal and AOML group. In the serum protein of AOML group, albumin was increased, on the other hand r-globulin was decreased. At the peripheral blood slide smear, lymphocytes were significantly decreased but neutrophils were increased and the morphological change of erythrocytes was observed. From these results we conclude that the AOML fed to mouse influences on the activity of various serum enzymes and blood cells in the mouse.

        질환자에서의 혈청 과산화 지질

        Lipoprotein Lipase 의 활성에 미치는 인삼 Saponin 의 영향

        In order to investigate the effect of ginseng saponins on the activity of lipoprotein lipase, it was attempted to conform the enzymatic hydrolysis of chylomicron with post-heparin induced plasma lipoprotein lipase of normal rabbit in vitro. And the activity of lipoprotein llipase was determined by the quantitative determination of liberated free fatty acids on the hydrolysis of chylomicron. As the result, it was observed that the ginseng ssaponins accelerated the hydrolysis of chylomicron by post-heparin plasma in vitro. And the optimum concentration of ginseng saponins for the activity of the lipoprotein lipase in the 」% bovine serum albumin was 10^-4%. But ginseng saponins on the hydrolysis of chylomicron was influenced by the presence and the absence of albumin. And the optimum concentration of albumin and Na-cholate on the activity of lipoprotein lipase was each of the 10^-6% albumin and 5mM Na-cholate. From these results, it seems that ginseng saponins might stimulate the intravascular hydrolysis of chylomicron.

      • Linoleic Acid 의 광산화와 항산화제의 방어작용

        Linoleic acid(LS)의 광산화와 이에 대한 각종 항산화제들의 억제효과를 in vitro에서 조사하였다. 공기의 공급하에서 백색광과 적색광에 의한 LA의 광산화 반응은 광증감제인 methylene blue를 첨가했을 때만 진행되었으며, 백색광을 조사했을 때는 적색광을 조사했을 때 보다 약 2배의 산화 증가를 나타내었다. LA의 ethanol 용액에 BHA(butylated hydroxyanisole), BHT(butylated hydroxytoluene), dl-$\alpha$-Toc(dl-$\alpha$-tocopherol) 등을 최종 농도가 0.1%되도록 첨가했을 때 LA의 광산화는 BHA의 경우에는 첨가농도에 비례하여 억제되었으며, dl-$\alpha$-Toc의 경우는 첨가농도가 0.1% 이상일 때만 억제효과를 나타내었다. 한편, methylene blue를 첨가하여 광산화 시켰을 때 BHA는 산화되지 않았으나, dl-$\alpha$-Toc은 $10^{-3}%{\sim}10^{-1}%$ 사이에서 산화가 일어났으며, BHT는 첨가농도에 비례하여 산화되었다. LA의 chloroform-ethanol 용액에 $\beta$-carotene, dl-$\alpha$-Toc, BHA, BHT를 각각 최종 농도가 $10^{-4}%$되도록 첨가했을 때 LA의 광산화 억제효과는 $\beta$-carotene>dl-$\alpha$-Toc이며, BAT와 BHT는 억제효과를 나타내지 않았다. The photooxidation of linoleic acid (LA) in the presence of methylene blue as a photosesitizer and inhibitory behavior of various antioxidants in vitro were investigated. Under air supply, LA that irradiated with visible light or red light was photooxidized only by addition of methylene blue. In the presence of methylene blue, the degree of photooxidation of LA with visible light was about twice as much as that with red leght. The photooxidation of LA in ethanol solution to which BHA (butylated hydroxyanisole), BHT (butylated hydroxytoluene) and dl-$\alpha$-Toc (dl-$\alpha$-tocopherol) was added to be 0.1% in final concentration, BHA was inhibited proportionally to the concentration of BHA, but in addition to dl-$\alpha$-Toc (dl-$\alpha$-tocopherol) was inhibited when it's concentration was higher than 0.1%. When methylene blue was added to ethanol solution, BHA itself was little oxidized, but dl-$\alpha$-Toc was at the concentration between $10^{-3}%{\sim}10^{-1}%$ and BHT was highly oxidized according to the increase of concentration. In the case of antioxidants was added to chloroform-ethanol solution containing LA to be $10^{-4}%$ in final concentration, the photooxidation of LA was significantly inhibited by both $\beta$-carotene and dl-$\alpha$-Toc but $\beta$-carotene was effective more than dl-$\alpha$-Toc. Also BHA and BHT were not effective.

      • 인삼 추출물이 흰 쥐의 dl-$\alpha$-Tocopherol의 흡수 및 혈청 Lipid Peroxide 양에 미치는 영향

        To investigate the effect of ginseng extracts on the absorption of total tocopherol, dl-$\alpha$-Tocopherol solution and ginseng extract solution were fed by stomach tube to rats and total tocopherol level and lipid peroxide value in serum were determined. The results compared with control were as followed. (1) Critical micelle concentration (CMC) of ginseng saponin was about 1.8% and that of ginseng extract solution in the presence of about 1 mM of dl-$\alpha$-Tocopherol was decreased to 0.14%. (2) The serum total tocopherol level of test group administered ginseng extracts was increased to 1.52 times. (3) The lipid peroxide value of test group administered ginseng extracts was decreased to about halves in serum. dl-$\alpha$-Tocopherol 흡수에 미치는 인삼 추출물의 영향을 규명하기 위하여 dl-$\alpha$-Tocopherol과 인삼 추출물 수용액 일정량을 흰 쥐에 경구 투여하여 혈청에서의 vitamin E 양과 lipid peroxide 양을 비교 정량한 결과는 다음과 같다. 인삼 사포닌 수용액의 CMC는 약 1.8%였으며, 1 mM dl-$\alpha$-tocopherol이 존재할 때 인삼 saponin의 CMC는 0.14%로 크게 저하하였다. 혈청에서의 total tocopherol 양은 dl-$\alpha$-tocopherol과 인삼 추출물은 같이 투여하였을 때 흡수가 증가하여 dl-$\alpha$-tocopherol만을 투여하였을 때 보다 약 1.52배 증가되었으며, lipid peroxide 양은 1/2로 감소되었다.

        자동산화 Methyl Linoleate가 Mouse간장의 효소활성에 미치는 영향

        자동 산화유가 mouse의 간장 효소에 미치는 영향을 구명하기 위하여 autoxidized methyl linoleate의 일정량을 mouse에게 1일 1회 20일 동안 경구 투여하여 간에서의 효소 활성을 조직화학적 및 생화학적 방법으로 검토한 결과는 다음과 같다.<br/> 실험군의 간에서의 POV, COV 및 TBA값은 정상군에 비해 크게 증가하여 AOML이 많이 흡수되어 있음을 알 수 있었다.<br/> 실험군의 간에서의 phospholipid, triglyceride 및 total cholesterol의 양은 정상군에 비해 증가하였다.<br/> 실험군의 간에서의 acid phosphatase, alkaline phosphatase의 활성은 정상군에 비해 증가하였으며, ATPase의 활성은 감소하였다.<br/> 실험군의 간에서의 형태학적 변화는 정상군에 비해 핵산의 감소, 지질의 축적 및 간 세포의 손상이 있었다.<br/> 이상의 결과로부터 mouse에 경구 투여한 자동 산화 methyl linoleate는 간에서의 효소 활성과 간의 형태학적 변화에 크게 영향을 주고 있음을 알 수 있었다. In order to investigate the effect of autoxidized oil on the enzyme activity in the mouse liver, we administered the fixed dosage of autoxidized methyl linoleate (AOML) to mice once per day for 20 days by using stomach tube and investigated the enzyme activity with the histochemical staining method and biochemical analysis.<br/> The following results were obtained:<br/> The POV, COV and TBA value in the liver of AOML group were significantly increased than those of normal group. The phospholipid, triglyceride and total cholesterol in the liver of AOML group were slightly increased than those of normal group. The activities of acid phosphatase and alkaline phosphatase in the liver of AOML group were increased than those of normal group but ATPase activity was decreased in the AOML group. The decrease of RNA, accumulation of fat and damage of liver cells were observed as the morphological changes in the liver of AOML group.<br/> From the results obtained we conclude that the autoxidized methyl linoleate influenced upon the various enzyme activity and the morphological changes in the mouse liver.

      • 생체 조직의 Estradiol Receptor 정량

        생체 조직에서의 estradiol receptor 양을 간편하고 정확하게 정량하기 위하여 종래의 $^3H$-estradiol을 사용한 dextran coated charcoal 방법을 개선한 결과는 다음과 같다. 새로운 방법으로 쥐 고환 조직내의 estradiol receptor 함량은 23.8 fmol/mg protein으로 비교적 높았고, 근육 조직에서는 0.57 fmol/mg protein으로 비교적 낮게 나타났다. 사람의 유암조직의 estradiol receptor 함량은 유암이 아닌 다른 종양성 환자의 유방 조직에서의 함량보다 높게 나타났다. Scatchard plot 상에서의 free form에 대 한 bound form의 비와 estradiol receptor 농도와의 linearity(r=0.925-0.977, P<0.01) 가 향상되었다. 따라서, 유방암 조직만의 estradiol receptor 양을 정량함으로써 유방암의 진단과 치료에 쉽게 이용할 수 있다. To quantitative the estradiol receptor in biological tissue, authors had modified the conventional dextran coated charcoal method using $^3H$-estradiol. The amount of estradiol receptor in the mouse testis was 23.8 fmol/mg protein, which is significantly higher than the value of the mouse muscle tissues (0.57 fmol/mg protein). In human breast cancer tissue, the amount of estradiol receptor was higher than that of chronic mastitis. The correlaton between the concentraion of estradiol receptor and the ratio of bound to free form (B/F) showed excellent linearity (r=0.925-0.977, P<0.01) by Scatchard plot with the modified method, therefore these results strongly suggested that the quantitation of estradiol receptor In breast cancer tissue would be useful for the diagnosis and treatment of breast cancer.
