Conformational Transitions in solubilized Bacteriorhodopsin

Bacteriorhodopsin has been the subject of intensive studies during the past decade and, structurally, it is the best known among the integral membrane proteins. The present study focusses on the conformational transitions of delipidated and detergent-solubilized bacteriorhodopsin under various conditions. A UV spectrophotometer was used for following the thermal transition of this protein as a function of pH as well as the concentrations of this protein as a function of pH as well as the concentrations of chemicals added. Below pH 7.5, the thermal transition curves showed two discrete steps in helix-random coil transition process while only one step was apparent above this pH. The thermal transition observed in the presence of methanol, ethanol and propanol, respectively, showed more complex behavior. While increasing alcohol concentration decreased baselines at lower temperatures for all three of these alcohols, the transition temperature and the transition range were different for different alcohols. The thermal transition behavior of the purple membrane itself was found to be complex. The thermal transition behavior of the delipidated and detergent-solubilized bacteriorhodopsin in compared with that of purple membrane as well as with that of non-membrane proteins.

PURPOSE : To report a case of Retinal capillary hemangioma initially misdiagnosed to idiopathic choroidal neovascularization CASE REPORTS : A 23-year-old female with a 6-month history of decreased vision in the left eye was referred to our hospital on June 17, 2003. A dilated fundus examination of the left eye was revealed a juxtapapillary subretinal lesion with adjacent exudative retinal detachment and retinal folds. A fluorescein angiogram demonstrated early hyperfluorescence and late leakage of the dye. Based on these results, a diagnosis of Idiopathic choroidal neovascularization was made. No treatment was performed. But, a new lesion of capillary hemangioma was found in the fundus examination on April 9,2004. Therefore, we confirmed the juxtapapillary subretinal lesion to a juxtapapillary capillary hemangioma. Medical investigations revealed no systemic involvement. Treatment with laser photocoagulation to the peripheral capillary hemangioma was performed. CONCLUSION : Juxtapapillary capillary hemangioma usually growths exophytically, and an endophytic form of juxtapapillry capillary hemnagioma has not been reported in Korea. However, the endophytic juxtapapillary capillary hemangioma as this case can be misdiagnosed to a choroidal neovascularization.
pH/온도 자극 응답성 하이드로젤에 고정화된 페니실린 G 아실레이즈

페니실린 G 아실레이즈를 pH나 온도에 민감한 자극 응답성 상전이 하이드로젤에 고정화 하였다. 95 몰 퍼센트의 N-isopropylacrylamide와 5 몰 %의 NN′-dimethylaminopropylmethacrylamide의 모노머 조성과 소량의 가교제를 포함한 양이온성 하이드로젤 미립구에 효소는 역서스펜션 중합에 의하여 물리적으로 고정화 하였다. 고정화 효소의 온도 의존성 및 pH 의존성과 같은 속도론적 성질은 고정화되지 않은 성질과 많이 달랐다. 이는 pH/온도 자극 응답성 하이드로젤 담체가 고정화 효소의 반응속도에 매우 큰 영향을 미친다는 것을 암시한다.
