        Heparin Derivatives Coated Drainage Catheter for Inhibition of Thrombus Formation

        A problem frequently encountered in the use of drainage catheters is that thrombus deposition tends to clog the drainage catheters, which destroy the efficacy of the device. In this study, a new heparin-releasing drainage catheter for inhibition of thrombus formation was proposed. The heparin-deoxycholic acid (heparin-DOCA) conjugate in the polyurethane solution was introduced to the drainage catheters using a simple casting method. From the results, the internal thickness of the coated drainage catheter was 15 μm and the coated surface of the catheter was even without defects. The pressures of the coated catheter measured at 7, 10, and 14 days were 10.8 ± 6.08, 25.0 ± 9.17, and 29.5 ± 12.02 mmHg, respectively. These values are relatively lower than the value of the bare catheter, showing that the pressure value of the bare catheter at 14 days was 198.5 ± 16.25 mmHg. This can be explained by the fact that the heparin-DOCA was released effectively from the catheter more than 2 weeks after inserting the catheter, which prevented thrombus deposition or fibrin clotting. In conclusion, the heparin release system is more effective for inhibiting thrombus formation than heparin-immobilized system.

        Evaluation of the Oral Absorption of Heparin Conjugated with Sodium Deoxycholate as a Facilitating Agent in GI Tract

        The oral delivery of heparin is the preferred therapy in the treatment of patients with a high risk of deep vein thrombosis and pulmonary embolism. New conjugates of heparin and sodium deoxycholate were synthesized in order to enhance the heparin absorption in the GI tract. After oral administration of DOC-heparin, the concentration in anti-FXa assay was increased with increasing amount of coupled DOC. The maximum concentration of DOC-heparin VIII conjugate was 3.3±0.5 IU/mL at an oral dose of 10 mg/kg, which was 3-fold higher than the baseline level. Finally, DOC coupled to heparin greatly enhanced the absorption of heparin in the GI tract, and this enhancing effect was not induced by changing the tissue structure of the GI wall.

        항응고제 코팅기술을 이용한 PVC와 실리콘 제품의 고기능성 흡입 배액용구의 개발

        Advanced anticoagulant suction drainage was prepared by anticoagulant coating with the hydrophobic multicomponent heparin conjugates, and named as MCK-Hepafilm (15K). The coating surface was analyzed by the staining test of azure-A and scanning electron microscope (SEM), which shows excellent coating effect on silicone as well as PVC. The coating condition of silicone tube was optimized to 3.0 wt% concentration of coating solution and the solvent casting temperature to 35 oC. In vitro anticoagulant effect is confirmed by observation of platelet adhesion, blood coagulation and adhesion of bacteria. Platelet adhesion on coating surface of PVC and silicone drainage tube was remarkably reduced, and completely prevented from whole blood clots of rabbit. The adhered number of Escherichia coil (E. coil) bacteria on coated surface of PVC drainage tube was reduced with 7.2?.0 CFU/cm2 as compared to the non-coated surface of 10.1?.2 CFU/cm2. Rabbit experiment has been carried out for the MCK-Hepafilm (15K) coated suction drainage device. The efficacy of the MCK-Hepafilm coat was examined by measuring the debris weight forming clogging in the tube (MCK-Hepafilm coated & uncoated) and analyzing the change in blood flow pressure through it, over a period of time, after insertion in the abdomen of the rabbit. In the coating tube, the internal resistance of PVC was reduced, and also the debris weight of silicone was reduced to 315.5?.4 mg as compared with non-coated 555.7?0.6 mg. The obtained results indicate the effectiveness of this MCK-Hepafilm (15K) coating technology for blood contacting medical devices including suction drainage devices in improving their hemocompatibility.

        소수성 다중복합 헤파린 결합체로 코팅한 항혈전성 혈관용 카테타의 평가

        An antithrombogenic angio-catheter was developed by coating hydrophobic multicomponent heparin conjugates, and named as MCK-Hepafilm (2K). MCK-Hepafilm (2K) using polyacrylic acid and octadecylamine cross linked with heparin conjugates, were prepared by Mediplex Corp. Korea. It has good organic solvent solubility for making an antithrombogenic coat on different medical devices. Coating both inside and outside of a polyurethane angio-catheter with MCK-Hepafilm (2K) showed excellent and stable coating, which was confirmed by SEM and the coloring test with azure-A. Varying the concentration of the coating solution can control the coating thickness: for 1.0, 3.0, and 5.0 wt%, thickness of 0.2, 0.6, and 1.1 mm were obtained, respectively. The coated surface had reduced contact angle and skin friction of rat was reduced to 152.7 g compared to 362.7 g of bare surface. Platelet adhesion was remarkably reduced, and whole blood clots were completely prevented in vitro condition. Safety and toxicity of coated angio-catheter have passed for variety of standard toxicity tests such as ISO and OECD guideline with 7 items, which were evaluated by Korea Test Laboratory (KTL). Dog experiment has been carried out for the MCK-Hepafilm (2K) coated angio-catheter (5F). The amount of thrombus formed on coated catheter was greatly reduced (103.7?9.9 mg) compared to non-coated catheter (374.6?1.6 mg). With these successful results, we propose that the MCK-Hepafilm (2K) coating technology can be applied as a potent platform technology for hemocompatibility of blood-contacting medical devices, including angio-catheter.

        River cross-section measurement data is one of the most important input data in research related to hydraulic and hydrological modeling, such as flow calculation and flood forecasting warning methods for river management. However, the acquisition of accurate and continuous cross-section data of rivers leading to irregular geometric structure has significant limitations in terms of time and cost. In this regard, a primary objective of this study is to develop a methodology that is able to measure the spatial distribution of continuous river characteristics by minimizing the input of time, cost, and manpower. Therefore, in this study, we tried to examine the possibility and accuracy of continuous cross-section estimation by estimating the water depth for each cross-section through multiple linear regression analysis using RGB-based aerial images and actual data. As a result of comparing with the actual data, it was confirmed that the depth can be accurately estimated within about 2 m of water depth, which can capture spatially heterogeneous relationships, and this is expected to contribute to accurate and continuous river cross-section acquisition.

        글로벌인턴십 파견 전 학습이 현지적응에 미치는 영향: LMX의 조절효과를 중심으로

        This study is to identify the relationships among the perceived satisfaction of pre-departure learning, adaptation to the host country, and the level of leader-member exchange. A total of 637 participants who were working globally after supporting global internship program by Korean government were subjected to this study, 261 available questionnaires were gathered, and only 111 of whom had joined four types of training programs including job-related training, host country language, English language, and cross-cultural training were used to final statistics analysis in order to ensure reliability and validity. The results of hierarchical regression analysis showed that the perceived learning satisfaction of pre-departure learning was positively related to adaptation for global internship participants to the host countries. In particular, English language training and host country language training both had positive effects on local adaptation, while job-related training and cross-cultural training had not related to adaptation to the host country. The results were not matched with precedent studies which were examined the relationship between pre-departure cross-cultural learning and adaptation of expatriates of multinational corporations. The reason is that expatriates are having clear assignments from corporations before leaving the host country, while participants who are joined global internship program were having a role of complementary assignment rather than key tasks. Furthermore, the perceived LMX between supervisor and internship participant had a positive effect on the adaptation to the host country. This study also found the moderating effects of LMX with the perceived satisfaction of pre-departure learning and adaptation The results of this study have expanded a field of global human resource research by identifying the effectiveness of the pre-departure learning being aimed at the global internship participants, not to the expatriates. We also discussed limitations and further research directions.

        이 연구의 목적은 해외 인턴십 참여자를 대상으로 해외 인턴십의 특성과 글로벌 역량, 고용가능성 간의 관계를 규명하는데 있다. 그리고 이러한 연구 목적을 달성하기 위하여 해외 인턴십의 특성인 현장지향성과 경력개발지원이 글로벌 역량과 고용가능성에 미치는 영향을 규명하고, 해외 인턴십 특성과 고용가능성 간의 관계에서 글로벌 역량의 매개효과를 규명하는 것을 구체적인 목표로 설정하였다. 2014년과 2015년에 정부의 지원을 통해 해외 인턴십에 참여하고 있는 인턴을 대상으로 조사가 실시되었다. 자료의 수집은 조사대상이 해외에 근무 중이라는 특성 때문에 이메일과 SNS를 통해서 실시되었으며, 총 637부가 배부되어 278부가 회수되었으나 이중 불성실한 응답을 제외하고 252부의 설문이 최종분석에 활용 되었다. 그리고 변수들 간의 관계를 규명하기 위해서는 SPSS 21.0 프로그램을 활용하여 위계적 다중회귀분석을 실시하였다. 연구 결과, 첫째 해외 인턴십의 특성인 현장지향성과 경력개발지원은 고용가능성의 하위 변인인 내적유연성과 외적이동성에 모두 긍정적인 영향을 미쳐 고용가능성에 정(+)의 영향을 미치는 것으로 나타났다. 둘째, 글로벌 역량의 하위요인인 이문화수용력과 외국어능력은 모두 고용가능성의 하위요인인 내적유연성과 외적이동성에 정(+)의 영향을 미치는 것으로 나타났다. 마지막으로 글로벌 역량은 해외 인턴십의 현장지향성과 경력개발지원 중 경력개발지원과 고용가능성 간의 관계에서 매개효과를 가지는 것으로 나타났다. 따라서 고용가능성은 해외 인턴십의 현장지향성과 경력개발지원으로부터 정(+)적인 영향을 받으면서도, 글로벌 역량 함양을 통해서 고용가능성이 높아지는 등 글로벌 역량을 매개로 높아질 수 있음이 검증되었다. 이와 같은 연구결과를 토대로 다음과 같은 시사점을 얻을 수 있다. 먼저 해외 인턴십이 청년들의 글로벌 역량과 고용가능성에 긍정적으로 영향을 미치는 것이 검증되 었으므로 해외 인턴십은 더 확대될 필요가 있다. 특히 각 대학에서는 재학생들의 취업준비를 위해 글로벌 역량을 함양한다는 측면에서의 접근이 필요하다. 글로벌 역량의 향상을 통해 고용가능성이 증가하기 때문이다. 둘째, 해외 인턴십은 일터학습의 일환으로 좀 더 구조화되어 운영될 필요가 있다. 인턴십의 현장지향성이 글로벌 역량에 유의미한 영향을 미치지 못하는 것은 인턴십의 현장학습이 구조화되지 못한 채 운영되고 있는 것에 기인하기 때문이다. 마지막으로 글로벌 역량이나 고용가능성은 취업 여부 등과 함께 인턴십의 효과성을 측정하는 주요한 성과지표로 활용될 수 있다.         해외 인턴십의 현장지향성과 경력개발지원이 고용가능성에 미치는 영향: 글로벌 역량의 매개효과를 중심으로

        The purpose of this study is to investigate the relationship among field orientedness and career development support―the traits of overseas internship program, global competencies, and employability of overseas internship program participants. The data used for this study is collected from internees who have participated or are participating in the overseas internship program operated with government’s financial support. To enhance reliability of the results, “Internees” were restricted to those who participating in the internship or working at the same company after the internship as of the survey date. This study found firstly, field orientedness and career development support have positive effect both on internal flexibility and external mobility and therefore positively affect to employability. In addition, it was found that field orientedness was more influential to internal flexibility while career development support has a greater impact in external mobility. Secondly, cross-cultural adaptability and language skills―lower factor of global competency― has positive effect both on internal flexibility and external mobility. Third, among the sub-variables of global competency, language skills have positive effect only on external mobility among the sub-variables of employment possibility, and cross-cultural adaptability has a positive effect both on internal flexibility and external mobility. Forth, global competency has mediating effect on the relationship between career development support and employability but not on the relationship between field oritentedness of the overseas internship and employability. Therefore, it is verified that employability can be increased by the mediating effect of global competency while being positively affected by field orientedness and career development support of overseas internship. Several implications can be drawn from these research results. Firstly, overseas internship need to be expended and more structured as a part of workplace learning, universities also need a global approach to improve students’ employability. Secondly, it is necessary to induce multi-national corporations or companies of host country to participate in the program since internees are less exposed to cross-cultural. Lastly, global competency and employability can be used as important outcome indicator.
