        A Genome-wide Scan for Selective Sweeps in Racing Horses

        Using next-generation sequencing, we conducted a genome-wide scan of selective sweeps associated with selection toward genetic improvement in Thoroughbreds. We investigated potential phenotypic consequence of putative candidate loci by candidate gene association mapping for the finishing time in 240 Thoroughbred horses. We found a significant association with the trait for Ral GApase alpha 2 (RALGAP2) that regulates a variety of cellular processes of signal trafficking. Neighboring genes around RALGAP2 included insulinoma-associated 1 (INSM1), pallid (PLDN), and Ras and Rab interactor 2 (RIN2) genes have similar roles in signal trafficking, suggesting that a co-evolving gene cluster located on the chromosome 22 is under strong artificial selection in racehorses.

        소셜커머스 외식상품 이용자의 구매만족에 영향을 미치는 요인 -소비자 혁신성, 상품 혁신속성, 지각된 위험, 차별적 대우를 중심으로-

        The main characteristic of social commerce is a significantly discounted price for a product or service. This may contribute to the satisfaction of social commerce users, but discriminatory treatment because a customer uses the discount can be a source of dissatisfaction. Perceived risk due to the uncertainty of social commerce may also inhibit satisfaction. The purpose of this study was to identify what influences satisfaction in consumers who purchase restaurant products through social commerce, including consumer innovation, product innovation, perceived risk, and discriminatory experience. A total of 384 respondents who shopped for restaurant deals using social commerce participated through online survey. Chinese tourists who visited Korea participated. Discrimination due to using discounts had the greatest influence on satisfaction among social commerce users. Also, as perceived risk declined, awareness of the innovations in products contributed to higher satisfaction in purchasing restaurant products through social commerce. The managerial implications are discussed. 소셜커머스의 주요 특성인 제품이나 서비스의 할인 가격이 경제적으로 만족요인으로서 작용할 수 있는 반면에 할인된 가격으로 구매한 서비스에서 차별적 대우에 따른 부정적 인식과 경험은 불만족 요인이 될 수 있다. 소셜커머스를 통해 구매한 외식상품으로 인해 차별을 받은 경험이 불만족 사유로 제기되어 왔지만 이에 대해 학술적으로 검증되지 못했다. 본 연구에서는 소셜커머스 이용자들의 구매 만족도에 영향을 미치는 요인을 소비자 혁신성에 대한 욕구, 소셜커머스 상품의 혁신 속성, 불확실성으로 인해 지각된 위험, 차별적 대우를 중심으로 파악하였다. 이를 위해 소셜커머스를 통해 외식상품을 구매한 고객을 대상으로 온라인 설문조사를 실시하였다. 설문은 2013년 4월부터 2013년 5월까지 실시되었으며 총 390부가 수거되었으며, 이 중 불성실하게 응답한 9부를 제외하고 384부가 실증분석에 사용되었다. 소셜커머스 외식상품 구매 만족도에 영향을 미치는 요인으로 소셜커머스에서 외식상품 구매로 인한 차별적 대우가 가장 큰 영향을 주었으며 외식 상품 구매 시 지각된 위험과 상품 혁신 속성도 유의적인 영향을 주는 것으로 나타났다. 차별적 대우 정도가 낮을수록, 지각된 위험이 감소될수록, 그리고 상품의 혁신 속성을 많이 인식할수록 외식 상품 이용자의 구매 만족도가 높았다.

        1H, 15N, and 13C Resonance Assignments and Secondary Structure of the SWIRM Domain of Human BAF155, a Chromatin Remodeling Complex Component

        Mammalian SWI/SNF complexes are evolutionary con-served, ATP-dependent chromatin remodeling units. BAF155 in the SWI/SNF complex contains several highly conser-ved domains, including SANT, SWIRM, and leucine zipper domains. The biological roles of the SWIRM domain re-main unclear; however, both structural and biochemical analyses of this domain have suggested that it could mediate protein-protein or protein-DNA interactions during the chromatin remodeling process. The human BAF155 SWIRM domain was cloned into the Escherichia coli expression vector pMAL-c2X and purified using affinity chro-matography for structural analysis. We report the backbone 1H, 15N, and 13C resonance assignments and secondary structure of this domain using nuclear magnetic resonance (NMR) spectroscopy and the TALOS+ program. The secondary structure consists of five -helices that form a typical histone fold for DNA interactions. Our data suggest that the BAF155 SWIRM domain interacts with nucleosome DNA (Kd = 0.47 M).

        Effects of Infection Control Training on Dental Hygienists’ Health Beliefs and Practices of Infection Control

        This study aims to determine the changes in health beliefs and practices of dental hygienists on infection control after having received special training on that subject. The study population consisted of dental hygienists working at the dental institutions located in Seoul and Gyeonggi areas. The intervention group and the control group each had randomly assigned 26 dental hygienists. The intervention group received training courses on infection control once a week for 3 hours, for a total of 4 training sessions. We used a survey tool to find the changes between the two groups regarding their health beliefs and practices on infection control measures. The survey was conducted prior to the training sessions, 3 months and 2 weeks after the training. The study results revealed statistically significant increases in perceived sensitivity, perceived seriousness, and perceived benefits after the training courses in the intervention group (p<0.05). A statistically significant decrease in perceived barriers was also noted in the intervention group (p<0.05). Additionally, there was a statistically significant increase in the practice of infection control in the intervention group as compared to the control group (p<0.05). Therefore, we conclude that infection control training is crucial in bringing about positive changes to the health beliefs of dental hygienists, and a structured system is necessary for continuous infection management along with training in order to improve infection control practices.

        랫드에서 thioacetamide의 반복 투여로 유도된 간 손상에 대한 민들레 추출물의 효과

        This study was conducted to investigate of hepatoprotective effect of dandelion water extract (DWE) according to repeated administration of thioacetamide (TAA) induced hepatotoxicity in Spraque-Dawley rats. Thirty rats were randomly assigned to 5 groups; normal control, DWE-control, TAA-control (TAA injection during the feeding of normal diet), TAA&DWE600 (TAA repeated injection during the feeding of DWE 600 mg/kg BW), TAA&DWE1200 (TAA repeated injection during the feeding of DWE 1,200 mg/kg BW). Rats in DWE-control and TAA&DWE groups were treated with DWE (600 or 1,200 mg/kg BW daily) by gavage for 20 days (twice a day). All the rats in the TAA-control and TAA&DWE groups were repeated injection of TAA (100 mg/kg BW) into the abdominal cavity 3 days interval and 12 hrs later, all rats were sacrificed. At the same time, normal control and DWE-control groups were injected normal saline. In TAA&DWE groups, serum alanine and aspartate aminotransferase (ALT, AST) were significantly decreased and triglyceride (TG) synthesis was significantly increased compared to TAA group. As well as total billilubin and GGT were slightly decreased by the treatment of DWE. Lipid peroxidation (MDA) concentration was significantly decreased and hepatic GSH content was slightly or significantly increased in the TAA&DWE groups compared to TAA group. Hepatic antioxidative enzyme activities, such as GSH, GST, SOD and catalase were slightly or significantly elevated by the treatment of DWE. According to these results, When dandelion extract was long term supplied, it could be used as a potential protective material for a longer time liver damage by repeated adminstration of the TAA.

        Early studies have been reported that there is little evidence of imprinted genes in birds. If imprinted genes are intimately connected with acquiring resources from parents, we would not expect any in the chicken that leave their offspring to their own devices after conception. We identified the imprinted genes of human and mouse as well as orthologues in human, mouse and chicken to show how considerable conservation can be existed in distant species, chicken. This result was significant using statistical test, providing an evidence that the oviparous genome is predicted to have imprinted genes.

        NMR and Fluorescence Studies of DNA Binding Domain of INI1/hSNF5

        INtegrase Interactor 1 protein (INI1/hSNF5) or BRG1-associated factor 47 (BAF47) is a SWI/SNF-related matrix associated actin dependent regulator of chromatin subfamily B member. DNA binding domain of INI1/ hSNF5 is cloned into E.coli expression vectors, pET32a and purified as a monomer using size exclusion chromatography. NMR data show that INI1DBD has folded state with high population of α-helices. By fluorescence-quenching experiments, binding affinities between INI1DBD and two double stranded DNA fragments were determined as 29.9 ± 2.6 μM (GAL4_1) and 258.7 ± 5.8 (GAL4_2) μM, respectively. Our data revealed that DNA binding domain of INI1/hSNF5 binds to transcriptional DNA sequences, and it could play an important role as a transcriptional regulator.

        Functional Prediction of Imprinted Genes in Chicken Based on a mammalian comparative Expression Network

        Little evidence supports the existence of imprinted genes in chicken. Imprinted genes are thought to be intimately connected with the acquisition of parental resources in mammals; thus, the predicted lack of this type of gene in chicken is not surprising, given that they leave their offspring to their own heritance after conception. In this study, we identified several imprinted genes and their orthologs in human, mouse, and zebrafish, including 30 previously identified human and mouse imprinted genes. Next, using the HomoloGene database, we identified six orthologous genes in human, mouse, and chicken; however, no orthologs were identified for SLC22A18, and mouse Ppp1r9a was not included in the HomoloGene database. Thus, from our analysis, four candidate chicken imprinted genes (IGF2, UBE3A, PHLDA2, and GRB10) were identified. To expand our analysis, zebrafish was included, but no probe ID for UBE3A exists in this species. Thus, ultimately, three candidate imprinted genes (IGF2, PHLDA2, and GRB10) in chicken were identified. GRB10 was not significant in chicken and zebrafish based on the Wilcoxon-Mann- Whitney test, whereas a weak correlation between PHLDA2 in chicken and human was identified from the Spearman's rank correlation coefficient. Significant associations between human, mouse, chicken, and zebrafish were found for IGF2 and GRB10 using the Friedman's test. Based on our results, IGF2, PHLDA2, and GRB10 are candidate imprinted genes in chicken. Importantly, the strongest candidate was PHLDA2.

        NMR characterization of SRG3 SWIRM Domain Mutant Proteins.

        SWIRM domain, a core domain of SRG3 is well conserved in SWI3, RSC8, and MOIRA family proteins. To understand structural basis for cellular functions of the SWIRM domain, we have initiated biochemical and structural studies on SWIRM domain and mutants using gelfiltration chromatography, circular dichroism and NMR spectroscopy. The structural properties of the mutant SWIRM domains (K34A and M75A) have been characterized, showing that the structures of both wild-type and mutant proteins are α-helical conformation. The data conclude that mutations at interaction sites of its binding partner protein do not affect its secondary and tertiary structure.
