        Recent trends in biosensors

        Rapid advances in biosensors have recently been reported. This has been possible due to rapid growth in the development of newbiomaterials such as conducting polymers, copolymers and sol gels etc and the reported improvements in sensing techniques. Bio-sensors are miniaturized devices employing biochemical molecular recognition as the basis for a selective analysis. The response gen-erated as a result of biochemical reaction is detected by a transducer to give a signal (optical/electrical/thermal) that can be used withor without amplication for the estimation of the concentration of an analyte in a given test sample. Among the various biosensors,electrochemical sensors, especially amperometric biosensors presently hold a leading position.Due to specicity, portability, simplicity, high sensitivity, potential ability for real-time and on-site analysis coupled with thespeed and low cost, biosensors have been projected to have applications in food analysis, environment control, clinical detection,drug and agriculture industries etc. Besides this, biosensors oer exciting opportunities for numerous decentralized clinical applica-tions, ranging from emergency room screening, home self testing and alternative site testing, continuous and real-time in vivo mon-itoring. New generation of biosensors combining new bioreceptors with the ever-growing number of transducers is emerging.         Application of polyaniline as enzyme based biosensor

        The PANI lms have been synthesized electrochemically and are used as matrix for immobilization of glucose oxidase (GOD)and lactate dehydrogenase (LDH) enzymes. The temporal aspects of anion self-exchange in PANI lms have been investigated. Theexchange of bulkier tosylateferricyanide ion with Cl. ion has been monitored by photometry and electrochemical techniques. Therelative changes in porosity brought about by self-exchange have been experimentally determined to be 323 and 2125/k in tosylate-exchanged and ferricyanide-exchanged polyaniline lms, respectively. It is seen that the polyaniline lms exhibit enhanced loading ofglucose oxidase after a self-ion exchange, and, hence they can be used for the fabrication of a third generation glucose biosensor.Lactate is determined by the photometric detection of NADH formed in the reaction catalysed by LDH. Studies have been car-ried out with PANI as a matrix for the immobilization of LDH and its feasibility as a biosensor. The results of the photometric andamperometric measurements conducted on such LDH/PANI electrodes show a response to pyruvate concentration upto 0.45 mM, aresponse time of 90 s and a shelf life of about two weeks..

        Pectobacterium carotovorum subsp. carotovorum을 침해하는 박테리오파지의 분리

        Bacteriophages of Pectobacterium carotovorum subsp. carotovorum which causes soft rot on diverse vegetables had been isolated from 6 major Chinese cabbage cultivation areas in Korea. In order to isolate bacteriophages, total 15 different strains of P. carotovorum subsp. carotovorum isolated from nation-wide of Korea had been used as a host. When we tested 30 different soil samples individually from Pyeongchang and Taebaek with 15 different strains as a host, Taebek soil samples showed bacteriophage plaques with almost all different indicator strains but Pyeongchang soil samples showed plaques only with P. carotovorum subsp. carotovorum Pcc2 and Pcc3 strains. Especially, P. carotovorum subsp. carotovorum Pcc3 strain was able to produce plaques with almost all soil samples. Thus, this strain can be used as an indicator strain for P. carotovorum subsp. carotovorum bacteriophage screening. Electron microscope observation revealed P. carotovorum subsp. carotovorum bacteriophages isolated in Korea were belonged to three different families,Myoviridae, Siphoviridae and Podoviridae in order Caudovirales. 0

        Characterization of electrochemically synthesized poly(2-fluoroaniline) film and its application to glucose biosensor

        Poly(2-uoroaniline) has been electrochemically deposited on ITO coated glass plates in the form of thin lms using 4 M per-chloric acid as electrolyte. These polymeric lms have been spectroscopically characterized by FT-IR and UV-visible spectropho-tometric methods. Room temperature electrical conductivity of the polymeric lms was measured and found to be lower than that ofpolyaniline lms. The SEM studies show the blunt needle like surface morphology of the polymeric lm.physical adsorption method. The redox characterization of poly(2-uoroaniline) and poly(2-uoroaniline)/GOX lms has beencarried out by cyclic voltammetry technique. Amperometric detection ofb-glucose was carried out by using the above enzymeimmobilized polymeric lm as a working electrode and Ag/AgCl as a reference electrode at a bias voltage of 0.2 V over a physi-ological range.         An experimental set-up for the study of electromechanical properties of conducting polymer films

        An experimental set-up have been designed and fabricated to study the electromechanical properties of a thick film of conducting polymer under load. Extension of the films versus voltage has been measured in terms of change in capacitance of parallel plate capacitor constituted by metal pan and a fixed metal plate. HP 4284A impedance analyzer measures absolute value of the capacitance. Change in capacitance is related to change in distance by pre-calibrating the assembly using traveling microscope. A computer programme is developed to convert the capacitance value with the corresponding distance and simultaneously plotting the graph between changes in length of polymer film versus applied voltage. The assembly has been used to study the electrochemomechanical behavior of solution cast polyaniline films (50 l thick). During first cycle the length is enhanced by about 6% of original value, while repetitive value of extension is ffi2.8%, in subsequent cycles. This value closely matches to that reported in literatures. 2002 Elsevier Science B.V. All rights reserved.
