      • 환경조건에 따른 가시광선 및 적외선 영역에서 청개구리의 변색 연구

        본 연구에서는 온도와 배경색의 변화에 따른 청개구리(Hyla japonica)의 색 변화의 양상을 가시광선 과 적외선 영역에서 연구하였다. 청개구리에게 같은 환경을 만들어 주기 위해 항온항습기를 이용하였 고 환경조건으로는 섭씨 15도와 25도의 온도조건, 그리고 흰색과 검은색의 배경색을 이용하였다. 국 내에서 채집한 청개구리 30개체를 주어진 온도조건과 배경조건에 노출시킨 뒤 각 환경조건에서 청개 구리의 등 색채를 full-spectrum 카메라를 활용, 가시광선과 적외선 영역에서 촬영하여 분석하였다. 청개구리는 온도조건과 배경색 조건 모두에 반응하여 색채를 변화하였으며, 가시광선 영역 뿐 아니라 적외선 영역의 색채 역시 환경조건에 따라 변화한다는 사실을 발견하였다. 또한, 온도 vs. 배경색에 서의 기능적인 색채가 충돌할 때에는 배경색이 청개구리의 색채를 결정하는 데 더 큰 영향을 준다는 사실을 확인하였다.

        Immune-enhancing effects of a traditional herbal prescription, Kyung-Ok-Ko

        Objectives : A traditional herbal prescription, Kyung-Ok-Ko (KOK), has long been used in oriental medicine as an invigorant for age-related diseases, such as amnesia and stroke. However, the beneficial value of KOK for immune responses is largely unknown. Based on the above mentioned effects of KOK, other previous reports, and its use in traditional medicine, we hypothesized that KOK displays beneficial effects against methotrexate (MTX)-induced immune suppression. Methods : We investigated the effects of KOK (0.6 g/kg/day, oral (p.o.)) on deteriorated immunity caused by MTX (2 mg/kg/day, p.o.) in an immune suppression mouse model. MTX was fed to mice once a day for 7 days. After the immune responses of the mice deteriorated by MTX treatment, KOK in water was fed to the mice once a day for 14 days. We then measured the expression levels of various cytokines, such as T helper cell (Th1, Th2) cytokines, and the number of immune cells, such as spleen T cells, B cells, and macrophages. Results : The data showed that MTX decreased Th1 profiles (interferon (IFN)-, interleukin (IL)-2, IL-12) and the number of immune cells, and increased Th2 profiles (IL-4, IL-5, IL-13), which were normalized significantly by post-administration of KOK. However, there was no significant difference in body-weight gain between MTX- and KOK-treated mice. Conclusion : These results indicate that KOK has immune-enhancing functions and reduces immunotoxicity of MTX, suggesting that supplementation with KOK will improve immune responses clinically and be useful for the prevention of immune-related diseases.

        Toosendan Fructus ameliorates the pancreatic damage through the anti-inflammatory activity in non-obese diabetic mice : Erratum

        Objectives : The present study was conducted to examine whether Toosendan Fructus has an ameliorative effect on diabetes-induced alterations such as oxidative stress and inflammation in the pancreas of non-obese diabetic (NOD) mice, a model of human type I diabetes. Methods : Extracts of Toosendan Fructus (ETF) were administered to NOD mice at three doses (50 mg/kg, 100 mg/kg and 200 mg/kg). Mice at 18 weeks of age were measured glucose tolerance using intraperitoneal glucose tolerance test. After 28 weeks of ETF treatment, glucose, total cholesterol (TC), triglyceride (TG), and proinflammatory cytokines in serum, western blot analyses and a histopathological examination in pancreas tissue, and on the onset of diabetes were investigated. Results : The results showed that levels of glucose, glucose tolerance, TC, TG , interferon-γ, interleukin (IL)-1β, IL-6, and IL-12 in serum were down-regulated, while IL-4, IL-10, SOD, and catalase significantly increased. In addition, ETF improved protein expression of proinflammatory mediaters (such as cyclooxygenase-2, and inducible nitric oxide synthase) and a proapoptotic protein (caspase-3) in the pancreatic tissue. Also, in the groups treated with ETF (100 mg/kg or 200 mg/kg), insulitis and infiltration of granulocytes were alleviated. Conclusions : Based on these results, the anti-diabetic effect of ETF may be due to its anti-inflammatory and antioxidant effect. Our findings support the therapeutic evidence for Toosendan Fructus ameliorating the development of diabetic pancreatic damage via regulating inflammation and apoptosis. Our future studies will be focused on the search for active compounds in these extracts.

        비파엽약침이 고지방식이 유발 비만 생쥐의 국소부위지방에 미치는 영향

        Objectives : This study was designed to investigate the anti-obesity effects in local area adipose tissue using by pharmacopuncture of Eriobotryae Folium on mice fed high fat diet. Methods : Male ICR mouse were divided into three groups. Normal diet group (N), High-fat diet(HFD) group, HFD+E(pharmacopuncture of the Eriobotryae Folium) group for 8 weeks. HFD+E group was injected in the concentration of 50 mg/kg once a day for 4 weeks and every other day during the next 4 weeks. We measured body weight, food intake, absolute weight and relative weight in the liver and adipose tissue. Also, we analyzed hematological levels and histopathological changes on adipose tissue. Results : 1. We observed no difference in food intake among three groups.2. HFD+E group significantly decreased body weight gain and absolute weight and relative weight in the liver and adipose tissue compared with HFD group.3. Blood AST and ALT were no differences among the experiment groups. However, HFD group was significantly increased higher TG, TC, HDL and LDL than in N group and HDL/LDL ratio was remarkably decreased. On the other hand, HFD+E group was significantly decreased in obesity markers and HDL/LDL ratio was remarkably increased.4. Levels of adiponectin in serum of HFD+E group was remarkably increased compared with those of HFD group. 5. HFD group was significantly decreased compared with N group in adipocyte number. Compared with the HFD group, HFD+E group was displayed a significant increase by adipocyte number. Conclusions : We suggest that pharmacopuncture of the Leaves of Eriobotrya japonica can play the anti-obesity effects in local area adipose tissue.

        Low-Molecular-Weight Oligonol, a Polyphenol Derived from Lychee Fruit, Attenuates Experimental Reflux Esophagitis and HCl/Ethanol-Induced Gastric Ulcer

        Oligonol, a polyphenol derived from lychee fruit, is produced by an oligomerization process that converts high-molecular-weight polyphenol polymers into low-molecular-weight oligomers. Evidence suggests that oligonol exerts its beneficial effects based on antioxidant and anti-inflammatory properties. This study was the first to investigate the antioxidative and anti-inflammatory effects of oligonol on gastroesophageal inflammatory models: surgically induced acute reflux esophagitis (RE) and gastric ulcer (GU) induced by HCl/ethanol. In the in vitro study, 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2′-azino-bis(3-ethylbenzothiazolin-6-sulfonic acid) (ABTS) radical scavenging assays were performed to determine the antioxidant activity of oligonol. The experimental groups were each composed of normal, vehicle, and oligonol groups. RE rats and GU mice were treated orally with oligonol (100 mg/kg bw) or distilled water as a vehicle (n = 8 for each group). Oligonol exhibited potent free radical-scavenging capacities for DPPH and ABTS radicals, activities that were similar to those of ascorbic acid. The in vivo study revealed that oligonol consumption significantly prevented RE and GU formation and decreased the gross mucosal injury from oxidative stress. Oligonol decreased the reactive oxygen species levels and elevated levels of both inflammatory mediators and cytokines (p-IκB, NF-κBp65, COX-2, iNOS, TNF-α, and IL-1β) in the RE and GU models. Oligonol had a protective effect against oxidative stress by regulating antioxidant enzyme (superoxide dismutase, catalase, and GPx-1/2) activities in GU mice. Oligonol has potential as a preventive and therapeutic agent for gastroesophageal inflammatory diseases, including RE and GU.

        Micronucleus Test of DHU001, a Polyherbal Formula, in Bone Marrow Cells of Male ICR Mice

        The genotoxic effects of DHU001, a polyherbal formula were evaluated using the mouse micronucleustest. DHU001 was administered once a day for 2 continuous days by oral gavage to maleICR mice at doses of 2000, 1000 and 500 mg/kg. Cyclophosphamide was used as a known genotoxicagent in a positive control. The appearance of a micronucleus is used as an index for genotoxicpotential. In addition, the changes on the total white blood cells and differential counts on thelymphocytes, neutrophils, eosinophils, basophils and monocytes in the prepared blood smears werealso conducted to observe the possible immunosuppression. The results indicats that DHU001showed no genotoxicity effects up to 2000 mg/kg dosing levels and did not influenced on the totalwhite blood cells and differential counts. In addition, it is also considered that there were no problemsfrom cytotoxicity of DHU001 tested in this study because the polychromatic erythrocyte ratiowas detected as > 0.41 in all tested groups.

        Mouse Single Oral Dose Toxicity Study of DHU001, a Polyherbal Formula

        This study was conducted to obtain acute information of the oral dose toxicity of DHU001, a polyherbal formula in male and female mice. In order to calculated 50% lethal dose (LD50) and approximate lethal dose (LD), test material was once orally administered to male and female ICR mice at dose levels of 2000,1000, 500, 250 and 0 (vehicle control) ml/kg (body weight). The mortality and changes on body weight,clinical signs, gross observation, organ weight and histopathology of principle organs were monitored 14days after treatment with DHU001. We could not find any mortalities, DHU001 treatment-related clinical signs, changes on the body and organ weights, gross and histopathological findings. The results obtained in this study suggest that LD50 and approximate LD in mice after single oral dose of DHU001 were considered over 2000 mg/kg in both female and male mice.

        Lithospermi Radix is in use in oriental medicine. Oriental medical doctors use Lithospermi Radix for clearing away heat and eliminating toxin(淸熱解毒), etc. We investigated toxicity, poisoning symptoms, poisoning treatment and prevention against poisoning of Lithospermi Radix. The results were summarized as follows: 1. Taking Lithospermi Radix excessively cause diarrhea, etc. 2. In order to prevent against side effects of Lithospermi Radix, the patients should keep usage, dosage and notes.

        Croci Stigma is in use in the Korean medicine. The Korean medical doctors use Croci Stigma for activating blood flow and removing blood stasis(活血祛瘀), removing heat from the blood(凉血), dispersing accumulations of pathogen(散結), etc. We investigated toxicity, poisoning symptoms, poisoning treatment and prevention against poisoning of Croci Stigma. The results were summarized as follows: 1. The ingredients of poisoning from Croci Stigma mainly can cause stimulating symptoms on digestive system, urinary system, nervous system, circulatory system, etc and cause hypersensitive reaction. 2. The representative methods of poisoning treatment are promotion of vomiting, washing out the stomach, supplies of grape sugar and symptomatic treatment, etc. 3. In order to prevent against poisoning of Croci Stigma, the patients should keep usage, dosage and notes.
