The vasorelaxant effect of an extract of Lophatherum gracile Brongn (ELB) and its possible action mechanism were ascertained in aortic tissues isolated from rats. ELB relaxed endothelium-intact thoracic aorta in a dose-dependent manner. However, the induced vascular relaxation was abolished by removal in endothelium of the thoracic aorta. Pretreatment of endothelium-intact vascular tissues with $N^G$-nitro-L-arginine methyl ester (L-NAME) or 1H-[1,2,4]-oxadiazole-[4,3-$\alpha$]-quinoxalin-1-one (ODQ) significantly inhibited vascular relaxation induced by ELB. Moreover, ELB significantly increased cGMP production in aortic tissues, which was blocked by pretreatment with L-NAME or ODQ. The vasorelaxant effect of ELB was attenuated by tetraethylammonium (TEA), and glibenclamide. ELB-induced vasorelaxation was not blocked by atropine, propranolol, indomethacin, verapamil, and diltiazem. Taken together, the present study demonstrates that ELB dilates vascular smooth muscle via an endothelium-dependent NO-cGMP signaling pathway, which may be at least in part related with the function of $K^+$ channels.

Objectives: This study was designed to investigate the basis for developing the herbal decoction that may help promote healthy blood vessels in accordance with the basic principles of Korean medicinal materials. Methods: In this study, we investigated the vascular relaxation effects of fifty herbal decoctions on vascular response in isolated thoracic aorta of phenylephrine-preconracted rats. Results: The results of identify the vascular relaxation effect of fifty herbal decoctions provided by Korea Institute of Korean Medicine, twenty-one herbal decoctions showed vascular relaxation effects. Among them, four herbal decoctions, Mahwang-tang (麻黃湯), Samchulgeonbi-tang (參朮健脾湯), Cheonwangbosim-dan (天王補心丹), and Socheonglyong-tang (小靑龍湯), with showed significantly concentration- dependent vasodilation was examined. Vascular relaxation level are 84.02±4.70, 79.39±13.99, 51.26±12.56, and 54.73±15.88%, respectively (P<0.05, 0.01, and 0.001). Conclusions: Thus these results provide a basic data for the treatment of the impairment of vascular function and blood pressure in traditional Oriental medicine.
당뇨병성 신병증 모델에서 블랙커런트의 사구체 섬유증 및 신장 기능장애 개선 효과

본 연구를 통하여 블랙커런트 추출물의 섭취가 제 2형 당뇨동물 모델인 db/db 마우스에서 혈당 개선 및 사구체 확장, 신장섬유증, 염증을 억제하여 당뇨병성 신병증을 개선했다는 것을 입증했다. 제 2형 당뇨병 db/db 마우스에서 블랙커런트 추출물의 섭취가 인슐린 저항성 및 신장 기능 장애와 같은 당뇨병 관련 대사 장애를 개선하고, 당뇨병성 신병증에 있어 신장 염증 및 신장섬유증을 현저하게 약화시켰다는 것을 보여주었다. 결론적으로, 이러한 연구 결과는 블랙커런트 추출물이 신장 섬유증, 염증 및 이에 따른 당뇨병성 신병증의 예방에 중요한 역할을 할 수 있다는 증거를 제공한다. 따라서 당뇨병 관련 신기능 장애에 대한 블랙커런트 추출물의 보호 역할은 당뇨병성 신병증 개선 건강성 식품 개발에 대한 새로운 통찰력을 제공 할 수 있다. Diabetic nephropathy is a major and representative complication of type 2 diabetes. Hyperglycemia increases the incidence of diabetic nephropathy, and induces kidney inflammation, thereby causing renal fibrosis, which is an important factor in the pathogenesis of diabetic nephropathy. This study investigated the effects of blackcurrant extract (BLC), which has been implicated in diabetic nephropathy in db/db mice, on glomerular fibrosis and renal dysfunction. The results showed that BLC consumption in type 2 diabetic db/db mice ameliorated diabetes-related metabolic disorders, such as insulin resistance and renal dysfunction, and significantly attenuated renal inflammation and renal fibrosis in diabetic nephropathy. In conclusion, these findings suggest that BLC consumption may help prevent renal fibrosis, inflammation, and consequent diabetic nephropathy.
고지혈증 동물모델에서 홍삼과 천마 혼합투여에 의한 혈관 염증 개선연구

Objectives : This experimental study was designed to investigate the inhibitory effects of combination with Korean red ginseng and Gastrodia rhizoma on vascular dysfunction in high-fat/cholesterol diet-induced hyperlipidemia. Methods : Sprague-Dawley rats were fed with 7.5% cocoa butter and 1.25% cholesterol for 10 weeks, with Panax ginseng (PG), and mixtures of Panax ginseng and Gastrodia rhizoma (PGM), respectively. Results : Chronic treatment with PG and PGM significantly decreased body weight. The aortic expression of cell adhesion molecules such as intracellular adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1 (VCAM-1), and E-selectin were markedly increased in hyperlipidemia rats. Interestingly, PGM significantly decreased cell adhesion molecules expression. However, there was no significant decrease in PG group. In addition, PG and PGM group inhibited high-fat/cholesterol diet-induced cytokine such as monocyte chemoattractant protein (MCP-1) mRNA expression. Furthermore, PG and PGM group significantly decreased c-reactive protein protein (CRP) level. Especially, PGM significantly accentuated the decrease of MCP-1 mRNA expression and CRP level. Conclusions : the present study provides an evidence that combination with Panax ginseng and Gastrodia rhizoma enhances anti-vascular protective effects through suppression of vascular inflammation in hyperlipidemic rats.
심근 경색 유발 심부전 모델에서 강리 추출물의 심장 보호 가능성

Gracilaria Verrucosa (GV), a seaweed used in traditional Korean medicine, was studied for its effects on MI-induced heart failure in rats. MI is caused by a blocked coronary artery, leading to severe cardiac dysfunction. The study used a rat model to assess cardiac changes over time and evaluate the impact of GV on heart failure. Ischemia was induced through LAD ligation surgery, and the extent of ischemic area was measured as a prognostic factor. GV extract administration significantly improved cardiac morphology and reduced cardiac weight compared to the MI group. GV treatment also improved cardiac function, as evidenced by positive effects on chamber dilation during MI-induced heart failure. Parameters such as ejection fraction (EF) and fractional shortening (FS) were measured. The MI group showed decreased EF and FS compared to the sham group, while these parameters improved in the GV group. GV treatment also reduced levels of LDH, CPK, and CK-MB in the serum, indicating reduced myocardial damage. Histological analysis revealed that GV treatment attenuated cardiac hypertrophy and fibrosis, with reduced collagen deposition in the myocardium. Immunohistochemistry analysis showed suppressed expression of TGF-β1 and collagen 1, involved in fibrosis. In conclusion, GV showed potential in improving cardiac function in a rat model of MI-induced heart failure. It alleviated myocardial damage, attenuated cardiac hypertrophy and fibrosis, and suppressed fibrotic markers. Further studies are needed to explore its clinical efficacy and underlying mechanisms in cardiac diseases beyond animal models.
천연물 복합 추출물 (HAE-06)의 기관지 확장에 미치는 영향

HAE-06 extract is a mixture of four medicinal plants, namely Lonicerae Folium et Caulis (Lonicera japonica), Scutellariae Radix (Scutellaria baicalensis), Adenophorae Radix (Adenophora triphylla var. japonica), and Polygonati Oddorati Rhizoma (Polygonatum odoratum var. pluriflorum). The HAE-06 extract demonstrated a concentration-dependent relaxing effect and enhanced cAMP production in bronchial smooth muscle that had been stimulated to contract with acetylcholine. Using a blocker, it was confirmed that the effect was through the β2-adrenergic receptor/cAMP/PKA pathway. In addition, it is thought that the HAE-06 extract has a bronchial smooth muscle relaxation effect by reducing the inflow of Ca<sup>2+</sup> through the K<sup>+</sup> and Ca<sup>2+</sup> channels present in the sarcoplasmic membrane. If research continues in the future, it is believed that it will be possible to use it as a material for pharmaceuticals and functional foods.
