        송암스님 화청의 음악구조 형태

        The purpose of this study is to investigate musical structure form of Hwachung by Songam Buddhist monk. Songam is considered to hand down Hwachung well through activity of passing intangible cultural asset as a holder of Yeongsanjae. Investigating musical structure form of Hwachung by Songam is needed so that traditional musical gene can communicate with new age and can be recreated as creative music. For this end, Songam’s sound sources were collected and then sound sources with completeness in terms of music were gathered. They were divided into melody structure and rhythm structure and classified into paragraphs and parts. Hwachung by Songam is Gyeongtori considering the mode of B♭bonchung Sol (F), La (G), Do (B♭), Re (C), Mi (D). Rhythm structure is composed of mixed beats of 3 beats+2 beats+3 beats. Rhythm structure is 8/8 beat. This seems to be closely related to regions where Hwachung by Songam has been handed down. Hwachung being handed down nowadays is dominated by Sol mode Gyeongtori. This is because Hwachung has been handed down centering around designation of national intangible cultural asset. It is desirable for us to inherit and develop traditional music escaping from prejudice of evaluation of traditional music and hand down musical gene to our descendants wholly. 본 연구의 목적은 송암스님 화청의 음악구조 형태를 규명하는데 있다. 송암스님은 영산재의 보유자로 무형문화재 전승활동을 통해 표본적인 화청을 잘 전승한 것으로 여겨진다. 전통 음악적 유전인자를 새로운 시대와 소통하고 창의적인 음악으로 재창조되기 위하여 송암스님 화청의 음악구조 형태를 규명할 필요가 있다. 이를 위해 송암스님 음원을 수집한 후 음악적으로 비교적 완성도가 높은 것으로 보이는 음원을 채보하였다. 그리고 선율구조와 리듬구조 형태로 나누고 단락과 마루로 구분하였다. 연구결과는 다음과 같다. 송암스님의 화청은 B♭본청 Sol(F), La(G), Do(B♭), Re(C), Mi(D) 선법으로 보아 경토리이다. 리듬구조는 3분박+2분박+3분박의 혼합박 형태로 8/8박자이다. 이는 송암스님의 전승지역과 밀접한 연관이 있다고 볼 수 있다. 오늘날 전승되고 있는 화청은 보편적으로 Sol선법 경토리가 중심을 이루고 있다. 그 까닭은 국가무형문화재 지정 중심으로 전승이 이뤄지기 때문일 것이다. 따라서 전통음악의 가치평가에 대한 편견에서 벗어나 이를 올곧게 계승하고 발전해 나아감으로써 음악적 유전인자를 온전히 물려줘야할 책임이 있다.

        활액막 추벽 증후군과 정상 추벽에서의 활액막 추벽의 자기공명영상 소견

        Purpose : To compare MRI appearance between plicae syndrome and normal plicae. Materials & Methods : MR imagesof 60 cases of arthroscopically-confirmed plicae syndrome and 18 of arthroscopically-proven normal plicae wereretrospectively analyzed. Sagittal T2-weighted MR images in all cases and MPGR(200) in 37 cases of plicae syndromewere obtained. Statistical analysis was performed using the chi-square test. Results : On the basis of operatingresults, we observed 55 medial plicae, eight combined medial and suprapatellar plicae, four suprapatellar plicae,and one lateral plica. T2-weighted sagittal MR scans of the 60 cases demonstrated 37 medial plicae, 8suprapatellar and one lateral plica. Joint effusion was found in 26 cases of 55 medial plicae. In T2-weightedsagittal MR scans, the identification of medial plicae was superior in the presence of joint effusion than itsabsence(plicae syndrome, p < 0.001 ; normal plicae group, p < 0.05). Medial plicae were well demonstrated onMPGR(200) axial images; on T2-weighted sagittal MR scans, they could be more frequently identified in the plicaesyndrome group than in the normal control group(p < 0.001). Plicae syndrome-associated pathology includeddegenerative change of the articular cartilage of the medial femoral condyle in eight cases(14.5%), discoidmeniscus in nine(16.4%), lateral meniscus tear in 12(21.8%), medial meniscus tear in 21(38.1%), anterior cruciateligament tear in three(5.5%), medial collateral ligament tear and osteochondritis dissecans in one case.Conclusion : The present study revealed that synovial plicae were well demonstrated in T2-weighted sagittalimages, particularly on the presence of joint effusion. Medial plicae could be more frequently identified in theplicae syndrome group than in the normal control group, especially on T2-weighted sagittal MR scans.

      • PCR 방법에 의한 결핵균의 신속한 동정

        Tuberculosis is caused by Mycobacterium tuberculosis. Long culture time of the strain limited early diagnosis of the disease. Polymerase chain reaction (PCR) has become one of the most important techniques in molecular biology, and will be more promising in the future. DNA was extracted from patients’ sputa, and the 123 bp DNA fragment on the chromosomal DNA of M.tuberculosis was amplified specifically just within 30 min by PCR. Optimal amplification conditions for rapid detection of the strain from patients’ sputa were established. PCR sensitivity was tested by amplification of the defined genomic DNA in specimens from patient s pleural fluid and lung tissue sections. However, the fragment was not amplified, suggesting that there was not the strain in the specimens.

        Bacillus anthracis와 그 유연종의 rpoB 유전자 컴퓨터 분석을 통한 동정

        Bacillus anthracis, B. cereus, B. thuringiensis 를 분류하기 위해 rpoB 유전자 배열을 이용한 컴퓨터 분석 작업을 수행하였다. 17개의 B. anthracis, 9개의 B. cereus, 7개의 B. thuringiensis 를 database에서 구하였다. B. anthracis 는 rpoB 유전자의 in silico 제한효소 절단에 의해, B. cereus, B. thuringiensis 2 group과 구별되었다. 그러나 B. cereus와 B. thuringiensis 는 제한효소 절단에 의해 구분되지는 않고, 염기배열과 Blast 탐색의 도움으로 구분이 가능하였다. 본 연구를 통해 3 종류의 Bacillus 종을 동정할 수 있는 알고리즘이 개발되었다. Computational analysis of partial rpoB gene sequence (777 bp) was done in this study to identify B. anthracis and its closely related species B. cereus and B. thuringiensis. Sequence data including 17 B. anthracis strains, 9 B. cereus strains, and 7 B. thuringiensis strains were obtained by searching databases. Those sequences were aligned and used for other computational analysis. B. anthracis strains were identificated by in silico restriction enzyme digestion. B. cereus and B. thuringiensis were not segregated by this method. Those sequencing and BLAST search were required to distinguish the two. In actual identification tests, B. anthracis strains could be identified by PCR-RFLP, and B. cereus and B. thuringiensis strains were distinguished by BLAST search with reliable e-value. In this study fast and accurate method for identifying three Bacillus species, and flow chart of identification were developed.

        Suppression of metastasis-related ERBB2 and PLAU expressions in human breast cancer MCF 7 cells by fermented soybean extract

        발효대두 청국장에는 대두단백질이 발효에 의해 분해 형성된 다양한 펩타이드류가 들어 있다. 청국장 추출물이 처리된 유방암세포의 microarray data와 잘 알려진 유방암 전이 마커를 합쳐서 새로운 연결망이 제조되었으며 이를 이용해 전이 마커와 발현 차이가 있는 단백질 사이의 상호작용을 체크하였다. 연결망 분석을 통해 PLAU (plasminogen activator, urokinase, uPA)와 ERBB2 (epidermal growth factor receptor 2)를 실제 전이 가능성을 보여주는 유전자로 선택하였다. MCF7 암세포를 청국장추출물로 처리하고 PLAU와 ERBB2 발현정도를 측정하였다. 청국장 추출물은 PLAU와 ERBB2 발현을 상당히 억제하였다. 청국장 추출물을 처리한 암세포에서 염증 마커인 NO의 생산이 감소하였다. 인간 유방암세포에서 PLAU와 ERBB2 발현을 특이적으로 감소시키는 펩타이드를 찾아내는 것은 흥미로운 일일 것이다. Chunkookjang, fermented soybean is rich in diverse oligopeptides which derived from cleavage of soybean proteins during fermentation. Microarray data containing differently expressed genes in breast cancer cells treated with fermented soybean extract and well known breast cancer metastasis markers were combined, and a new network was constructed. It is used to check interactions between the marker proteins and the differently expressed genes. Based on the network analysis, PLAU (plasminogen activator, urokinase, uPA) and ERBB2 (epidermal growth factor receptor 2) are chosen as possible metastasis genes. We treated breast cancer MCF7 cells with fermented soybean extract and measured expression levels of PLAU and ERBB2. Fermented soybean extract suppressed PLAU and ERBB2 expressions conspicuously. In the cancer cells treated with fermented soybean extracts, an inflammation marker, NO production was also reduced. It will be interesting to find specific peptides to suppress PLAU and ERBB2 expressions in human breast cancer cells.

        Genistein, daidzein 은 ERα 보다는 ERβ를 30배 정도 선택적으로 자극한다 . 이들 isoflavone 의 ERβ 자극효과는 유방,전립선 암의 억제에 기여한다 . Genistein 이나 daidzein은 ERα,ERβ 를 매개로 하여 histone acetyltransferase 활성에 영향을 미친 다 . 어미 쥐가 섭취한 genistein은 histone을 acetylation 시켜 태아쥐의 genome에 영 향을 미쳐 영구적으로 새끼쥐의 비만을 막고 털색깔을 바꿀 수 있다. 암세포에서는 특정 microRNA 가 증감되는 양상을 보인다. Isoflavone 에 의한 특정 microRNA 발 현의 조절은 암치료에 적용될 수 있다 . Genistein은 estrogen 에 반응을 보이는 유전 자에 영향을 미쳐 자궁 biology 나 초기 임신에 도움이 될 수 있다 . Genistein 과 daidzein 은 COX-2 의 활성을 억제하여 항염증효과를 발휘한다. Daidzein 이 풍부한 청국장추출물을 유방암 세포인 MCF7 세포에 처리하면 염증억제에 관련된 TGFβI와 Smad3 유전자의 발현은 증가하고,염증 촉진 관련 CSF3 , CCL3 유전자 발현은 감소하면서 MCF7 성장을 억제할 수 있다 .

      • Bacillus sp. SH-5와 그것의 알칼리성 단백질 분해효소의 특성에 관한 연구

        For selection of bacteria that produce protease, well growing bacteria were isolated from Korean Soybean paste in the media containing 1% skim milk. Out of about 100 colonies which showed protease activity with a clear zone on the media containing 1% skim milk, a colony producing the largest amount of protease was selected. The isolated and selected strain SH-5 was an aerobic, spore-forming, Gram positive, motile, rod-shaped bacterium. Considering its characteristics, the bacterium belongs to genus Bacillus. The optimum pH for growth and alkaline protease production of the bacterium was 7.0, and it grew at range of pH 6.1~9.4. The optimum temperature for its growth and the alkaline protease production was 37°C The enzyme from this strain was most active at pH 9 in casein containing buffer and stable at a range pH 6.0~12.0 for 30 min. The optimum temperature for the enzyme action was 60°C in the presence of calcium ion.

      • 된장에서 단백질 분해효소를 분비하는 Bacillus cereus SH-7의 특성에 관한 연구

        To know whether or not there are bacteria producing excretory protease in Korean traditional fermented food, soybean paste (Doen-Jang), it was screened by methods of casein and gelatin hydrolysis, and seemingly different groups of bacteria were obtained. One strain was identified to be Bacillus cereus SH-7 through about fifty physiological, biochemical tests, and automatic identification system, VITEK system and MIDI system. This strain was a sporeforming, Gram(+), motile, and rod-shaped bacterium. The strain grew well at pH 7 and 37°C. Its extracellular protease showed optimal activity at pH 8 and 50°C. When 1%(W/V) casein was added to the medium, its activity was increased about 40% at 24 h. Also, hemolysis activity of Bacillus cereus SH-7 was observed.

        Overexpressed Chloramphenicol Acetyltransferases Are Soluble but Mostly Inactive

        We constructed an E. coli strain that produces chloramphenicol acetyltransferase (CAT) in high yield by utilizing the phage T7 RNA polymerase's stringent recognition and active transcription of T7 promoter. The T7 expression system was constructed such that the T7 RNA polymerase gene is under the control of lacUV5 promoter in one plasmid, and that the target gene, a promoterless CAT gene along with E. coli ribosome binding site is under the control of a T7 promoter in the other plasmid. Without induction the T7 expression system is repressed by lacI repressor. The T7 RNA polymerase produced by IPTG induction is capable of specific and efficient transcription of the CAT gene. Upon IPTG induction CAT is produced up to 36% of the total protein content. The overproduced CAT proteins are soluble but mostly inactive, as assayed for specific activity of the protein.
