        Penicilium sp . 를 이용한 토끼풀의 생물학적 방제

        Among 980 fungal strains isolated from the lesions of Trifolium repens L. and various weeds, three Penicillium sp. F40362, F40496, F40497 were selected by the first selection test and a pathogenicity test. The spores of these strains germinated readily 90 to 100% and readily infected the respective plant. The wheat bran-corn starch formulation of F40362 strain showed 100% mycoherbicidal activity against clover plant at 4×10^8 spores/pot. The same formulations of F40496 and F40497 strains showed 100% mycoherbicidal activity against clover plant at 2×10^8∼4×10^8 spores/pot. The same formulations of three strains showed over 30% mycoherbicidal activity against Leguminosae plants. This method of pelletiation was potentially useful for the production of inoculum formulation as mycoherbicides and it was effective enough to treat 2∼2.5 g formulation(4.5×10^7 spores/㎎) to a 350 ㎠ pot. The three strains, F40362, F40496 and F40497 have selective mycoherbicidal activity between Trifolium repens L. and Zoysia japonica and nonselective mycoherbicidal activities against some other crop plants and weeds.

        동물 사료화를 위한 음식물 쓰레기의 미생물 분포 변화

        This study investigated the microbiological changes and distribution of bacteria producing enzyme in order to change food wastes to animal feed during 6 days at room temperature. Food wastes were divided as follows: one is untreated food wastes containing 80% water content and another wastes containing 40% water content adiusted by wheat bran. During the fermentation of food wastes, Lactobacillus sp. and Streptococcus sp. were grown the top position among investigated microorganisms in both of food wastes. Numbers of total microorganisms were much more than untreated wastes during fermentation of flood wastes with 40% water content. But, Streptococcus sp. and Enterobacteriaceae from 4 to 6 days were not detected in treated wastes with 40% water contents. This fact indicated that the adiustment of water content in food waste was effective in fermentation. However, the numbers of microorganisms producing pretense were low in both treated and untreated food wastes. These results suggested that flood wastes should be treated as food wastes with 40% water content to maintain a lot of fermentative microorganisms such as Lactobacillus sp., Streptococcus sp. and yeasts together with pretense to make final animal feed.

        Penicillium sp.를 이용한 토끼풀의 생물학적 방제

        유기합성농약에 의한 환경오염 문제를 해결할 수 있는 미생물제초제를 개발할 목적으로 토끼풀로부터 분리된 Penicillum 곰팡이를 사용하였다. 토끼풀에 대한 제초활성 검정 실험을 통하여 선발된 F40362, F40496, F40497등 3균주의 포자를 여러 가지 생물고분자물질과 담체를 이용하여 제제화한 후 다양한 식물에 대한 제초활성을 조사하였다. 선발된 3 균주의 제제형태에 따른 제초활성은 $2{\times}10^8{\sim}4{\times}10^8$ 포자농도에서 토끼풀에 대하여 100%의 활성을 나타내는 rice bran-corn starch 제제가 가장 우수하였다. 동일한 제제로 잔디와 주요 식량작물, 그리고 밭잡초에 대하여 제초활성을 조사한 결과 잔디종인 Zoysia japonica에서 $1{\times}10^{10}$ 포자농도에서도 영향이 없었으며 작물중에서는 목화에 대하여 비교적 높은 감수성을 나타내었고 콩과 식물에 대해서는 $4.5{\times}10^{7}$ 포자농도에서 30 이상의 병해를 나타내었다. 농도에 따른 밭조건 실험결과에서는 rice bran-corn starch 제제($4.5{\times}10^{7}$ 포자수/g)를 $2{\sim}2.5\;g/350\;cm^2\;pot$ 수준으로 처리하는 것이 적당한 것으로 나타났다. 이상에서 본 제제는 잔디와 토끼풀에 있어서는 높은 선택성을 나타내며 다른 작물과 잡초에 있어서는 비선택적인 제초활성을 나타내었다.         Nine hundred and eighty fungal strains were isolated from lesions of Trifolium repens L. and various weeds. Among them, F40362, F40496. F40497 strains were selected by the screening of herbicidal activity on Trifolium repens L. and Zoysia japonica. Selected three strains showed selective activity between Trifolium repens L. and Zoysia japonica, and were identified as Penicillium sp.

        자연 동굴 토양 방선균의 속 다양성 분포

        국내에 소재 하는 자연 동굴 중 충청북도의 고수굴, 천둥굴, 노동굴, 강원도의 고씨굴, 경상북도의 성류굴, 제주도의 협재굴, 쌍용굴, 만장굴 등 8개 동굴의 내부에 존재하는 토양 방선균 분포를 조사하였다. 그 결과 전체적인 속 수준의 분포로서는 Streptomyces가 52.5%, Micromonospora가 16.3%, Nocardioform 22.8% 정도 분포하고 있었으며 Kineosporia, Actinomadura, Nocardiopsis, Streptosporangium의 순서로 방선균이 0.3~1.4% 정도 분포하고 있었다. 지금까지 보고된 일반적인 방선균 분포와 비교해 볼 때 특히 Streptomyces의 분포 비율이 낮고 상대적으로 Micromonospora, Nocardioform이 많이 분포하고 있었다. 한편 노동굴 지역을 대상으로 동굴 내부와 외부로 나누어 방선균 분포를 비교해 보았다. 그 결과 동굴의 내부에는 Streptomyces가 52.0%, Micromonospora가 16.8%, Nocardia가 32.0% 정도 분포하였고 외부에서는 Streptomyces가 75.8%, Micromonospora가 10.7%, Nocardia가 7.1% 정도 분포하였다. 따라서 동일 지역 내에서도 동굴의 내부와 외부에 따라서 큰 차이를 나타내고 있었다. 그리고 분리된 방선균 중에서 Streptomyces속 균주의 형태적인 특징을 조사한 결과, 기균사의 색에 있어서는 회색 계열 45.7%, 적색 계열 7.8%, 황색 계열 2.3%, 녹색 계열 13.2%, 백색 계열 27.1%, 불량 3.9%의 분포를 나타내었다. 일반적으로 Streptomyces속 방선균에서 잘 분리되는 회색과 적색 계열의 분리 비율은 크게 낮아 졌으나 희소한 녹색과 백색 계열은 분리 비율이 크게 증가하였다. 그리고 성류굴에서는 황색, 녹색, 백색, 계열등 다양한 계열의 Streptomyces속 방선균이 많이 분리되었다. 포자 연쇄의 형태에 있어서는 retinaculiaperti 형태가 35.7%, spirales 형태가 12.4%, rectiflexibiles 형태가 51.9%로 나타났다. Total 276 soil actinomycete strains were isolated from 46 soil samples collected at domestic natural caves; the Kosu, Chundong, and Nodong caves at Chungbook province, the Kossi cave at Kangwon province, the Sungruye cave at Kyungbook province, the Hyupjae, Ssangyoug, and Manjang caves at Cheju province. All of these isolates were identified to the genus level based n morphological and physiological characteristics. As the result, 52.5% of those isolates were Streptomyces, 16.3% were Micromonospora, 22.8% were Nocardioform group, 1.1% were Actinomadura, 0.3% were Nocardiopsis, 0.3% were Streptosporangium, 0.3% were Nocardioides, 1.4% were Kineosporia, 4.7% wee the others. Streptomycete strains were the most abundant, but were relatively less comparing to general distribution pattern. Nocardioform and Micromonospora strains were quite abundant, and other rare actinomycete groups were somewhat abundant comparing to general distribution pattern previously reported. Especially Nocardioform strains were highly abundant at almost of the natural caves.

      • 축사환경개선제의 비육돈 생산 효과에 관한 연구

        Five commerdy avatlable probiotics or yeast culture were compared with the performance of growing-hhgpigs and odorgenerating substances in the feces. Total of 240 pigs were used for group feedig trial for 80 days with 6 treatments(T0 : Control, T1 : DS Cleaner 1 kgl 1 ton feed, T2 Atapon 0.5 kg/l ton feed, T3 : Bio-pro 1 kgl 1 ton feed, T4 : Photoplus 1 kg/ 1 ton feed and T5 CYC 2000, 5 kg/ 1 ton feed). Each treatment has 20 pigs with two replicates. At the end of trial 7 pigs from each treatment were slaughtered for carcase quality evaluation. Fecal nitrogen, ammonia nitrogen, VFA concentration and pH were analysed as a indicator of ordorgenerahg substance. The bactenal counts in the feces and acid resstance and enzyme activity of isolated bacteria were also evaluated. Average body weght at D-70 were T1: 97.46 kg, T3 : 95.18 kg, T2 : 94.44 kg and T4 : 93.49 kg, sgruficantly(p<O.05) higher body weights were obtained in probiotic feeding group than the control. However, there was no significant difference in feed intake and feed converjlon between treatments. Backfat thickness were Sgnficantly(p<O.05) affected by probiotic or yeast culture feeding, lowest backfat was in Tl(15.29 mm) and higher backfat were found in probiotic feedug group than the control. Total bactenal counts in feces were in the range of 10^(7)-10^(8) cfu/g in the first week of tnal but was gradually decreased by 106-107 at the end of experiment. Number of coliform bactenal cell was not in the other group. Total nitrogen, ammonia nitrogen and total VFA umcentration in the feces were decreased in all treatment during probiotic feeding period. Total nitrogen concentration were decreased by 68.28% in T1, 60.19% in T4, 45.63% in the control, 38.99% in T2, 31.83% in T5 and 18.10% in T3 when compared between week 1 and week 10. Fecal ammonia nitrogen concentration also expressed the similar pattern of total nitrogen. Bactenal enzyme activity and the add resistance capability of bacteria which were cultured from commercial probiotics were also evaluated.

        국내 분리 방선균의 항균활성 특성

        제주도지역, 충청남도 서해안지역, 강원도 및 충청북도 자연 동굴지역, 그리고 충청북도 청원군 등지에서 채집한 205점의 토양시료로부터 분리된 총 1,322주의 방선균을 대상으로 Eschericia coli KCTC 1924, Salmonella typhimurium KCTC 1926, Staphylococcus aureus KCTC 1926, Staphylococcus aureus KCTC 1928, Candida albicans KCTC 1940등 5종의 시험균주에 대하여 항균활성을 조사하였다. 액체배양을 위해서는 G.S.S(glucose-soybean meal-starch) 배지와 fish meal 배지등 2종류의 배지를 사용하였으며 그 결과를 균분리원 지역별, 배지별, 시험균주별, 그리고 방선균 종류별로 비교하였다. It is well known that actinomycetes would be useful for screening of biologically active compounds. Not only to isolate diverse actinomycete strains but to ferment those strains effectively would be important. Seven hundred and forty six strains were isolated from Cheju province, 216 strains were from Chungnam province, 158 strains were from the natural caves at Chungbuk and Kangwon provinces and 202 strains were from Chungwon area at Chungbuk province. All of these 1,322 strains were fermented on a small scale using two different media and tested for their antimicrobial activities against four bacterial strains and one yeast strain. As the result, 12.3% of those isolates were active against Staphylococcus aureus KCTC 1916, 7.6% were Staphylococcus aureus KCTC 1928, 3.9% were Eschericia coli KCTC 1924, 3.0% were Canida albicans KCTC 1940, and 2.2% were Salmonella typhimurium KCTC 1926. About 40% of those isolates showed antimicrobial activities at both two media but the others showed at either one. According to the genus of isolated strains, Streptomyces and Micromonospora showed activities with higher frequencies than others.
