      • Use of transrenal DNA for the diagnosis of Tuberculosis

        There have been attempts to use biological samples other than sputum for the diagnosis of TB. Urine is promising clinical specimen because of its availability, ease of access, processing and storage, and the low infection risk to healthcare workers during sample collection. In this study we evaluated the outcomes of urine diagnostics in hospitalized MDR, XDR pulmonary TB patients. In order to detect TB-specific transrenal DNA, 10 urine samples were collected during the study period, and quickly added to 1 ml of a stabilization buffer to a final concentration of 10 mM, in order to minimize degradation of soluble DNA by nucleases. DNA was extracted from 8ml of whole urine and eluted in 25ul with an automated system based on magnetic silica particle technology. We used droplet PCR, it might be an ideal alternative to conventional real-time PCR for microorganism detection. We aimed to assess the capacity of ddPCR for detecting low levels of circulating Mycobacterium tuberculosis DNA target (known as IS6110). ddPCR was found to provide results in good agreement with the other methods tested and to be highly reproducible.In this study, the results show that ddPCR could be used to measure low levels of MTB DNA, and it has the potential to be used to diagnose pulmonary TB based on clinical samples. The application of the Tr-DNA test could provide a significant advance in the diagnosis of TB. The availability of new method using urine that is more convenient and accessible may greatly usefulness for individual hospitalized patients for monitoring their treatment process.

        Reversible Splenial Lesion in the Corpus Callosum on MRI after Ingestion of a Herbicide Containing Glufosinate Ammonium: A Case Report

        Glufosinate ammonium (GLA) is a broad-spectrum herbicide used worldwide. Transient lesions involving the splenium of the corpus callosum have been described in patients with encephalitis or encephalopathy of varied etiology, but have not been reported in association with GLA poisoning. We describe a patient who had attempted suicide by ingesting GLA-containing herbicide, with a focal diffusion-restrictive lesion in the splenium of the corpus callosum on MRI, which had disappeared four weeks later with no sequalae.

        3대째 우성유전된 Disseminated Superficial Actinic Porockratosis

        Disseminated superficial actinic porokeratosis (DSAP) has an autosomal dominant mode of inheritance with reduced penetration at young age, characterized by multiple superficial keratotic lesions surrounded by a slightly raised border. The aggrevation of the lesion by exposing UV light indicates that phenotypic expression of gene regluating DSAP is related to sun light. We observed a family of which 13 members had DSAP in 3 successive generations. They were grandrnother, two sons of her 3 offsprings, 2 grandsons and 8 granddaughters. Lesions of all cases were found on the sun exposed areas, especially, face, extremities and upper trunk but exceptionally one had a lesion on the unex posed area, abdomen below the iliac crest. The number of lesions of the individual patient were 12-116, and there appeared to be increasing number of lesions with age.

        Enhanced structural and electrical features of amorphous InGaZnO thin film transistors via a heavy Kr gas process

        We report the influence of a heavy Kr gas sputtering process on the electrical and structural features of amorphous indiumegalliumezinc-oxide (a-IGZO) thin film transistors (TFTs). Electrical observation revealed effective reduction of threshold voltage shifts by adapting a simple sputtering process with Kr gas during growth of a-IGZO TFTs. In addition, the application of Kr-gas resulted in a reduction of oxygen vacancies associated with defect sites in the a-IGZO active channel layer. Structural analyses including atomic force microscopy, X-ray reflectivity, Auger electron spectroscopy, and X-ray photoelectron spectroscopy depth profiling were carried out, along with electrical bias stability tests that convincingly confirm progress of heavy Kr gas process-induced features.

        Segmentation of Neuronal Axons in Brainbow Images

        In neuroscientific research, image segmentation is one of the most important processes. The morphology of axons plays an important role for researchers seeking to understand axonal functions and connectivity. In this study, we evaluated the level set segmentation method for neuronal axons in a Brainbow confocal microscopy image. We first obtained a reconstructed image on an x–z plane. Then, for preprocessing, we also applied two methods: anisotropic diffusion filtering and bilateral filtering. Finally, we performed image segmentation using the level set method with three different approaches. The accuracy of segmentation for each case was evaluated in diverse ways. In our experiment, the combination of bilateral filtering with the level set method provided the best result. Consequently, we confirmed reasonable results with our approach; we believe that our method has great potential if successfully combined with other research findings.

      • 의원입법 규제영향평가의 개념과 대안: 한국규제학회의 분석경험을 중심으로

        This paper aims at showing the necessity of some kinds of the Regulatory Impact Assessment on the law proposals by the Korean Legislature and at proposing the institutional change needed. Chapter I and II illustrate and show why it is necessary to somehow monitor and evaluate the law proposal by the Korean Legislature, in terms of the over-production and quality of the proposals for the last couple of years. Chapter III tries to show that there are some formal or informal institutional settings of the Regulatory Impact Assessment on the law proposal by the legislative branch in the lawmaking process of the so-called advanced countries, such as the USA, England, France, Germany and so on. In addition, Chapter IV tries to extract some learnings from the Korea Regulatory Studies Association’s experience of the monitoring the regulatory legislative law proposals by the Korean congressmen for the last two years, regarding the purpose, potential use, evaluation tools and processes of the Regulatory Impact Assessment. Finally Chapter V concludes with the alternatives and author’s recommendations on the institutional and policy settings and priorities of the Regulatory Impact Assessment system on the law proposal by the Korean Legislature. 본 연구는 우리나라 의원입법 규제에 대한 규제영향평가 도입의 필요성과 그 방안 등을 제안하는 것을 목적으로 한다. 이를 위하여 제I~II장에서는 의원입법 규제에 대한 관찰과 심사가 필요한 배경과 이유를 최근 우리나라 국회의 입법 동향을 중심으로 피력하였다. 제III장에서는 선진국의 제도를 음미하여 비록 공식적으로는 의원입법 규제에 대한 규제영향평가 제도가 존재하지 않을 수도 있으나, 그 기능은 관례적으로나 실질적으로 구현되고 있음을 보이고자 하였다. 제IV장에서는 의원입법 규제에 대한 규제영향평가의 일종의 사례로서, (사)한국규제학회가 지난 2년간 수행한 의원입법 규제의 모니터링에 대한 개요 및 평가지침, 관련 경험, 향후 분석 전망 등을 제시하였다. 이는 향후 규제영향평가 제도 도입과 관련하여 평가의 틀 및 수행방식 등의 체계를 마련하는 데 있어 참고할 여지가 있을 것으로 판단하였기 때문이다. 마지막으로 제V장에서는 의원입법 규제에 대한 규제영향평가의 정책 대안들을 보여주고 저자가 주장하는 정책의 우선순위와 대안들을 제시하였다.

        Evaluation of Volumetric Texture Features for Computerized Cell Nuclei Grading

        The extraction of important features in cancer cell image analysis is a key process in grading renal cell carcinoma. In this study, we applied three-dimensional (3D) texture feature extraction methods to cell nuclei images and evaluated the validity of them for computerized cell nuclei grading. Individual images of 2,423 cell nuclei were extracted from 80 renal cell carcinomas (RCCs) using confocal laser scanning microscopy (CLSM). First, we applied the 3D texture mapping method to render the volume of entire tissue sections. Then, we determined the chromatin texture quantitatively by calculating 3D gray-level co-occurrence matrices (3D GLCM) and 3D run length matrices (3D GLRLM). Finally, to demonstrate the suitability of 3D texture features for grading, we performed a discriminant analysis. In addition, we conducted a principal component analysis to obtain optimized texture features. Automatic grading of cell nuclei using 3D texture features had an accuracy of 78.30%. Combining 3D textural and 3D morphological features improved the accuracy to 82.19%. As a comparative study, we also performed a stepwise feature selection. Using the 4 optimized features, we could obtain more improved accuracy of 84.32%. Three dimensional texture features have potential for use as fundamental elements in developing a new nuclear grading system with accurate diagnosis and predicting prognosis.

        Estimation of Water Quality Model Parameters

        A modified Gauss-Newton method was utilized to estimate water quality model parameters, which is important in the modeling of water quality transport process. This method served the unknown parameters of three engineering problems exceptionally well. We applied this method to obtain a set of water quality parameters by solving the kinetic equations for dissolved and particulate water quality species during episodic events in the Indian River Lagoon, FL. The method was successful in producing water quality model parameters, and the relative errors between measurements and model predictions are below 46.4% and 39.4% for all dissolved and particulate water quality state variables, respectively. In this study we evaluated a good capability of suggested parameter estimation method by comparing a couple of water quality model simulations: (1) water quality model simulation using model parameters produced by the modified Gauss-Newton method and (2) water quality model simulation employing model parameters obtained by the trial-and-error method. The average relative error and ROC score between observed data and water quality model results using model parameters calculated by the modified Gauss-Newton method are 58.3% and 0.565, respectively. In the case of water quality model simulation using model parameters obtained by the trial-and-error method, the average relative error and ROC score are 60.5% and 0.555, respectively. This suggests that the modified Gauss-Newton method efficiently yields water quality model parameters resulting in better or similar model prediction errors when compared to the trial-and-error method.
