The dissociation constant of erythrosine in the mixed solvent systems of water-alcohols and water-acetone at various pH and in concentration media were determined by spectrophotometric method. Methanol, ethanol, and acetone were employed as solvents and studied the changes of pK values of erythrosine according to the kind and change of concentration of organic solvent which is mixed into water solution. The effect of organic solvent on pK is negligible at 40% while in the case of above 40% mixing, it is not negligible.

Microanalysis of rodenticide by visible absorption method was attempted. The following procedure was found to be very simple and to give accurate results. RH-28787 was added to dimethylformamide (DMF), and extracted after stirring for about 20 minutes in extractor (30℃). After extraction, the absorbance of DMF solution was measured at 346 ㎚ by using spectrophotometer. The result agrees closely with the experimentally determined value by means of the gas chromatograph.

Antidiabetic activity of several herbal drugs, which are used as antidiabetics in folkmedicine, was evaluated. Among tested herbal drugs, extract of Astragali Radix significantly lowered blood glucose level in streptozotocin-induced diabetic model rat.

Systematic study on the extraction of ion pair by the use of picric acid (PCA) as an ion pair forming reagent to the aliphatic amines has not be done by spectrophotometric method. The extraction of ion pair by the use of PCA and 23 kinds of the aliphatic amines was examined. The procedure is as follows; Elving''s buffer solution (pH 1.3-10, ionic strength 0.5) each amine solution, and PCA solution were taken into a test tube. The mixture was shaken mechanically with chlorform. The organic phase was filtered through a filter paper to remove water droplets. The absorbance was examined at lambdamax against a reagent blank. Primary and secondary whose carbon number were more than 7 or 6, respectively, are extractable as ion pairs with PCA, while tertiary amines and quaternary ammonium salts are also extractable without the correlation of carbon number. It was considered that the ion pair extraction of primary and secondary amines was affected by the number of carbon of amines, but its extraction of tertiary amines or quaternary ammonium salts was affected by kind of aliphatic amines rather than pKa values or carbon number of amines.

HPLC/fluorescence detection method for the analysis of pancuronium bromide in biological fluids was developed. The method depends on the formation of insoluble red complex between pancuronium bromide and rose bengal in aqueous layer. This complex is quantitatively extracted from aqueous layer into chloroform layer. The complex is stable for 1 day in chloroform layer at room temperature. It was possible to analyze pancuronium bromide in the range of 0.05-0.5 mcg/ml without the effect of co-prescribed drugs.

박층크로마토그라피 및 농도 측정법에 의한 Ampicillin Trihydrate 중의 N,N-Dimethylaniline의 검색

A simple and convenient method of detecting N, N-dimethy laniline (D.M.A.) residues in ampicillin trihydrate (A.T.) was established. D.M.A. was extracted by chloroform from the chloroform presaturated N/10-ammonia water solution of A.T. and chromatographed on silica gel G thin layer. Blue spot appeared in 15minutes after spray of 2, 6-dichloroquinonechlorimide solution was compared with the blue spot of reference concomitantly processed. The developing solvent was prepared by mixing equal volume of cyclohexane and chloroform. To quantitate the amount of D.M.A. in A.T., T.L.C. was performed with the Eastmann Chromatogram sheet, then color density was measured by Cosmo Superclick densitometer. The developing solvent at this time was cyclohexane-chlorofonn (3+7) mixture. The peak areas obtained with the amount of D.M.A ranging from 0.05 to 2.0 .mu.g were linear to color density. Better sensitive results would be available with the densitometer equipped with monochromator.
