        Efficient Cultivation Conditions for Human Limbal Epithelial Cells

        To compare the stem niche in different culture conditions of limbal epithelial cells, the suspended human limbal epithelial cells (HLECs) were seeded on the 3T3-pretreated plates and the other suspended cells were plated on amniotic membranes (AMs) which were either cryo-preserved or freeze-dried. All were cultured for 10 to 12 days. Reverse transcription-polymerase chain reaction (RT-PCR) for ATP-binding casette, subfamily G, member 2 (ABCG2), p63, cytokeratin 12, and connexin 43 were performed in cultivated HLECs and their expression levels were compared. The mRNA expression of all markers examined showed no statistically significant differences between the cells on cryo-preserved and on freeze-dried AM. The expression of p63 and cytokeratin 12 in cultivated cells on AMs were significantly lower than those in 3T3-cocultured cells on RT-PCR and immunofluorescent staining. Cultivated HLECs on AMs showed reduced proliferation and differentiation while maintaining stem-property regardless of the preservative method of AM.

        汉韩书面省略句比较研究 - 以中韩2019年新年贺词为例

        Ellipsis is a common linguistic phenomenon, which not only helps to construct cohesion and coherence among sentences in context but also facilitates to highlight new information. Therefore, there is ellipsis in both spoken and written texts. Ellipsis is also an old problem in the study of Chinese and Korean. In the past, Chinese and Korean grammarians mainly focused on the elements omitted and ellipsis types, and less research was done on the comparative study of ellipses in Chinese and Korean. To be concrete, that is almost in a blank state. This paper chooses 2019 New Year's greetings of the two country leaders as the example to get the overall statistics of ellipsis use frequency and intends to find out whether there is difference between Chinese and Korean written language resulted from diversified languages, different thinking styles and cultural distinctions by means of comparing the following three aspects: sentence structure, syntactic component and recall constituent types so as to prove the prevalence of elliptical sentences. Furthermore, not all the ellipsis phenomena are the same. In Chinese President Xi jinping's 2019 New Year's speech, ellipses account for 77%, most of which occur in clauses. It is common to omit time adverbial and subject due to the linguistic context. While in President Moon Jaein's 2019 New Year's speech, 61% of the ellipses take place in the subordinate structure, and it is common to omit the subject due to the situational context.

        DNA Microarray-Based Gene Expression Profiling in Porcine Keratocytes and Corneal Endothelial Cells and Comparative Analysis Associated with Xeno-related Rejection

        Porcine to rat corneal xenotransplantation resulted in severe inflammation and rejection of the corneal stroma, whereas an allograft showed mainly endothelial cell-associated rejection. We, therefore, investigated and compared the gene expression between porcine keratocytes and corneal endothelial cells. RNA was isolated from primary cultured porcine or human keratocytes and porcine corneal endothelial cells. Gene expression was comparatively analyzed after normalization with microarray method using Platinum pig 13 K oligo chip (GenoCheck Co., Ltd., Ansan, Korea). Real-time polymerase chain reaction (PCR) was performed for C1R, CCL2, CXCL6, and HLA-A in porcine keratocytes and corneal endothelial cells. As a result, upregulated expression more than 2 folds was observed in 1,162 genes of porcine keratocytes versus porcine endothelial cells. Among the immune-regulatory genes, SEMA3C, CCL2, CXCL6, F3, HLA-A, CD97, IFI30, C1R, and G1P3 were highly expressed in porcine keratocytes, compared to porcine corneal endothelial cells or human keratocytes. When measured by real-time PCR, the expression of C1R, CCL2, and HLA-A was higher in porcine keratocytes compared to that in porcine corneal endothelial cells. In conclusion, the increased expression of C1R, CCL2, and HLA-A genes in porcine keratocytes might be responsible for the stromal rejection observed in a porcine to rat corneal xenotransplantation.

        VOF법과 DEM에 의한 사석구조물 변형예측모델과 그 적용성에 관한 연구

        A numerical model, which can compute deformation of rubble mound structures composed with various size materials, was proposed. In the numerical model, wave field into the mound structures was computed by CADMAS-SURF and the deformations of mound structures were computed by DEM. Interaction between wave field and sectional deformation of structure was considered and to present the variation of behaviors caused by various properties of materials, computation was carried out with random coefficients by Monte Carlo simulation method for contact stiffness and friction angle. The experiments were carried out with rubbles and glass balls with radius of 2.9cm, 2.6cm and 1.5cm. And the deformation characteristics of rubble mounds composed with various size materials were clarified. Furthermore the validity and the applicability of the model were discussed by comparing with the experimental results. 사석 방파제와 같은 불연속 구조물의 안정 해석이 수행되었다. 파랑장의 계산에는 CADMAS-SURF를 이용하고 마운드의 변형 해석에는 개별요소법을 이용하였다. 파랑장과 구조물 변형과의 상호작용을 고려하였고, 마운드를 구성하는 요소의 다양성에서 비롯되는 물성치들을 고려하기 위해 스프링계수, 점성, 마찰계수를 몬테카를로법에 의해 랜덤하게 입력하였다. 또한 수치해석 결과를 검증하고 해석 모델의 타당성을 검토하기 위해 모형실험을 실시했다. 모형실험에서는 마운드를 구성하는 요소로써 유리구를 이용했고, 실험결과와의 비교를 위하여 요소의 크기를 수치해석의 모델요소와 일치시켰다. 그 결과 수치해석결과와 모형실험결과가 정성적으로 일치했으며, 본 해석 모델의 타당성이 검증되었다.

        형광 흥분세포분리를 이용한 추정 윤부상피줄기세포의 분리

        Purpose: To analyze the isolating pattern of slow cycling cells as putative limbal epithelial stem cells (PLESCs) using Hoechst exclusive cell sorting. Methods: Rabbits were injected with 5-bromo-2-deoxyuridine (Brd U) 1 month prior to be sacrificed. After obtaining limbal tissues, fluorescence-activated cells were sorted on a Coulter EPICS 753 after they had been incubated with Hoechst 33342 and propidium iodide. Two different methods were applied to sort PLESCs. Side-population(Sp) cells were obtained using gates with dichroic mirror to detect low Hoechst blue and red after verapamil was treated. Hoechst negative cells were obtained using gates exhibiting low Hoechst blue with a 424/44 BP filter. Brd U-retaining cells were counted and their sizes were evaluated in each gated sample to compare isolating pattern of PLESCs in each method. Results: The percentages of Sp cells and of the Hoechst negative fraction were 0.96±0.79% and 16.01±13.60%, respectively(p=0.021). Homogeneity and density of the small cells were higher in Hoechst negative fraction than in Sp cells. The percentage of Brd U-retaining cells was 47.36±10.34% and 47.14±14.94% in Sp cells and Hoechst negative fraction, respectively(p>0.05), and they were 10 times higher than in non-Sp and Hoechst positive fraction(p=0.000). Conclusions: Hoechst negative exclusion without verapamil more efficiently isolated PLESCs than Sp did.

      • DEM을 이용한 해안구조물 변형해석

        In this paper I proposed a model that the deformation of the submerged rubble mound breakwaters composed with various size materials induced by the wave can be computed. The water particle kinematics by waves in porous mound structure are computed by CADMAS-SURF, then the deformation of structure is computed using OEM module. To investigate the interaction of wave and sectional deformation of structures, analysis is accomplished by two steps analysis of incipient mound at the rust step and that of the deformed mound by wave action at the second step. Furthermore, behaviors of materials are influenced by various properties such as the contact stiffness and the friction angle. Therefore, in order to present the behavior of the element caused by various properties, computations are accomplished with random coefficients by using the Monte Carlo simulation.
