        In vitro Synthesis of Ribonucleic Acids by T7 RNA Polymerase That was Fast Purified with a Modified Procedure

        세포 내에서 매우 적은 양으로 존재하는 RNA 전사체와 기능적으로 동일하거나 비슷한 RNA를 RNA 중합효소를 써서 in vitro에서 생화학적으로 의미 있는 양 만큼을 합성할 수 있다. T7 RNA중합효소를 발현하는 재조합 유전자를 지닌 대장균주 BL21/pAR1219로부터 순수한 T7 RNA중합효소를 손쉽게 얻는 방법을 본 논문에서 소개한다. 황산암모늄 분획화와 sephadex SP 컬럼 크로마토그래피법으로써 여타의 방법과 비교하여 더 간단하고 빠르게, 그리고 경제적으로 T7 RNA 중합효소를 분리할 수 있었다. 정제된 T7 RNA중합효소를 이용하여 보통의 화학적 합성법으로 불가능한 긴 길이(1.54 kb)의 RNA전사체를 합성 하였다. 한편,정제된 T7 RNA중합효소에 의해 생성된 망치머리 리보자임은 표적 RNA를 in vitro에서 절단함으로써, 생성된 RNA가 생화학적 기능성을 유지한다는 것을 입증하였다 따라서 본 연구에서 소개되는 절차들은 다양한 길이의 RNA를 목적에 따라 간단하고 경제적으로 합성하는데 유용하게 이용될 수 있다. Biochemical amounts of RNA molecules can be synthesized in vitro, which is functionally equivalent or similar to those transcripts normally existing at extremely low levels in vivo. In this study we described a method for efficient preparation of pure T7 RNA polymerase from Escherichia coli strain BL21/pAR1219. The procedure, which used ammonium sulfate fractionation and preparative column chromatography on sephadex SP, was shown to be simple, rapid, and cost effective in comparison with other methods reported previously, Using the purified T7 RNA polymerase we were able to synthesize very long RNA transcript of 1.54 kb length, which is not feasible by conventional chemical synthesis. RNA molecule that was also synthesized by the purified T7 RNA polymerase, such as hammerhead ribozyme, retained its biochemical activity by cleaving the target RNA successfully in vitro. Thus, the procedure shown in this study can be useful to synthesize any length of RNA molecules in vitro in a simple and cost effective way for a variety of purposes.

        The choice of a geostationary orbit for telecommunications has been recently reevaluated for the personal communications, demanding reasonable size antenna and receivers and quality voice services with reasonable delays, which pushes new classes of satellite communications to exploit the lower orbits. The new satellitebased systems, so called Global Mobile Personal Communication Systems, are expected to be deployed and start services in a few years. In this paper, Odyssey, employing 12 Medium Earth Orbit satellites in 3 orbit planes is reviewed. It is observed as an alternative among various Global Mobile Personal Communication Systems, and the orbit altitude and constellation configurations are described. Furthermore, fundamental technical parameters including uplink and downlink power budgets are tabulated to see<br/> performance of the communication links, especially with considering interferences between other satellite-based systems.<br/>

      • 수평연속공정으로 주조된 회주물의 열처리 영향에 관한 연구

        This study was based on the examination of the microstructure depending on the heat-treatment temperature and cooling rate of the gray cast ion produced of fabricated by the horizontal continuous casting method, The heat-treatment were performed at 900, 1,000, and 1,100℃. for 1 hour, respectively, The cross sectional microstructures of the as-cast and the treated specimens were observed by the optical microscope and the hardness numbers were measured by Rockwell hardness tester. The type A and type B graphites were observed on the center of the as-cast specimen. The matrix of the specimen was ferrite structure. The dendrite structures and the type D graphites were formed at the outside of the matrix. The pearlite and the nodular graghite in the matrix were formed by the dissolution of the graphites according to the heat treatment temperature increase. The martensite structure was observed in case of water cooling. The pearlite structure and the martensite structure were formed in case of heat-treatment at 1,100℃. The hardness number of treated specimens with the pearlite and martensite matrix increased.

        본 연구는 비대칭-팽이 분자인 CH_(2)O가스의 기본진동 6개중 A형인 v_(2) 띠를 택하여, 고분해 진동-회전 스펙트럼을 이론적으로 계산하여 합성하고, 측정된 적외선 스펙트럼과 비교 검토 하였다. 여기서 진동수를 계산하기 위하여 Fortran프로그램을 사용하였으며, 방정식은 CH_(2)O의 세 관성 모멘트 중 2개가 거의 같은 성질을 이용하여 비대칭-팽이분자의 방정식으로부터 근사식을 사용하였다. 이론 스펙트럼에 나타난 회정양자수 J와 그투사 양자수 K를 실험 스펙트럼에서 확인 할 수 없었으나 분해능이 좀 더 나은 분광기를 사용하면, 확인 할 수 있을 것이다.

      • Procast를 이용한 AISI 316합금의 주조방안 연구

        In order to improve the casting process of a steel casting, the casting design method of AISI 316 alloy was investigated using a Procast. The design for a casting and a gate system were performed by solidworks. The analysis of solid fraction, solidification time, shrinkage and pore for AISI 316 models have been done with a Procast. As a result of solidification time, steep thermal gradient was observed where the thickness of casting change. The probability of macrocrack formation increased at casted area between large head feeders because of large cooling rate difference. Shrinkage was predicted around head feeders with medium thermal gradient. It were very important to increase the molten pressure and to maintain a gate with smooth curvature in order to get a good flow. The larger head feeder was, the less directional solidification was. Head feeder, especially it's volume and location, gave an strong influence on the casting defects.

        Abstract From years ago, 'Sul' was called differently according to various nations. They were called as 春節(chiness), car γ an ca γ z(Mongolia), 元旦(Manchu), Sul(korea) etc During this peried, the folk scholar assumed the name of 'Sul' as below. First, according to the record, the first day of the year was thought ad the meaing of lonely and sadness. In this situation, We used to be careful. Second. 'Sul' has the meaning of age where we used to call as 'Sool' on the old days. But lately it has changed to 'Sul' Third, the name of 'Sul' came from the Sanscrit where the word 'Sal' has the meaning of growing. Fourth, in the Chinese Character 'Sul' has the meaning standing '立'or building '建'. Fifth, from years ago, the researchers wonders the word 'Sul' has changed in the middle century as the meaning of age. So when years past, the ages become more at the same time. According to history the 'Sul' of Korea and Mongolia was continued until 3~4 century(B. C) According to history the 'Sul' of Manchu was affected mostly bu China but in Manchu thyu strenth their specialities and expressions. In Mongolia there are various kinds of customs. But they think 'Sul' as the special and January is the stsrting point of the year for the Mongolia They called this month as 'white month'. They belive white as the symbol of purity
