        Preliminary Application of Molecular Monitoring of the Pacific Herring (Clupea pallasii) Based on Real-time PCR Assay Utilization on Environmental Water Samples

        Pacific herring, Clupea pallasii, a keystone species with significant ecological and commercial importance, is declining globally throughout much of its range. While traditional fishing equipment methods remain limited, new sensitive and rapid detection methods should be developed to monitor fisheries resources. To monitor the presence and quantity of C. pallasii from environmental DNA (eDNA) extracted from seawater samples, a pair of primers and a TaqMan® probe specific to this fish based on mitochondrial cytochrome b (COB) sequences were designed for the real-time PCR (qPCR) assay. The combination of our molecular markers showed high specificity in the qPCR assay, which affirmed the success of presenting a positive signal only in the C. pallasii specimens. The markers also showed a high sensitivity for detecting C. pallasii genomic DNA in the range of 1 pg~100 ng rxn-1 and its DNA plasmid containing COB amplicon in the range of 1~100,000 copies rxn-1, which produced linear standard calibration curves (r2 = 0.99). We performed a qPCR assay for environmental water samples obtained from 29 sampling stations in the southeastern coastal regions of South Korea using molecular markers. The assay successfully detected the C. pallasii eDNA from 14 stations (48.2%), with the highest mean concentration in Jinhae Bay with a value of 76.09±18.39 pg L-1 (246.20±58.58 copies L -1). Our preliminary application of molecular monitoring of C. pallasii will provide essential information for efficient ecological control and management of this valuable fisheries resource.

        Complete Mitogenome of the Russian Sturgeon Acipenser gueldenstaedtii (Acipenseriformes; Acipenseridae)

        Sturgeons and paddlefishes are frequently referred to as ‘living fossils’ among the action-pterygian lineage. They are increasingly facing threats to their existence because of various anthropogenic pressures. In this study, we present the complete mitogenome sequence of the Russian sturgeon Acipenser gueldenstaedtii (Acipenseriformes; Acipenseridae). The mitogenome showed highly homogeneous molecular features compared to previously known vertebrate mitogenomes. Phylogenetic tree inferred from concatenated protein-coding and tRNA genes unambiguously revealed the monophyly of A. gueldenstaedtii, Acipenser stellatus, and Huso huso. Genetic information of the endangered A. gueldenstaedtii will provide baseline data needed to develop molecular markers for stock identification and assessment of population diversity and also to develop future conservation strategies. geon

        Phylogenetic Relationships Among Diverse Dinoflagellate Species Occurringin Coastal Waters off Korea Inferred from Large Subunit Ribosomal DNASequence Data

        We analyzed the nuclear-encoded large subunit ribosomal RNA gene (LSU rDNA) sequences of 19 dinoflagellates occurring in costal waters off Korea and reconstructed a phylogenetic tree containing 74 representative species from 37 distinct genera. Of these, the LSU rDNA sequences of Amylax triacantha (Jörgensen) Sournia, Gonyaulax verior Sournia (= Amylax diacantha Meunier), Gyrodinium fissum (Levander) Kofoid et Swezy, Katodinium glaucum (Lebour) Lebour III, Noctiluca scintillans (Macartney) Kofoid et Swezy, Oxyphysis oxytoxoides Kofoid, and Pyrophacus steinii (Schiller) Wall et Dale are reported for the first time. Our LSU rDNA tree consistently placed Oxyrrhis marina Dujardin and N. scintillans at the most primitive positions, giving rise to a strongly supported monophyletic group of typical dinoflagellate species belonging to the Dinophyceae. The phylogenetic relationships among the typical dinoflagellates, however, were not resolved in the higher taxonomic levels in general. Only genera at terminal branches were usually supported with high confidence. The Dinophysiales, represented by Dinophysis species and O. oxytoxoides, formed a strongly supported monophyletic assemblage. The Gymnodiniales and Peridiniales were recovered as polyphyletic groupings. Members of the Gonyaulacales were consistently grouped together, but lacked statistical support. Within this order, the Ceratiaceae and Goniodomataceae each formed a monophyletic group, but the Gonyaulacaceae was polyphyletic. The phylogenetic relationships of the Gonyaulacaceae were generally congruent with differences in the combinations of the apical pore complex, hypothecal organization and thecal formula.

        황해 부유 녹조 면적 산출을 위한 멀티 위성센서 활용

        황해 녹조 대발생은 2008년 이후 매년 여름 대규모로 발생하고 있으며, 인공위성 분석을 통해 녹조 규모를 추정하기 위한 연구가 활발하게 진행되고 있다. 하지만 정확한 녹조 면적 산출을 위한 위성영상 선택 기준이 마련되어 있지 않은 실정이다. 따라서 이 연구에서는 위성영상의 공간해상도에 따른 녹조 면적 산출 결과의 차이를 알아보고, 녹조 면적 산출에 적합한 위성영상을 제시하고자 한다. 이 연구에서 이용한 위성영상은 Landsat ETM+, MODIS, GOCI로 영상의 공간해상도는 각각 30, 250, 500 m 이다. NDVI 알고리즘을 적용하여 녹조 픽셀을 분류하였고, 임계값에 따른 녹조 면적 산출 결과를 비교하였다. 또한, Linear Spectral Unmixing(LSU) 기법을 이용하여 한 픽셀 내에 녹조가 차지하는 비율을 추정하였고, 녹조 비율의 차이가 면적 계산에 미치는 영향을 평가하였다. NDVI 알고리즘을 이용한 녹조 면적 산출 결과, 공간해상도가 낮을수록 녹조 면적이 과대추정되는 경향을 보였고, 최대 1.5배 차이를 보였다. 또한, LSU 분석 결과에서 픽셀 내의 녹조 비율이 0.1(10%) 미만인 픽셀이 대부분이었고, 0.5(50%) 이상 녹조 비중을 차지하는 픽셀은 세 타입의 영상 모두에서 약 2% 수준으로 극히 적었다. 즉, NDVI 분석 결과에서 녹조로 분류된 픽셀의 경우 한 픽셀의 공간을 녹조가 100% 채우고 있지 않더라도 모두 동일 면적으로 간주되기 때문에 실제보다 과대추정되는 것으로 생각되고, 이러한 현상은 공간해상도 차이에 의해 심화되는 것으로 나타났다. The massive green tide occurred every summer in the Yellow Sea since 2008, and many studies are being actively conducted to estimate the coverage of green tide through analysis of satellite imagery. However, there is no satellite images selection criterion for accurate coverage calculation of green tide. Therefore, this study aimed to find a suitable satellite image from for the comparison of the green tide coverage according to the spatial resolution of satellite image. In this study, Landsat ETM+, MODIS and GOCI images were used to coverage estimation and its spatial resolution is 30, 250 and 500 m, respectively. Green tide pixels were classified based on the NDVI algorithm, the difference of the green tide coverage was compared with threshold value. In addition, we estimate the proportion of the green tide in one pixel through the Linear Spectral Unmixing (LSU) method, and the effect of the difference of green tide ratio on the coverage calculation were evaluated. The result of green tide coverage from the calculation of the NDVI value, coverage of green tide usually overestimate with decreasing spatial resolution, maximum difference shows 1.5 times. In addition, most of the pixels were included in the group with less than 0.1 (10%) LSU value, and above 0.5 (50%) LSU value accounted for about 2% in all of three images. Even though classified as green tide from the NDVI result, it is considered to be overestimated because it is regarded as the same coverage even if green tide is not 100% filled in one pixel. Mixed-pixel problem seems to be more severe with spatial resolution decreases.

        수중음향과 Kompsat-2 위성영상을 이용한 해초지 분포 추정

        해초지의 생태적 중요성에도 불구하고 국내 연안에 분포하는 해초지 규모에 대한 정보가 미비하다. 장흥군 회진면 일대의 해초지를 대상으로 수중음향측심기와 고해상도 Kompsat-2($4{\times}4m$) 위성영상을 이용하여 식생유무를 탐지하고 분포크기를 파악하는 연구가 수행되었다. 위성영상을 이용한 식생분석의 정확도는 음향측심기를 통해 얻은 자료분석과 이를 비교하여 검증되었다. Kompsat-2 영상분석으로 계산된 회진면 일대의 해초지 면적은 약 $3.9km^2$로 수중음향 탐사를 통해 구해진 $4.5km^2$ 보다 과소평가 되었다. Kompsat-2 위성영상을 객체기반 영상분류법으로 해초 식생을 분석한 결과는 수중음향 결과 값에 대해 90%의 정확도를 보였는데, 이와 같이 높은 정확도는 Kappa 지수(0.85)로도 확인되었다. 또한 위성영상과 수중음향 결과 간의 유사도는 77.1%로 비교적 높았다. 생물 비파괴적인 수중음향조사와 Kompsat-2 영상분석으로 국내 연안에 산재해 있는 해초지 식생의 광역적인 조사가 가능할 것으로 기대되며, 보다 정확한 탐지를 위해서 다양한 고해상도 위성을 이용한 연구가 활발히 이루어져야 할 것이다. Despite the ecological importance of seagrass beds, their distributional information in Korean coastal waters is insufficient. Therefore, we used hydroacoustic system to collect accurate bathymetry and classification of seagrass, and Kompsat-2 (4 m spatial resolution) image for detection of seagrass beds at Deukryang Bay, Korea. The accuracy of Kompsat-2 image classification was evaluated using hydracoustic survey result using error matrix and Kappa value. The total area of seagrass beds from satellite image classification was underestimated compared to the hydroacoustic survey, estimated 3.9 and $4.5km^2$ from satellite image and hydroacoustic data, respectively. Nonetheless, the accuracy of Kompsat-2 image classification over hydroacoustic-based method showing 90% (Kappa=0.85) for the three class maps (seagrass, unvegetated seawater and aquaculture). The agreement between the satellite image classification and the hydroacoustic result was 77.1% (the seagrass presence/absence map). From our result of satellite image classification, Kompsat-2 image is suitable for mapping seagrass beds with high accuracy and non-destructive method. For more accurate information, more researches with a variety of high-resolution satellite image will be preceded.
