        회생에너지 저장시스템이 제동 브랜딩 안정화에 미치는 영향

        Regenerative Energy Storage System(ESS) is a system that saves regenerative energy which generated instantly in the regenerative braking of Electric Multiple Unit(EMU) and disappear, and reuse the stored energy when EMU is in powering. Such system related to a research field of renewable energy which emerged concerning climate change and high oil prices. In the case of existing domestic rolling stock, about 25% to 30% of generated regenerative energy is restored to power source and is regarded as direct factor of raising catenary voltage. Such rapid change of catenary voltage is a cause of the failure of EMU's electronic equipment and lowering its reliability and is also a cause of train's fault occurred by tripping circuit breaker. In this paper, we intend to investigate the effect on blending characteristics of electric-braking and pneumatic-braking whether the regenerative energy storage system is used or not in urban transit DC 1,500V feeding system, while trains run. And we also intend to investigate its effect on stabilization of the blending, fluctuation of catenary voltage and various electric equipments.

        Pleurotus ostreatus에 의한 리그닌 분해에 미치는 수종 탄수화물의 영향

        To clarify the effects of several carbohydrates on the biodegradation of lignin by Pleurotus ostreatus. The strain was cultured on the media formulated with lignin and carbohydrates such as cellulose, xylan, collobiose, glucose and xylose, which was added individually. The culture mixtures grown 36 days were filtered and then estimated the degree of lignin biodegradation. It was found that the growth of P. ostreatus was stimulated and the depoly-merization was also increased by the addition of carbohydrates. When the carbohydrates were not added, polymerization was apparent in stead of depolymerization. In the case of glucose as an added carbohydrate, the content of lignin by the nitrosolignin method was greatly (about 7.4 times) decreased than control which contains lignin as a carbon source. The peak of lignin at 280nm in UV spectra was decreased about 27% after 27 days of culture. As results, it was assumed that lignin biodegradation was correlated to the carbohydrates and especially glucose was very significant role in lignin degradation.

        Pleurotus ostreatus의 laccase 작용특성

        Extracellular laccase (E.C. 1.10.3.2) from the culture filtrate of Pleurotus ostreatus was purified by ammonium sulfate precipctation, protamine sulfate precipitation, DEAE-Sephadex A-50 ion exchange chromatography and Sephadex G-100 gel permeation chromatography. The molecular weight of the enzyme was estimated by SDS-polyacrylamide gel electrophoresis to be 58,000 and the isoelectric point was 3.75. The optimum temperature for the enzyme was about $45^{\circ}C$ and the optimum pH was 6.5. The enzyme was found to be stable at temperature below $35^{\circ}C$ and rapidly inactivated at higher temperatures. Km values for ferulic acid, vanillic acid, dihydroxyphenylalanine (DOPA) were 48.6.$\mu$M, 0.52mM, and 2.73mM, respectively, which indicates that the enzyme has much higher affinity towards ferulic acid. The reaction products of the enzyme were separated by TLC and HPLC.

        Induction of Extracellular Polyphenol Oxidase from Two White-rot Fungi

        리그닌과 관련된 대표적인 페놀화합물인 ferulic acid, vanillic acid 및 gallic acid와 단백질합성저해항생제인 cycloheximide와 puromycine 중에서 Lentinus edodes JA01 경우는 gallic acid가 Pleurotus ostreatus는 ferulic acid가 각각 가장 효과적인 inducer였다. gallic acid는 2.0mM에서, ferulic acid는 1.0mM에서 induction이 가장 잘 되었다. Inducer의 처리시기는 접종후 L. edodes는 2일후에, 그리고 P. ostreatus는 3일후에 최고의 활성을 나타내었다. 또한 polyphenol oxidase activity는 균주의 생장곡선에 대체적으로 비례하여 증가하였으나, P. ostreatus 경우는 대수기에서 최고의 활성을 나타내고 그 이후 감소하였다. Among the representative phenolic compounds in relation to lignin derivatives and protein synthesis inhibitors, the most effective inducer for the extracellular polyphenol oxidase (PO) of Lentinus edodes JA01 was gallic acid and ferulic acid for Pleurotus ostreatus. Optimum concentration of these inducers was 2.0 mM and 1.0 mM, respectively. Addition of gallic acid after two days culture had the best effect on production of PO enzyme of L. edodes JA01 and for P. ostreatus, and addition of ferulic acid after three days culture had the best effect. Also, in case of L. edodes JA01, polyphenol oxidase activity was parallel to growth curve, whereas the maximum enzyme activity of P. ostreatus was shown at exponential growth phase and declined thereafter.

        The Roles of Polyphenol Oxidase and Glucose Oxidizing Enzymes in the Lignin Metabolism of Pleurotus ostreatus

        To investigate the effects of several carbohydrates on the biodegradation of lignin by Pleurotus ostreatus, the strain was cultured on the media formulated with individually added carbohydrates such as cellulose, cellobiose, glucose, xylan, and xylose. Fractionation of the lignin biodegradation products by Sephadex G-75 column chromatography revealed that polymerization was more apparent rather than depolymerization in the absence of carbohydrates. However, depolymerization and lignin biodegradation were promoted by the addition of carbohydrates. Several inducers such as gallic acid, ferulic acid, 2,5-xylidine, cycloheximide and puromycine were added respectively for the higher production of polyphenol oxidase and it was found that ferulic acid was the most effective inducer. Extracellular polyphenol oxidase was purified by ammonium sulfate fractionation, protamine sulfate precipitation, DEAE-Sephadex A-50 ion exchange chromatography and Sephadex G-100 gel permeation chromatography. The molecular weight of the enzyme was estimated to be 58,000 by SDS-polyacrylamide gel electrophoresis, and the isoelectric point was 3.75. The optimum temperature for the enzyme activity was about 45℃, and the pH optimum was 6.5. The enzyme was found to be stable at temperature below 35℃ and able to oxidize various phenolic compounds containing at least one hydroxyl group regardless of its position. After the reaction of ferulic acid with the purified polyphenol oxidase, the reaction products were separated by TLC and HPLC and analyzed by H-NMR, Mass and IR spectroscopy. The products were found to be styrenoid dimer with two benzene ring called 2-(3'-hydroxy-4'-methoxyphenyl)-4-hydroxy-5-methoxystyrene and styrenoid trimer. Glucose oxidizing enzymes were purified using Sephadex G-200 chromatography, DEAF-Sephadex A-50 ion exchange chromatography and CM-Sephadex chromatography. Glucose dehydrogenase and glucose oxidase activity were detected in the same position during the column chromatography, and polyacryamide gel electrophoretogram indicated that one enzyme showed bifuctional activities. The optimum temperature for their activities was about 60℃ and the pH optima for glucose oxidase and glucose dehydrogenase activity were 4.5 and 6.0 respectively. The activities of these two enzymes showed different heat stability: the half life of oxidase activity was 2 hours at 40℃, while dehydrogenase was 1 hour at 80℃. In order to investigate the function of glucose oxidase and glucose dehydrogenase, the reaction mixture of polyphenol oxidase, glucose oxidizing enzymes and ferulic acid was incubated in the presence and absence of O₂ and the reaction products were also analyzed by TLC. In the presence of O₂, the substrate was converted synergetically to the products when glucose oxidizing enzymes were added with polyphenol oxidase and ferulic acid. In the view point of electron tranfer system involved in lignin metabolism, the results suggest that the polymerization of lignin was promoted by supplying H₂O₂ from glucose oxidase to polyphenol oxidase-glucose oxidizing enzyme system in the presence of O₂. In the absence of O₂, however, no reaction can proceeded due to the inhibition of polyphenol oxidase.

        탄수화물과 황산암모늄이 Pseudomonas diminuta의 리그닌 분해에 미치는 영향

        To investigate the influence of cosubstrate supplement and ammonium sulfate on lignin degradation by Pseudomonas diminuta KM-4-2, isolated in the laboratory, the strain was cultured on the lignin media which contained lignin as a source of carbon and the culture filtrate was analyzed by Sephadex G-75 column chromatography. It was found that polymerization was not appeared unlike wood-rot fungi. When the carbohydrates were added, the peak of lignin at 280nm by UV scanning spectra of the filtrate, was significantly increased. In order to determine the effect of ammonium sulfate on the ligninolytic activity, the isolated strain was incubated in the media containing 0.1%, 0.25% and 0.5% of nitrogen concentration in the Warburg flask and the rate of oxygen uptake was esitmated by Warbuge Respirometer. As a result, the activity was maximum at 0.1% of nitrogen concentration and thereafter decreased in parallel with nitrogen concentration.

        연어 회귀 하천인 양양 남대천에서 수질과 섬유소 분해율의 계절적 분석

        The analyses of physicochemical factors and the distribution of microbial populations were measured at seven sites from May, 1994 to April, 1995 in Yangyang Namdaecheon river, Kwangwon-do. Abrupt increases of nitrogen and phosphate salt values were showed from area below the Yangyang old bridge, Ohsackcheon and Namdaecheon, parallel with the increase of cellulose degradation rate. The cellulose degradation rate was highly correlated with the degree of temperature, the concentration of NO_3-N, the population of fecal coliform bacteria and heterotrophic bacteria. The attached bacteria on cellulose film at Ohsackcheon were succeeded from high density of rod form to low density of filamentous form.
