The lateral hoof of right hind limb of a pregnant Masai giraffe was accidentally injured, and its coronet, hoof wall, and sole were detached in a mass. The bleeding was observed from the exposed dermis of the affected foot for several hours, with second scoreof supporting lameness. The bleeding from the digital artery at the exposed dermis was stopped by epinephrine spray and direct pressing. Nitrofurazone ointment was topocally applied for 5 days and sxytetracycline solusion was delivered by a fluid jet in the affected hoof one month. The calf was born 5 months after the avulsion. The horn of the hoof wall was grown from the coronet to sole, and was gradually filled from the heel to the toe. The growth rate and filling rate of the hoof wall were 0.32mm and 0.54mm a day, respectively.

Hematological and serum biochemical values were assessed from 20 clinically healthy Japanese Macaques raised in Everland Zoological Gardens and compared to the International Species Information System (ISIS) reference range that is used internationally as standard for wildlife animals. Taking our standard on sexual maturation at age 4, tRBC values in Macaques under age 4 were significantly lower than those over age 4, but the Hb and PCV values were significantly higher. Compared to the ISIS standard, the tRBC values in Macaques under age 4 were significantly lower whereas the Hb and MCHC values were significantly higher. Moreover, in the samples of Macaques over age 4, the PCV and MCV values were significantly lower than the ISIS standard. On serum biochemistry values the creatinine and amylase values in the Macaques under age 4 were significantly lower than those over age 4. In comparison with the ISIS standard, the values of ALT, ALP, BUN, IP, Ca2+ and K+ in the Macaques under age 4 did have no significant difference. The values of TP, GGT, tBil, amylase, TG and UA were significantly higher than the ISIS standard, but the values of albumin, AST, glucose, creatinine, cholesterol, CPK, LDH, Na+ and Clwere significantly lower. In contrast, the values of TP, albumin, ALT, ALP, creatinine, cholesterol, amylase, TG, IP and Na+ in the Macaques over age 4 did have no significant difference, but the values of GGT, BUN, tBil, UA and Ca2+ were significantly higher, while the values of AST, glucose, CPK, LDH, K+ and Cl- were significantly lower. On the other hand, there was no significant difference in hematological and serum biochemical values between the groups of male and female.
E. coli에서 GroEL/ES chaperone 공발현에 의한 활성형 cyclodextrin glucanotransferase의 생산 증대

Chaperone 분자는 세포 내에서 새로 합성된 polypeptides의 misfolding을 보호하는 역할을 가진다. 이런 chaperone 분자와의 공발현은 활성형 재조합 단백질의 생산을 증가를 기대할 수 있다. 본 연구에서는 E. cozi에서 B. macerans 유래 cyclodextrin glucanotransferase (CGTase)의 활성형 생산에 GroEL/ES chaperone과의 공발현의 효과에 대해 조사하였다. cgt와 groEL/ES 유전자출 발현하는 pTCGT1과 pGro7은 각각 T7 promoter와 araB promoter에 의해 조절되고 이들을 E. coli cell에 co-transformation시켰다. 재조합 E. coli에서 IPTG와 L-arabinose의 최적 농도를 결정하기 위해 행한 결과 1 mM IPTG, 0.3 mg L-arabinose/$m\ell$에서 가장 높은 CGTase 활성을 나타내었다. 그리고 tube에서는 L-arabinose와 IPTG를 각각 0.4~0.5 $OD_{600}$과 0.8~l.0 $OD_{600}$에서 첨가하였을 때 활성형 CGTase의 생산이 증가되었다. GroEL/ES 공발현 조건에서는 가용성 CGTase 활성이 0.7~0.73 unit/$m\ell$로 단독 발현의 0.36~0.56 unit/$m\ell$에 비해 약 1.5 배 정도 증가함을 알 수 있었다. SDS-PAGE 분석에서는 GroEL/ES 공발현 조건에서 총 CGTase의 33.6%정도가 가용성 형태로 생산됨을 알 수 있었다. Molecular chaperones prevent the misfolding of newly synthesized polypeptides in the cell. The coexpression of molecular chaperones could be expected to improve the production of soluble and active recombinant proteins. In this study, the effect of coexpression of E. coli GroEL/ES chaperone on the active production of Bacillus macerans cyclodextrin glucanotransferase (CGTase) in E. coli was investigated. Two plasmids, pTCGT1 and pGro7 in which the cgt and the groEL/ES genes are under the control of 77 promoter and araB promoter, respectively, were co-transformed into E. coli. With a series of cultures of recombinant E. coli cells, the optimal concentrations of IPTG and L-arabinose were found be 1 mM and 0.3 mg/$m\ell$, respectively. When IPTG and L-arabinose were added at 0.8~1.0 $OD_{600}$ and 0.4~0.5 $OD_{600}$, active CGTase production was increased significantly. This coexpression condition resulted in 1.5-fold increased level of soluble CGTase (0.7~0.73 unit/$m\ell$), compared to the level of CGTase in the single expression (0.36~0.56 unit/$m\ell$). An SDS-PACE analysis revealed that about 33.6% of CGTase in the total CGTase protein was found in the soluble fraction by coexpression of GroEL/ES chaperone.
Cyclodextrin Glucanotransferase와 Cyclodextrinase를 생산하는 Bacillus 속 세균의 분리와 그 효소들의 특성

토양으로부터 CGTase와 CDase를 함께 분비·생산하는 내열성 세균을 분리하였으며, 동정결과 Bacillus stearothermophilus로 판명되어 KJ16으로 명명하였다. Bacillus stearothermophilus KJ16 균주는 회분배양할 경우 2단계 성장을 하며 일차증식기에 CGTase를 생산하고 이차증식기에 CDase를 생산하는 양상을 보였다. 배치 pH를 7.0으로 조절하면서 배양할 경우, CGTase는 균의 증식에 따라 증가하고 CDase는 CGTase보다 뒤에 생산되었다. 그 생산양은 pH 비조절 배양에 비해 CGTase는 최대 12배(1300 mU/ml)까지, CDase는 2배(225 mU/ml)정도 증가되었다. 분리균주가 생산하는 CGTase는 분자량이 65,000 dalton으로 pH 6.0, 60℃에서 최적활성을 보이며 HPLC 분석을 통해 α-, β-, γ-CD를 42: 46: 12의 비율로 생산하는 α/β 혼합형 CGTase였다. 또한 CDase는 분자량이 68,000 dalton으로 pH 6.0, 55℃에서 최적활성을 보이며 최적기질은 γ-CD였다. When the medium pH was controlled at 7.0, the maximun enzyme activities of CGTase and CDase were increased by 12-fold(1300 mU/ml) and 2-fold (225 mU/ml), respectively, compared with the pH-uncontrolled batch culture. The CGTase of the isolate converted soluble starch to CDs with the ratio of α-CD: β-CD: γ-CD=42:46:12 at 55℃. The optimal pH and temperature of CGTase were 6.0 and 60℃, respectively and the optimal pH and temperature of CDase were 6.0 and 55℃. The molecular weights of the purified CGTase and CDase were estimated to be 65,000 and 68,000 dalton, respectively. Among serveral substrates, γ-CD was most rapidly hydrolyzed by the purified CDase.
인터넷 포탈의 성공전략 분석: 인수합병 사례를 중심으로

인터넷을 활용한 비즈니스의영역이 확대됨에 따라 인터넷 포탈이 주목 받고 있다. 점차 인터넷 포탈이 종합화, 대형화 되어가면서 그 역할 또한 커지고 있다. 이제 고객의 다양한 욕구와 변화된 경영환경 속에서 인터넷 포탈의 성공전략에 대한 논의가 필요한 시점이 되었다. 따라서 본 논문에서는, 인수합병을 통한 인터넷 포탈의 성공요인들을 제시하고자한다. 이를 위하여, 본 연구에서는, 국내외 주요 인터넷 포탈 기업들의 사례를 정성적 연구방법론을 바탕으로 분석하였다. 연구변수로는 인터넷 포탈의 기본적인 성공요인 2가지와 지속적인 성장을 위한 인수합병의 성공요인 3가지를 선정된 사례에 각각 적용하여 분석을 실시하였다. 분석결과, 독점적 컨텐츠를 확보하는 것과 임계점 이상의 가입자 규모를 확보하는 것이 인터넷 포탈의 성공을 위해 동시에 만족되어야 할 가장 중요한 성공요인들임을 확인하였다. 또한, 인수합병의 성공요인 분석을 통해 효율적인 인수합병 방안을 제시하고 있다.
