        Biologically Active and C-Amidated HinnavinII-38-Asn Produced from a Trx Fusion Construct in Escherichia coli

        The cabbage butterfly (Artogeia rapae) antimicrobial peptide hinnavinII as a member of cecropin family is synthesized as 37 residues in size with an amidated lysine at C-terminus and shows the humoral immune response to a bacterial invasion. In this work, a synthetic gene for hinnavinII-38-Asn (HIN) with an additional amino acid asparagine residue containing amide group at C-terminus was cloned into pET-32a(+) vector to allow expression of HIN as a Trx fusion protein in Escherichia coli strain BL21 (DE3) pLysS. The resulting expression level of the fusion protein Trx-HIN could reach 15~20% of the total cell proteins and more than 70% of the target proteins were in soluble form. The fusion protein could be purified successfully by HiTrap Chelating HP column and a high yield of 15 mg purified fusion protein was obtained from 80 ml E. coli culture. Recombinant HIN was readily obtained by enterokinase cleavage of the fusion protein followed by FPLC chromatography, and 3.18 mg pure active recombinant HIN was obtained from 80 ml culture. The molecular mass of recombinant HIN determined by MALDI-TOF mass spectrometer is 4252.084 Da which matches the theoretical mass (4252.0 Da) of HIN. Comparing the antimicrobial activities of the recombinant hinnavinII with C-amidated terminus to that without an amidated C-terminus, we found that the amide of asparagine at C-terminus of hinnavinII improved its potency on certain microorganism such as E. coli, Enterobacter cloacae, Bacillus megaterium, and Staphylococcus aureus.

        In this paper, steady laminar flow in a tank of double cylinder with rotating outer cylinder and upper disc was analyzed numerically. For simplicity, the stream function and vorticity have been introduced into the Navier-Stokes and the continuity equations. The transformed governing equations were converted into the fimite difference equations which were solved numerically by ADI (Alternating direction implicit) and SUR (Successive under relaxation) method.

        The propagations of crack in rotary bending fatigue test were investigated. The results can be summurized as follows. (1+ / _0)=(1+ _f/ _0)^(Exp[K(N*-1)]) It contains initial crack(or notch) length, final crack length which can be expressed in terms of stress, and experimentally determined constant. The final crack lengthes of smooth or notched specimen are equal near the fatigue limit of the smooth specimen and show a certain linear relationship with the stresses.

      • 國産 鋼板의 成形性에 對한 硏究

        To investigate the Plastic-Formability of domestic steel sheets, the sheets were cold-rolled to final thickness and were annealed at various temperatures for various soaking times in a furnace filled with inert-gas. The properties relative to plasticformability, n--values, r--values, CCV and Erichsen values, along with the common mechanical properties are carefully examined for each annealing condition. The optimal conditiconfor plastic-formability are investigated and determined as 600℃-700℃ annealing temperature with 30 min soaking time. Data of all the test results are contained in Appendix for reference.

      • Coil Spring의 振動傳達 特性에 관한 考察

        This study attempts an analytical investigation of coil springs with significant mass under vibratory motion. Equations are presented for both displacement of the free end and transmitted force on support of spring. Equations were solved by applying standard Fourier's techniques and several features of the results are pointed out. It is shown that the common spring may cause an uncommon problems unless more attention is paid to the dynamic characteristics of spring.

      • JPEG2000 코덱을 위한 최적의 데이터 형식 모델링

        최근의 통신매체로는 함축적이고 인식이 용이한 영상/비디오가 매우 선호되고 있다. 그러나 이들 데이터는 그 양이 과다해 영상/비디오 데이터의 압축의 큰 연구과제가 되어 왔다. 본 논문에서는 영상압축 시 성능을 크게 좌우할 수 있는 데이터 형식의 최적화에 관련되고, 웨이블릿 변환과 이를 이용한 JPEG2000을 기반으로 한다. 본 논문에서는 이산 웨이블릿 변환 시 이용되는 데이터 형식을 결정하는데 있어서의 기준을 주파수 변환 및 양자화 과정에서의 데이터 손실 양으로 설정하였으며, 이를 바탕으로 실험적으로 최적의 데이터 형식을 도출하였다. 그 결과, 필터계수는 (1, 9)의 10-비트 고정소수점 형식, 웨이블릿 계수는 (9, 7)의 16-비트 고정소수점 형식이 최적임을 확인하였다. Recently, images/videos have been preferred as the communication media because of their information-implication and easy recognizability. But the amount of their data is so large that it has been the major research area to compress their data. This paper is related to optimization in the image data format which can make a great effect in performance of data compression and is based on the wavelet transform and JPEG2000. This paper established a criterion to decide the data format to he used in wavelet transform which is on the bases of the data errors in frequency transform and quantization. This criterion has been used to extract the optimal data format experimentally. The result were (1, 9) of 10-bit fixed-point format for filter coefficients and (9, 7) of 16-bit fixed-point data format for wavelet coefficients and their optimality was confirmed.

        The selection of maching conditions based on minimum production cost and maximum production rate is analyzed for a multi-stage machining model which consists of rough cuttings, finishing cutting and honing. The problems are formulated as standard mathematical programming problems, which subject to several constraints such as cutting velocity, feed rate, required roughness etc., and mixed penalty method is used to solve the problems. The procedure used in this study can be applied to many actural multi-stage machinings.

      • 나노 구조에서 실리콘 산화 절연막의 스트레스 유기 누설전류

        In this paper, the stress induced leakage currents of thin silicon oxides is investigated in the ULSI implementation with nano structure transistors. The stress and transient currents associated with the on and off time of applied voltage were used to measure the distribution of high voltage stress induced traps in thin silicon oxide films. The stress and transient currents were due to the charging and discharging of traps generated by high stress voltage in the silicon oxides. The transient current was caused by the tunnel charging and discharging of the stress generated traps nearby two interfaces. The stress induced leakage current will affect data retention in electrically erasable programmable read only memories. The oxide current for the thickness dependence of stress current, transient current, and stress induced leakage currents has been measured in oxides with thicknesses between 113.4${\AA}$ and 814${\AA}$, which have the gate area $10^3cm^2$. The stress induced leakage currents will affect data retention and the stress current, transient current is used to estimate to fundamental limitations on oxide thicknesses. 본 논문에서 얇은 실리콘 산화막의 스트레스 유기 누설전류는 나노 구조를 갖는 트랜지스터의 ULSI 실현을 위하여 조사하였다. 인가전압의 온 오프 시간에 따른 스트레스전류와 전이전류는 실리콘 산화막에 고전압 스트레스 유기 트랩분포를 측정하기 위하여 사용하였다. 스트레스전류와 전이전류는 고스트레스 전압에 의해 발생된 트랩의 충방전과 양계면 가까이에 발생된 트랩의 터널링에 기인한다. 스트레스 유기 누설전류는 전기적으로 기록 및 소거를 실행하는 메모리 소자에서 데이터 유지 능력에 영향이 있음을 알았다. 스트레스전류, 전이전류 그리고 스트레스 유기 누설전류의 두께 의존성에 따른 산화막 전류는 게이트 면적이 10/sup -3/㎠인 113.4Å에서 814Å까지의 산화막 두께를 갖는 소자에서 측정하였다. 스트레스 유기 누설전류, 스트레스전류, 그리고 전이전류는 데이터 유지를 위한 산화막 두께의 한계에 대해 연구 조사하였다.

        Industrial aluminium sheets cold-rolled to a final thickness of 0.8mm are annealed in two ways. In the first, the specimens are annealed for one hour at different annealing temperatures: in the second, specimens are annealed at various temperatures for different annealing intervals. The common mechanical properties, n-values and r-values are carefully examined in each way. The r--values show high peak during sheets are partially recaystallized. The n-values show maximum value at full recrystallization and remain constant after it. And also the effects of impurities, such as Cu, Mg, Zn, on the mechanical properties, n-values and r-values of pure aluminium are investigated.

      • 超音速 노즐흐름에 있어서 凝縮이 傾斜衝擊波에 미치는 影響

        본 연구에서는 작동유체로서 수증기와 거동이 유사한 습공기를 대기 흡입식 풍동을 사용하여 원호 노즐로서 팽창시키는 경우에 대하여 응축충격파가 발생하는 흐름이 측정부내에 쐐기를 설치하여 발생시킨 경사충격파에 미치는 영향에 대해 실험적으로 조사하였다. Last several stages of high capacity fossil power steam turbine and most stages of nuclear power steam turbine operate on wet steam. As a consequence, the flows in those cascades are accompanied by condensation, and the latent heat caused by condensation affects an oblique shock wave being generated at the vicinity of trailing of the blade. In the case of expanding of moist air through a suction type indraft wind tunnel, the effect of condensation affection the oblique shock wave generated by placing the small wedge into the supersonic part of the nozzle was investigated experimentally. In these connections, the relationship between condensation zone and reflection point of the incident oblique shock wave, angle between wedge bottom wall and oblique shock wave, and the variations of angles of incident and reflected shock waves due to the variation of initial stagnation relative humidity are discussed. Furthermore, the relationship between initial stagnation relative humidity and load working on the nozzle wall, obtained by measuring static pressure at the nozzle centerline, is discussed.
