      • 인간의 meningioma에서 nm23-H1의 발현과 세포이동능은 역비례한다

        The ㎚23 gene is widely documented as a suppressor of the metastatic phenotype in many human cancers. However, its role with respect to the invasive behavior of primary central nervous system tumors has scarcely been addressed. The present study was aimed to examine the expression of the ㎚23-Hl gene product in 14 meningiomas in order to clarify the association of its expression with the histological types or migration rate, The ㎚23-Hl protein levels in human meningiomas were analyzed using Western blotting analyses. The ㎚23-Hl protein levels and cell migration rate was not correlated with different histologic types of meningiomas. However, the expression of ㎚23-Hl is inversely correlated with the migration rate in meningiomas. These results suggest that ㎚23-Hl may play an important role in the cell migration of meningiomas in female patients.

        nm23-H1 유전자가 주입된 U87MG 세포의 이동능과 침윤능의 감소

        nm23-H1 gene expression has been inversely correlated with tumor metastatic potential in certain tumors including melanomas, breast carcinomas, and hepatocellular carcinomas. However, its role with respect to the invasive behavior of central nervous system tumors has scarcely been addressed. Because cell motility and invasion plays an essential role in metastatic dissemination, we have studied whether motile human glioma cell(U87MG) transfected with nm23-H1 complementary DNA have any alterations in their ability to migrate and invade. There was no significant changes in the shape and size of the cells following nm23-H1 transfection. The role of nm23-H1 in glioma migration and invasion have been evaluated by in vitro simple scratch technique and brain slice invasion model. Basal migration ability of nm23-H1 transfectants cell(U87MG-pEGFP-nm23) were lesser than U87MG. Accordingly, U87MG-pEGFP-nm23 didn't migrate away apparently from the tumors implanted site comparing U87MG in brain slice invasion model. These results suggest that nm23-H1 may play an important role in suppressing the human glioma migration and invasion.

        Multiplication and Transformation of Medicinal Plants for Production of Useful Secondary Metabolites 2. Establishment of Hairy Root Cultures of Centella asiatica

        The hairy root cultures of Centella asiatica were established by infection leaf explants with Agrobacterium rhizogenes A4, 15834 in 1/2 Murashige and skoog liquid medium supplemented with 50 μM acetosyringone. The induced hairy roots were subjected to paper electrophoresis for the detection of opine and opine-positive clones which were considered to have been transformed. Five hairy root clones were selected according to the different bacterial strains used, growth rate and pattern. Among media tested, MS basal medium substituted phosphate concentration by 2.5 mM K_2HPO_4 showed the highest growth rate in the dark condition.

        Increased Expression of Nitric Oxide Synthase Coincides with Reversal of Renovascular Hypertension

        <P> The present study was aimed at investigating whether there are changes in the expression of nitric oxide synthase (NOS) in relation with the unclipping-induced fall of blood pressure in two-kidney, one clip (2K1C) hypertension. Male Sprague-Dawley rats were made 2K1C hypertensive by clipping the left renal artery for four weeks. Sham-clipped rats served as control. The expression of endothelial constitutive (ec) NOS proteins and tissue levels of NO metabolites were determined in the kidney. Systolic blood pressure was significantly increased in clipped rats compared with that in the control. The development of hypertension was associated with decreases in the expression of ecNOS proteins and tissue levels of NO metabolites in the clipped kidney. The blood pressure at twenty-four hours after removal of the renal arterial clip fell to the control level. Accordingly, in the unclipped kidney, the expression of ecNOS proteins and tissue contents of NO metabolites were increased to the control level. The contralateral kidney was not affected by the development or reversal of hypertension. It is suggested that an enhanced expression of ecNOS in the unclipped kidney is an important component in the reversal of renovascular hypertension.

        테라테인먼트적 코어운동프로그램과 간섭파 전기자극이 복부비만 여성에 미치는 영향

        본 연구는 20대 비만여성의 복부비만에 코어운동 프로그램과 간섭파 전기자극을 적용하여 그 효과를 알아보고자 하였다. 복부비만 여성 30명을 무작위로 선정하여 코어운동프로그램군과 간섭파 전기자극군, 그리고 대조군으로 나눴다. 코어운동프로그램군과 간섭파 전기자극군의 테라테인먼트중재는 각각 1회 30분, 주 3회, 6주간을 실시하였고 연구대상자들의 체질량지수, 복부둘레, 피하지방을 중재 전, 후에 측정하였다. 대응표본 T검정의 결과에서 코어운동프로그램군과 간섭파 전기자극군에서 체질량지수와 복부둘레에서 유의한 차이가 나타났고 일원배치분산분석후 사후검정에서도 코어운동프로그램군과 간섭파 전기자극군에서 유의한 차이가 나타났지만 피하지방에서는 세군 모두 유의한 차이가 나타나지 않았다. 그러므로 향후 연구에서는 비만을 가진 다양한 그룹을 대상으로 테라테인먼트적 전기자극을 제공하여 객관적인 평가를 통한 검증을 하고 더 나아가 가정용 전기자극 건강보조기구 개발이 필요할 것으로 보인다. This research investigates the effects of theratainment core exercises and theratainment interferential current on abdominal obesity for women in their twenties. A total of thirty female participants with abdominal obesity were randomly divided into three groups: Group A participated in a theratainment core exercise program, Group B received theratainment interference electrical stimultation, and Group C was designed as the control group. The theratainmnet intervention for the Group A and B were conducted for six weeks, thirty minutes per day, three days per week. BMI, waistline, and skin-fold thickness data were taken as pretest and posttest measures. The results of the paired-t test showed that there were significant differences in both BMI and waistline for participants involved in the theratainment core exercise program as well as thertainment interferential current(p<.05). In addition, there were significant differences in the core exercise program group and the interferential current group in post-hoc test from one-way ANOVA. There were significant differences in BMI and waistline measures within both experiment groups. However, no difference was observed in the skin-fold thickness measurement. For future studies, it is suggested that researchers should control participants’ eating habits, apply electrical stimulation to many abdominal muscles, and employ objective measurements.

        Carrageenan으로 유도된 염증성 근통증 흰쥐 모델에서 경피신경전기자극과 냉치료에 의한 척수 cyclooxygenase-2의 감소

        근육통에 의해 유발된 이차성 통각과민과 척수의 허리팽대부에서의 cyclooxygenase-2 (COX-2)의 발현에 경피신경전기 자극(transcutaneous electrical nerve stimulation, TENS)과 냉치료에 대한 효과를 알아보기 위하여 연구되었다. 흰쥐의 장딴지근에 2% carrageenan을 근육주사하여 근통증 실험모델을 만들었다. 이차성 통각과민을 측정하기 위하여 열에 대한 발움추림잠복기 (paw withdrawal latency, PWL)와 꼬리치기검사 (tail flick latency, TFL)를 조사하였다. 척수의 허리팽대부의 COX-2 발현을 역전사중합연쇄반응 (reverse transcription-polymerase chain reaction, RT-PCR)과 면역조직화학염색법에 의해 조사하였다. 열에 대한 이차성 통각과민은 대조군에 비교하여 경피신경전기자극과 냉치료를 적용한 그룹에서 유의적으로 감소하였다. 척수의 허리팽대부에서 COX-2 mRNA 수준은 대조군에 비교하여 경피신경전기자극과 냉치료를 적용한 그룹에서 감소하였으며 또한, COX-2 면역반응성도 척수의 뒤뿔에서 대조군에 비교하여 경피신경전기자극과 냉치료를 적용한 그룹에서 감소하였다. 이러한 결과들은 경피신경전기자극과 냉치료를 적용한 그룹에서 COX-2 발현의 감소는 근육통에 의해 유발된 이차성 통각과민을 감소시키는데 중요한 요인으로 작용할 가능성이 있음을 시사한다. The purpose of this study was to investigate the effects of transcutaneous electrical nerve stimulation (TENS) and cold therapy on cyclooxygenase-2 (COX-2) expression in lumbar spinal cord and on secondary hyperalgesia produced by muscle pain. Muscle pain was experimentally induced by injection of 2% carrageenan into gastrocnemius muscle of Sprague-Dawley rats. For assessment of secondary hyperalgesia, the rats were tested for paw withdrawal latency (PWL) and tail flick latency (TFL) to heat stimulus. COX-2 expression in lumbar cord was examined using RT-PCR and immunohistochemistry. Secondary hyperalgesia to heat stimulus was significantly reduced in the both TENS- and cold-treatment groups, compared to the control group. The COX-2 mRNA levels were down-regulated in the lumbar spinal cord in the both TENS- and cold-treatment groups. In addition, COX-2 immunoreactivity was decreased in the dorsal horn of the lumbar spinal cord in the both TENS- and cold-treatment groups. These results suggested that decreased COX-2 expression in the lumbar spinal cord of the subjects receiving TENS treatment and cold therapy might be an important factor for reducing secondary hyperalgesia produced by muscle pain.

        2일 간격의 교대근무가 요중 17-OHCS와 Free Cortisol 농도에 미치는 효과

        In this study, 20 students in C university were selected by control group and 14 shift work nurses working in C university hospital by test group. It took part in case of day shift(8AM-4PM), evening shift(4PM-12MN), and night shift (12MN-8AM) to know a urinary 17-OHCS and free cortisol changes which works in a rapidly rotating shift work system. The working team were an rest period of a day and an observation of a week. It obtained an urine specimen before and after work shift in 2nd day. Test group and control group of shift work compared to change of levels of urinary 17-OHCS and free cortisol of urine collected from nurses of day shift, evening shift, and night shift. The data was analyzed by t-test, paired t-test. The results are as follows. 1. Compare 8AM with 4PM in day shift. Control group increased 4.1mg at 8AM, 4.2mg at 4PM in a case of 17-OHCS(p=.84) and also test group increased a little 3.5mg at start time of work, 3.6mg at stop time(p=.97). In a case of free cortisol control group decreased 3.8㎍ at 8AM, 2.4㎍ at 4PM(p=.12) and test group decresed 8.3㎍ at start time of work, 3.2㎍ at stop time(p=.22). 2. Compare 4PM with 12MN in evening shift. Control group decreased 4.2mg at 4PM, 2.9mg at 12MN in a case of 17-OHCS(p=.54), but test group increased 1.7mg at start time of work, 3.4mg at stop time(p=.07). In a case of free cortisol control group decreased 2.4㎍ at 4PM, 1.9㎍ at 12MN(p=.23) and also test group decresed a little 2.6㎍ at start time of work, 1㎍ to stop time(p=.43). 3. Compare 12MN with 8AM in night shift. Control group increased 3.9mg at 12MN, 4.1mg at 8AM in a case of 17-OHCS(p=.79) and also test group increased 6.3mg at start time of work, 8.4mg at stop time(p=.16). In a case of free cortisol control group increased 1.9㎍ at 12MN, 3.8㎍ at 8AM(p=.08) and test group increased 4.4㎍ at start time of work, 11.6㎍ at stop time(p=.04). As a result of this study reveals that health level of nurses is decreased in a rotating shift work(2days interval). As mentioned above, it was confirmed that the rotating shift work had influence on the health of the nurses. For it, I present the objective base data to mesure the level healthy in order to extent the understanding of the physical aspects of the nurses. Moreover, it is considered that one can make use of it as an objective base with a view to the rational management for the nursing administration.

      • Carrageenan으로 유도된 염증성 근통증 흰쥐 모델에서 냉치료에 의한 척수의 c-fos의 발현

        Expression of c-fos, an immediate early gene, has accepted to be a marker of functional activity in neurons. This study was aimed to investigate the effects of cold therapy on the expression of spinal cord c-fos in rats of carrageenan-induced muscle pain. Muscle pain was induced in male Sprague-Dawley rats by intra-muscular injection of gastrocnemius with $2\%$ carrageenan. The paw withdrawal latency (PWL) and tail flick test (TFT) responses to heat stimuli were used to detect secondary hyperalgesia produced by the muscle pain and measured to assess the effects of cold. The expression of c-fos was determined in the lumbar regions of the spinal cord by reverse transcription-polymerase chain reaction (RT-PCR) and immunohistochemistry assays. The secondary hyperalgesia to heat simuli (PWL and TFT) were significantly reduced in cold therapy compared with that in the controls. In RT-PCR assays the expression of c-fos mRNA was down-regulated in the lumbar spinal cord in cold group. In addition, Fos immunoreactivity in the dorsal horn of the lumbar spinal cord was decreased in cold group. These results suggested that application of cold attributed to increase PWL and TFT responses and to decrease expression of the c-fos produced by muscle pain.
