<P><B>Abstract</B></P> <P>Physostigmine, an acetylcholinesterase inhibitor, is known to affect the brain function in various aspects. This study was conducted to test whether physostigmine affects cell proliferation in the telencephalon of zebrafish. BrdU-labeled cells was prominently observed in the ventral zone of the ventral telencephalon of zebrafish. The increased number of BrdU- and proliferating cell nuclear antigen-labeled cells were shown in zebrafish treated with 200μM physostigmine, which was inhibited by pretreatment with 200μM scopolamine. iNOS mRNA expression was increased in the brain of zebrafish treated with 200μM physostigmine. Consistently, aminoguanidine, an iNOS inhibitor, attenuated the increase in the number of BrdU-labeled cells by physostigmine treatment. Zebrafish also showed seizure-like locomotor activity characterized by a rapid and abrupt movement during a 30min treatment with 200μM physostigmine. Neural activity in response to an electrical stimulus was increased in the isolated telencephalon of zebrafish continuously perfused with 200μM physostigmine. None of the number of BrdU-labeled cells, neural activity, or locomotor activity was affected by treatment with 20μM physostigmine. These results suggest that 200μM physostigmine increased neural activity and induced cell proliferation via nitric oxide production in zebrafish.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Physostigmine increased the cell proliferation and neural activity in the zebrafish brain. </LI> <LI> Scopolamine suppressed the increased cell proliferation by physostigmine. </LI> <LI> The increased cell proliferation by physostigmine was accompanied with the increased expression level of iNOS mRNA. </LI> <LI> The increased cell proliferation by physostigmine was suppressed by an iNOS inhibitor. </LI> </UL> </P>
Ginsenoside Rg1 Reduced Spontaneous Epileptiform Discharges and Behavioral Seizure in the Zebrafish

Epileptifrom discharges were induced in the telencephalon of the adult zebrafish via perfusion with pentylenetetrazole (PTZ), bicuculline methiodide, kainic acid-treated artificial cerebrospinal fluid (aCSF), and Mg<SUP>2+</SUP>-free aCSF. Ginseng total saponin [GTS (50 ㎍/ml)] was shown to attenuate the occurrence rate of epilpetiform discharges by 50-75%, compared to the control. Ginsenoside Rg₁ (130 ㎛) reduced the epileptiform discharges in the isolated telenephalon and delayed the occurrence of behavioral seizures observed from the adult zebrafish placed in the PTZ (10 mM)-containing aquarium water. However, Re was not effective in the suppression of epileptiform discharges and behavioral seizures. These results indicate that Rg1 may be useful in the control of epileptiform discharges and effective in controlling behavioral seizures, and that the zebrafish can be used as a model animal for the testing of potential anticonvulsant drugs.
증례 : 호흡기 ; 폐암과 전립선 암으로 오인된 면역 정상 환자의 파종성 효모균증 1예

면역 정상인 환자에서 파종성 효모균증은 드물지만 지속적으로 보고되고 있으며, 임상양상이 다양하여 종양 또는 결핵 등의 다른 감염성 질환과 감별해야 한다. 본 증례는 왼쪽에만 국한된 흉수를 주소로 내원한 환자에서 열이나 다른 염증인자의 증가가 없으며, 흉부단층촬영에서 폐결절 및 흉막비후, 다발성 결절, 림프절 비대 등의 소견을 보여 일차적으로 폐암을 의심하여 검사를 진행하였다. 양전자방출단층촬영에서 전립선의 대사항진이 추가로 발견되어 전립선암을 배제하기 위해 전립선 조직검사와 종격동 림프절의 조직검사를 통해 파종성 효모균증으로 진단한 사례이다. 치료로 amphotericin B는 전립선 투과력이 fluconazole에 비하여 상대적으로 낮아 fluconazole을 투약하였고 호전 중인 상태이다. 면역 정상인 환자에서 뇌수막염을 동반하지 않은 파종성 효모균증에서 폐와 전립선을 동시에 침범한 경우는 보고된 적이 없으며 fluconazole 치료로 호전 중인 증례를 경험하여 문헌고찰과 함께 보고하는 바이다. Cryptococcosis can occur in immunocompetent patients; in fact, it accounts for 20% of all cryptococcal infections. In immunocompetent patients, although clinical symptoms may not be obvious, cryptococcosis should be considered in the differential diagnosis for cancer and other diseases. We recently encountered a case of disseminated cryptococcosis involving the lung and prostate. A 71-year-old male patient was referred for evaluation of a large pleural effusion. He was a 40 pack years ex-smoker, and showed no evidence of immunodeficiency. Chest computed tomography and positron emission tomography images suggested primary lung cancer with pleural metastases and prostate cancer. Tuberculous pleurisy was suspected due to a lymphocyte dominant exudative pleural effusion and high level of adenosine deaminase. Prostate and bronchoscopic biopsies demonstrated chronic granulomatous inflammation, and periodic acid Schiff’s and Grocott’s methenamine silver staining confirmed cryptococcosis in both specimens. Antifungal therapy with fluconazole was started, and follow-up images showed complete resolution of the pleural effusion. (Korean J Med 2015;89:91-96)

Objectives: The detection methods of cell-free tumor DNA (cfDNA) continue to evolve in non-small cell lung cancer (NSCLC). Peptide nucleic acid-based methods have been developed to analyze EGFR mutation in plasma cfDNA in patients with NSCLC. Methods: From January, 2014 to December, 2016, we obtained tissue and plasma of 60 patients before treatment with EGFR-tyrosine kinase inhibitor (TKI). Quantitative analysis was also conducted on 11 patients who could obtain plasma samples during EGFR-TKI treatment. cfDNA testing was performed using PANAMutyper and a semi-quantitative index (SQI). Results: Concordance rate matched tissue and plasma results were 41.7% (35% for E19del, 50% for L858R). In 10 patients, the test failed because the amount of DNA was small. G719S mutation was detected in the plasma of one patient with E19del mutation in tissue biopsy. In 11 patients, concordance rates higher in SQI (PANAMutyper 27.3%, SQI 54.5%). When the copy numbers of EGFR mutation were less than 100 by SQI test, it was not detected by PANAMutyper. Five patients (45.5%) showed dynamic change of mutation status before disease progression (PD). (T790M mutation=1, negative conversion=3, decrease in copy numbers<100 =1) T790M mutation status was 100% consistent with tissue and plasma results after PD. Conclusions: The PANAMutyper test could not detect EGFR mutations at less than 100 copies. The sensitivity can be increased by applying the quantitative test. Dynamic changes in the cfDNA EGFR mutation status can help predict clinical outcome.
Isomer specificity of conjugated linoleic acid (CLA): 9E,11E-CLA

Conjugated linoleic acids (CLA) were identified in 1980’s, since then it has been intensively studied due to its various beneficial health effects such as anti-inflammatory, anti-atherogenic, anti-carcinogenic and anti-diabetic/obesity effects. Isomer specificity of a number of CLA isomers, especially predominant isomer 9Z,11E- and 10E,12Z-CLA, is now recognized. However, the less prevalent CLA isomers have not been well characterized. Recently, studies have reported the distinctively different effects of 9E,11E-CLA in colon cancer cells, endothelial cells, and macrophage cells compared to the rest of CLA isomers. In this review, various effects of CLAs, especially anti-inflammatory and anti-atherogenic effects, will be discussed with focusing on the isomer-specific effects and potential mechanism of action of CLA. At last, recent studies about 9E,11E-CLA in in vitro and animal models will be discussed.

Vinous-throated Parrotbills and Daurian Redstarts are the major host species of Common Cuckoos in Korea. Here, we report egg color polymorphism observed in Common Cuckoos and these specific hosts. Two morphs of Common Cuckoo eggs (immaculate blue and pale blue) were found in the nests of Vinous-throated Parrotbills, and a single morph (pale blue with firebrick squiggles) was identified in the nests of Daurian Redstarts. Moreover, three egg morphs (immaculate blue, pale blue, and white) were found in the nests of Vinous-throated Parrotbills, and two morphs (pale blue with firebrick spots and pinkish-white with firebrick spots) were observed in the nests of Daurian Redstarts.

BACKGROUND/OBJECTIVES: Gelidium amansii (GA) contains plenty of agars and various biological substances, which make them a popular functional food to control body weight in previous studies. Unlike previous studies focused on agar in GA, objectives of this study were to investigate the effects of agar-free GA extract (AfGAE) on preventive and treatment models by using diets-induced obese (DIO) C57BL/6J mice. MATERIALS/METHODS: AfGAE were used to test their effects on the prevention (Exp-1) and treatment (Exp-2) against obesity after pilot study in DIO mice. The weight changes of the body and fat tissues and protein expression related to lipid metabolism and inflammation as well as plasma lipid profile and insulin were detected. RESULTS: Although AfGAE did not prevent long-term DIO, it did increase the levels of anti-inflammatory cytokine production and lipolysis protein. We further evaluated various doses of AfGAE in preventive and treatment models. As a result, our findings suggested that an AfGAE administration as a preventive model might be a better approach to achieve its anti-inflammatory and lipolysis-promoting effects in DIO mice. CONCLUSION: Although future studies to investigate the target materials such as polyphenols in AfGAE are required, the result suggests that GA without agar might be a therapeutic tool to improve health conditions related to inflammation.
