        Misorientation Characteristics at the Growth Front of Abnormally-Growing Goss Grains in Fe–3%Si Steel

        Penetrating morphologies at the growth front of abnormally-growing Goss grains in Fe–3%Si steel was examined in view ofthe possibility that they are the two-dimensional section of the solid-state wetting (SSW) along the triple junction line. Toobtain the statistically meaningful data, 102 penetrated grain and 204 penetrating abnormally-growing Goss grain morphologieswere examined. The misorientation angles of penetrating and penetrated grains were measured by electron backscattereddiffraction. Among the 102 examined penetrated grain boundaries, none has low misorientation angles less than 15°,whereas 17.2% of the 204 penetrating grain boundaries have low misorientation angles, and 23.5% of them have coincidencesite lattice boundaries. Besides, boundary energies of penetrating Goss grains, which were estimated from misorientationangles of the three grains in the penetrating morphology, satisfied the energetic condition for SSW along the triple junctionline. These results imply that the abnormal grain growth of Goss grains in Fe–3%Si steel occurs by the mechanism of subboundaryenhanced SSW.

        [γ-^32P]ATP was used to test phosphorylation of membrane proteins of mating type a cells of heterobasidiomycetous yeast Rhodosporidium toruloides separated by non-denaturing electrophoresis. The phosphoprotein was observed in the membrane proteins. The phosphorylation was inhibited by the pheromone rhodotorucine A (Rh. A) secreted by mating type A of the yeast. Rh. A didn't inhibit the phosphorylation in the presence of a trigger peptidase (TPase) inhibitor, antipain. Partially digested Rh. A by trypsin maintained the phosphorylation inhibitory activity. These results show that TPase activity plays an important role in the transduction of pheromone signal in the yeast.

      • 이담자 효모균의 성분화과정에서 막단백질 중 Ca^2+ -ATPase와 trigger peptidase(TPase)의 상호관계

        We have studied the relation between Ca^2+-ATPase and trigger peptidase(TPase) which are membrane protein well known as their significant role for signal transduction of mating pheromone in heterobasidiomycetous yeast, Rhodosporidium toruloides. We found out that there were Ca^2+-ATPase and TPase together in isolated calmodulin binding protein(CBP), using clamodulin affinity column chromatography after solubilization of mating type a cell membrane protein, and that the dependence of enzyme activity of both the enzymes of Ca^2+, phospholipid and nonionic detergent are similar. However, Ca^2+-ATPase had quite absolute dependence on calmodulin and, on the other hand, TPase didn't have any dependence. Judging from the fact that there are both enzymes in CBP which the dependence of calmodulin are quite different, we found out that both enzymes were made to their compound and existed in mating type a cell membrane.

      • 성인 급성 부고환염에서 Nested PCR에 의한 Chlamydia trachomatis의 검출

        연구목적 : 성교가능 성인남자의 급성 부고환염에서 Chlamydia trachomatis가 차지하는 빈도를 Nested PCR을 통해 검출하여 보고자 하였다. 대상 및 방법 : 1996년 9월부터 1999년 2월까지 본 충북대학교 병원 비뇨기과 외래 및 응급실을 내원하여 급성 부고환염으로 진단된 환자 중 성교가능 성인남자 28명을 대상으로 하여 환자로부터 얻은 뇨검체로 일반 미생물 배양검사, 그람염색검사 및 Chlamydia PCR을 시행하였다. 결과 : 급성 부고환염으로 진단된 28명의 평균 연령은 30.9±12.4세 였으며 총 28명중 17명 (60.7%)에서 Nested PCR 검사에 양성으로 나타났으며 연령별로는 20대가 10명중 7명(70%)으로 가장 높은 양성율을 보였다. 그람염색도말 결과 28명중에서 6명(21%)이 비임균성 요도염에 양성을 나타냈으며 또한 일반 미생물 배양 검사 결과 2명에서 균이 자랐는데, 각각 S. epididermidis와 E. coli였다. 결론 : PCR을 통한 Chlamydia trachomatis의 검출이 지금까지 행해진 다른 검사 방법들에 비해 월등하게 민감도가 높으며 성접촉이 가능한 연령의 성인 남자가 급성 부고환염 증세로 내원시 기본적으로 PCR을 통한 Chlamydia trachomatis 검출 검사를 시행하여 적절한 치료가 이루어지는 것이 중요할 것이다. Purpose : We studied the incidence of Chlamydial epididymitis in sexually active male with acute epididymitis by the Nested polymerase chain reaction( PCR ). Materials and Methods: Urine specimens, collected from 28 patients that was 30.9±12.4 years old and presented with acute epididymitis, were tested for the presence of Chlamydia trachomatis by the Nested PCR and for other bacteria by the urine gram stain and culture. Results: Out of total 28 patients, 17 patients(60.7% ) showed positive for Chlamydia trachomatis by the Nested PCR. In thirtieth decade, 7 of 10 patients(70%) showed positive, revealed the highest incidence. 6 patients(21%) showed positive for other bacteria by the urine culture. Conclusion: Detection rate of Chlamydia trachomatis by the Nested PCR was higher than other current diagnostic technique. For the high incidence of Chlamydial epididymitis, all men with acute epididymitis should undergo Chlamydia trachomatis evaluation for adequate treatement of this infection.

        양측 하악과두 골절에 대한 관혈적 정보술후 나타난 근약화와 시력장애 : 증례보고

        The condylar fracture show high occurrence rate in the following of symphysis and angle, because it is easy to have concentration of outer pressure due to its long and narrow anatomical shape and mouth opened state when trauma is attacked. For the clinical findings, there are trismus, opening deviation, mal-occlusion and open bite The classification of condylar fracture is varied due to the location of condylar bone fragment, fracture site. There are conservative and surgical methods for treatment modalities. Because of many advantages, conservative method is widely used it has no particular limits of treatment adjustment but the cases of absolutely surgical correction indication are easily found in clinical experience. Although many methods are suggested for treatments of condylar fracture, there are no particular surgical methods for precise reduction due to difficult accessibility in high condylar neck fracture. For precise reduction of condyle, we used Nam's method. However, in Nam's method, because condyle is fixed when once it is kept off externally reduced in avascular state, so complication such as condyle resorption is pointed out as a problem. In our clinic, we obtained some knowledge by treating a patient who had muscle weakness and vision disorder after temporary airway obstruction follownig the both condylar fracure operated using Nam's method, so here we reported the case with a study of documents.

        이담자균 효모 Rhodosporidium toruloides에서 Rhodotorucine A에 의한 막단백질 인산화의 저해와 Trigger Peptidase의 관련성

        이담자 효모균 Rhodosporidium toruloides 접합형 a 세포의 막단백질 가용화 분획을 비변성 전기영동에 의하여 인산화 한 결과 인산화 되는 단백질을 발견하였다. 이 단백질의 인산화는 Rhodotorucine A (Rh. A)에 의하여 저해되었다. 본 인산화 단백질에는 trigger-peptidase (TPase)가 존재하고 있는 것을 확인 하였으며, TPase inhibitor(antipain)를 반응액에 첨가할 경우 Rh. A에 의한 인산화 저해작용은 나타나지 않았다. 그리고, Rh. A를 trypsin으로 분해한 산물을 반응액에 첨가했을 경우 인산화 저해현상이 있는 것으로 보아, 인산화의 저해반응에는 TPase가 중요한 작용을 하는 것이 확인되었다.       • glpD and glpE 유전자의 조절영역 결손변이주기가 전사조절에 미치는 영향

        glpD와 glpE는 같은 조절 영역 하에서 121 bp 간격을 두고 역방향으로 전사되어지며, 이들 유전자 상류영역에 존재하는 하나의 CRP binding site가 두 유전자의 전사 조절에 관여함을 이미 여러 연구를 통하여 확인되어졌다. 본 연구에서는 glpD와 glpE의 조절영역이 다양한 크기로 결손된 변이주를 작성하였다. 각 변이된 promoter의 발현조절 영향을 보고자 LacZ와의 융합 plasmid를 작성하였고, β-galactosidase 활성을 측정하여 cAMP에 의한 발현조절을 검토하였다. 발현조절 특성을 확인한 결과 CRP binding site 전까지 결손이 된 변이주는 wild와 비슷한 활성이지만 CRP bin-ding site가 없어진 변이주의 경우는 promoter 활성이 크게 감소했고 cAMP에 의한 발현조절은 나타내지 않았다. 한편 CRP의 공통배열을 삽입시키면 회복이 가능하였다. glpE의 조절영역도 glpD와 마찬가지로 다양하게 결손시킨 변이주를 작성하여 promoter 조절활성을 확인한 결과 CRP binding site가 없어진 변이주의 경우는 promoter 활성이 크게 감소하였으며, 결손된 CRP 결합영역을 삽입시키니 cAMP에 의한 전사촉진이 거의 회복되어졌다. The glpD gene encoding gly-3-p dehydrogenase is essential for the aerobic growth of E. coli on glycerol or gly-3-p. The glpE gene, the function of which is unknown, is transcribed divergently with respect to glpD gene. Expression of the adjacent but divergently transcribed glpD and glpE genes is positively regulated by the cAMP-CRP complex. In this study, for a precise investigation of the functional elements in the regulatory region for transcription activation by cAMP-CRP, deletion mutation have been introducted into the regulatory region. The effect of the deletion mutant on transcriptional regulation was tested in vivo by β-galctosidase activity. Deletion mutants in the regulatory region of glpD demonstrated that the presence of the CRP-binding site resulted in an sixfold increase in promoter activity. And also deletion mutants of glpE gene demonstrated that the presence of the CRP-binding site resulted in an eightfold increase in promoter activity. Insertion of 22 bp oligomer in the deletion mutants has shown that the CRP binding site is need for maximal expression of glpD and glpE genes.

        This study researched the improvement of water treatment process for lake water to drinking water. Especially, we examined water in Lake Shin Gal located nearby Kyung Hee University and investigated the possilbility of the treated water for drinking purpose. Analysis of the treated water gave the satisfactory results for the standard values of drinking water except the several items. However, the water pollution will become serious if the number of factories increase. Therfore, in order to keep possibility of the water in Lake Shin Gal, the number of factories musts be restricted and the application of membrane separation process with general water treatments must be considered.
