      • B형 간염 바이러스 단백질에 있어서 HLA-A2에 의해 표현되는 Epitope 펩타이드 들의 분석

        The cytotoxic T lymphocyte (CTL) are an important component in host defense mechanism against viral infection. They can recongnize virus-derived peptides presented by the Class I MHC molecule at the cell surface of the infected cells. On searching for effective CTL epitopes of hepatitis B virus(HBV), we synthesized a distinct set of 9-10 mer peptide containing amino acid sequence of hepatitis B virus surface protein that are selected on the basis of a computer modeling and the previously described HLA-A2 specific motifs.Binding assay of the synthetic peptides to HLA-A2 molecules using human antigen processing defectant T2 cells showed that 3 out of 4 synthetic peptides enhanced the expression of HLA-A2 molecule on T2 cell surface.Two anchor positions, namely P2 and P9(or P10) appeared to play a decisive role for binding.Structural. characteristics of the peptides addressed by molecular dynamics simulation was analysed and compared.These peptides also partially triggered CTL isolated from human peripheral blood mononuclear cells of HBV positive patients, and the response was peptide-spcific.These results showed that negatively-charged amino acid residue at P2 hampered binding affinity of the peptides to HLA-A2 molecules, and that binding affinity of the peptides are not always reflected by their immunogenicity among natural T cell repertoire.

      • 감꼭지나방 생태적 특성 및 방제약제 선발

        경남지역과 충북지역 감나무의 재배, 해충 방제실태를 농가 방문하여 설문조사 한 결과 감 재배경력은 경남지역이 평균 10년이며, 충북지역은 평균 5년 정도이다. 감 재배 주수는 대다수 농가가 20주 이상이고, 재배관리에 대하여 자문을 받는 곳은 농업센터나 농약사 및 연구소 등이었다. 김 품종은 경남지역에서는 부유와 서촌조생, 충북지역에서는 등시와 월하 등을 재배하고 있었다. 시비종류는 이른봄 에 퇴비나 복합비료를 주었고, 감나무 생장 중 열매나 저장 중 열매에 발생하는 해충이 발견되었다. 해충방제를 위해 살충제를 살포하고 있는데 주로 감꼭지나방, 진딧물, 잎말이나방 및 깍지벌레 등으로 파마치온, 파단, 코니도, 신파마치온, 수프라사이드 등을 살포하였다. 감꼭지나방(Stathmopoda masinissa)의 발육생태를 조사하기 위하여, 감꼭지나방을 야외에서 채집하여 온도±1℃, 광주기 16L:8D, 상대습도 50~60%에서 실내 사육하였다. 각 층태별 기간은 알 7.4일, 유충기간 34.7일, 용 15.5일이었다. 각 령기간은 1령 3.5일, 2령 4.2일, 3령 5.2일, 4령 6.5일, 5령 15.4일이었으며, 특히 유층의 두폭을 측정하여 평균한 결과는 1령 0.20mm, 2령 0.40mm, 3령 0.65mm, 4령 0.87mm, 5령 1.07mm일 이었다. 성충의 수명은 수컷이 6.2일, 암컷이 10.1일 이었다. 용화율은 68.0% 우화율은 59.9%이었다. A survey in Kyungnam and chungbuk areas revealed that the average number of years for persimmon tree farming is 10 and 5 years, respectively. The number of trees per orchard is at least over 20, and the farmers have been usually advised through an agricultural center, an agrochemical store, or agrochemical research center. The varieties of the persimmons they grow are Buyu and Seocheon in Kyungnam, and Doogsi and Wolha in Chungbuk. The kind of fertilizers they sprayed in early spring is either compost or composite fertilizer. persimmon pests are often found in persimmon fruits while on a tree or in storage. Phamathion, Padan, Konido, Sinpamathion, and/or supuraside is usually used to control persimmon fruit moths, aphides, leafrollers, and scale insects. To understand the developmental ecology of the persimmon fruit moth, Stathmopoda masinissa Meyrick, we collected and reared them under the laboratory condition which is at 25±1℃ of temperature, 16L:8D of photoperiod, and 85% of relative humidity. As a result, the developmental period of egg, larva, and pupa were 7.40, 34.7, and 15.5 days, respectively. The average period of each larval instar was 3.50, 4.17, 5.17, 6.46, and 15.42 days, respectively, and the average head capsule width was 0.20, 0.40, 0.65, 0.87, and 1.07mm, respectively. The average adult longevity was 6.2 days for males and 10.1 days for females. The pupation rate and the emergence rate were 68.0% and 59.9%, respectively.

      • 벼멸구의 有機燐系 殺蟲劑 低抗性에 對하여 (Ⅰ) 生物學的 特性 比較

        This paper was carried out to evaluate the difference in the biological characteristics of the brown planthoppers of the insecticide resistant, susceptible strains and their hybrid progenies. The nymphal period was shortened in the susceptible strain as compared with the resistant strain and the F1. The adult emergence was lower in the resistant strain than in the susceptible strain and the F1, but the F1 was not significantly different as compared with the susceptible strain. The sex ratio was not significantly different between the susceptible and resistant strains, but low in the F1. The average body weight of the female and male adults was higher in the resistant strain than that of the susceptible strain and the F1. The preovipositional period was long in the resistant strain as compared with the susceptible strain, but the F1(S X R) was shortened and the F1(R X S) was not significantly different as compared with the susceptible strain. The average number of eggs laid by a female was not significantly different at the 1% level among the susceptible, resistant strains and the F1(S X R), but remarkably decreased in the F1(R X S). The average longevity of the adult was significantly shortened in the Fl than that of the susceptible and resistant strains, but not significantly different between the susceptible and resistant strains. The feeding activity of the female adult BPH infested for 24 hours was remarkably high in the resistant strain as compared with the susceptible strain and the F1, but the feeding activity of the male adult was variable.

      • 萬頃江流域에 있어서 井水中의 弗素含量에 관한 硏究

        This study is the survey about Fluorine contents in the well water of area of Mankyung River side and water of Mankyung River. The result of this survey is as followings: The water of well in this area contained 0.5∼1.3ppm and it's average value was about 0.8 ppm. The water from Mankyung River contained 0.8∼0.9ppm and it's average value is 0.87ppm. By the regionally sectional survey the well water of the area of Mankyung River side, the well water of Chun-Ju, Iri, Samye, and Kusan contained 0.5∼0.8ppm, but the well water of Baikku area contained 0.6∼1.3ppm. Especially in Baikku area 63% of well contained 0.6∼1.0ppm and 38% of well contained 1.1∼1.3ppm. From the above results one can conclude that the 38% of well water of the Baikku area is troubled one.

        GH_3 세포(rat somatomammotropic tumor cell)에서 TRH가 Phospholipase D 활성에 미치는 효과

        연구배경: GH_3 세포는 TRH에 반응하여 세포막의 수용체와 G 단백, PI-PLC, PKC를 활성시켜 성장호르몬 및 프로락틴을 분비한다고 알려져 있다. PLD는 phosphatidylcholine을 phosphatidic acid(PA)와 choline으로 가수분해하는 효소로서 세포의 증식과 호르몬 분비에 관여한다. 본 연구는 GH_3 세포에서 아직 알려지지 않은 TRH의 PLD 활성에 대한 영향을 규명하고자 하였다. 방법: GH_3 세포를 1.5×10^6씩 분주하고 [^3H] myristate로 표지한 다음에 0.3% 알코올로 전 처치하였다. TRH 등의 시험제를 처치한 후에 메탄올로 반응을 종결하고 세포에서 총 지질을 추출하였다. PLD 활성은 박층크로마토그래피를 이용하여 총 [^3H] phospholipid에서 [^3H] phosphatidylethanol의 비율로 구하였다. 결과: TRH (1μM)의 처치 시에 PLC 활성은 44배 증가하였다. PLD 활성은 TRH (1μM), mastoparan (5μM), PMA(500nM)를 30분간 처치 시에 각각 1.9, 1.5, 2.2배 증가하였다. TRH(1μM)의 시간에 따른 PLD 활성 변동은 15, 30, 60, 120, 240분에 각각 142%, 170%, 172%, 160%, 115%의 증가를 보였다. 결론: GH_3 세포에서 TRH의 호르몬 분비와 세포증식의 신호전달 기전으로서는 PLC 활성뿐 아니라 PLD의 활성도 관여함을 시사한다. Backgroud: GH_3 cells are a well characterized and widely used model used for the in vitro study of growth hormone (GH) secretion. Thyrotropin releasing hormone (TRH) binds to receptors belonging to the family of G protein-coupled receptors, and secrets both GH & prolactin. Phospholipase D (PLD) is and enzyme that hydrolyses phosphatidylcholine to yield phosphatidic acid and choline, and plays important roles in cellular proliferation and hormonal secretion. To elucidate the pathway of the action of TRH in GH_3 cells, we investigated the activities of PLC and PLD in GH_3 cells treated with TRH or phorbor 12-myristate 13-acetate (PMA). Methos : GH_3 cells were labeled with [^3H] myristate, followed by incubation of with 0.3% ethanol, prior to before the addition of the agonists. The total lipids were extracted from the harvested cells following treatment with the agonists. The PLD activity was assessed by measuring [^3H] phosphatidylethanol from the [^3H] phospholipid using thin layer chromatography. Results : TRH (1μM) stimulated the PLD activity by 44-fold over that of the control values. TRH (1μM), mastoparan (5μM), and PMA (500μM) for 30 minutes increased PLD activity by 1.9, 1.5 and 2.2 fold, respectively, in comparison to the controls, The PLD activities after 15, 30, 60, 120 and 240 min treatments of TRH (1μM) were 142%, 170%, 172%, 160% and 115%, respectively. Conclusion : These results suggest that TRH stimulates not only PLC activity, but also the PLD activity in GH_3 cells (J Kor Soc Endocrinol 17:465∼472, 2002).

      • 대동맥 이첨판과 승모판 이탈증을 동반한 대동맥 판하부 협착 1예

        Discrete subaortic stenosis is a rare cardiac disorder especially in Orientals which occupies eight to ten percent of aortic stenosis, It occurs mainly before age of eighteen and accompanies with aortic valve abnormalities such as aortic stenosis, supravalvrlar stenosis, bicuspid valve and muscular hypertrophic subaortic stenosis, and other cardiac abnormalities such as ventricular septal defect, pulmonary stenosis and coarctation of aorta. There is no report of discrete subaortic stenosis accompanied with mitral valve prolapse. The major complications of discrete subaortic stenosis are aortic insufficiency, and subacute bacterial endocarditis. We report a case of asymptomatic nineteen-year-old male of discrete subaotic stenosis with bicuspid aortic valve and mitral valve prolapse, which is comfirmed by two-dimensional and Doppler echocardiography including transesophaigeal echocardiography.

      • 장기재령에 따른 고로슬래그를 사용한 모르타르의 강도특성

        The purpose of this study is to examine strength characteristics of mortar using blast-furnace slag for long ages, 365days. In this paper, the mortar replaced by varying fineness and content of blast-fumace slag was investigated through the flow value and the compressive strength. This experimental study was acquired following results by the material tests of fresh and hardened mortar proportioned standard sand and natural sand, mixing proportion of 1:2, 1:3, 1:5, slag fineness of 4500, 5500, 6500㎠/g, water binder ratio 40, 45, 50, 55, 60% and replacement rate of 0, 15, 30, 45%. The results are as follows 1. Blast-furnace slag increased somewhat higher flow value. 2. The higher fineness of slag developed the higher compressive strength. 3. The highest compressive strength appeared at 30% replacement rate. 4. Slightly higher compressive strength appeared at standard sand mortar rather than at river sand mortar.

      • 열대열 말라리아 치료 후 발병한 삼일열 말라리아 1예

        Endemic malaria in the northen parts of the Republic of Korea is caused by P. vivax. And imported malaria cases in Korea from south Asia, Indian subcontinents are usually caused by P. falciparum species. We report a case P. vivax malaria after successful treatment of P falciparum malaria with a review of literatures. Co-infection of P. vivax and P. falciparum, which relapsed only with P. vivax, after successful treatment of P. falciparum malaria was considered. A 38-year old Bangladeshi woman who had lived in Korea visited our hospital due to fever and headache. Eight months ago, she was admitted to a hospital in Calcutta in India because of P. falciparum malaria. She was treated successfully with mefloquine and doxycycline. In this time she was presented with features of malaria with every-other day fever. Awaiting the species identification, we empirically prescribed mefloquine and doxycycline. But the peripheral blood smear showed erythrocytic schizogony, trophozoite and gametocytes typical of P. vivax. So she was finally treated with hydroxychloroquine and primaquine. The patients was well during outpatient follow up period.

        GPS-X 시뮬레이션을 이용한 2단탈질 공정에서 외부탄소원 적용성 평가

        The purpose of this study was to evaluate adaptability of external carbon source using GPS-X program in pilot plant composed with 2-stage denitrification process. The result from analysis of pilot plant operation and GPS-X simulation showed that effluent concentration could be simulated similarly by modifying operation conditions, such as DO concentration, C/N ratio and other calibrated parameter. In order to satisfy the standard of the effluent water quality on T-N of 20㎎/L, it required approximately 3.1 of C/N ratio and 50% of nitrogen removal efficiency when influent T-N is 36.9㎎/L. To maintain the stable water quality of the receiving water, the effluent T-N concentration should be less than 10-15㎎/L and the appropriate C/N ratio to remove nitrogen was 4.27-6.82. The analysis of sensitivity to kinetic coefficient and reaction constant showed that Y_(H) and μ_(mAUT) were most sensitive to nitrate and ammonia nitrogen, relatively and sensitivity coefficient of their were 1.32, 1.98. It was concluded that as Y_(H) decreased and μ_(mAUT) increased, the reaction rates of denitrification and nitrification increased and the removal efficiencies of NO₃^(-)-N and NH₄^(+)-N improved.
