      • Effect of ER signal peptides (KDEL) on expression and function of monoclonal antibody in plant

        Plant genetic engineering led to the production of plant-derived mAb (mAbP), which provides a safe and economically feasible alternative to the current animal expression systems. In this study, the expression level of mAbP SO57 between with and without Lys-Asp-Glu-Leu (KDEL) was compared in transgenic plant. PCR and PT-PCR analyse showed stable gene insertion transcription of heavy and light chain genes of mAbP SO57 with or without KDEL in plant, respectively. We validated expression of mAbP SO57 by western blot. Western blot showed the significantly higher expression level of mAbP SO57 with KDEL compared to without KDEL. Flow cytometry (FACS) analysis showed that the Fc domains of both purified mAbP and mammalian-derived mAb (mAbM) evidenced similar binding activity to the FcγRI receptor (CD64). High performance liquid chromatography (HPLC) analysed, glycosylation patterns of mAbP SO57 with or without KDEL. The mAbP SO57 with KDEL had glycan profile with both oligomannose type (47.6%) and golgi type (52.4%), while the mAbP SO57 without KDEL had only golgi type (100%) glycans. Neutralizing analysis with rabies virus CVS-11 showed the similar neutralizing activity between mAbP SO57 with and without KDEL. These results suggest the potential of mAbP SO57 for rabies immunotherapy, regardless of plant specific glycan structures.

        가스상 대기오염물질에 의한 종이 기록물의 가속열화 특성 연구

        Paper records were generally degraded by some factors from atmospheric environments, like temperature, relative humidity or air pollutants. In this study, the degradation behavior of paper records by single or mixed gases of NO₂, SO₂, HCHO and TVOC was evaluated. The mechanical, optical and chemical properties of 4 kinds of paper (acid and neutral-based printing paper, traditional Hanji, and filter paper) were directly and indirectly affected by gaseous harmful materials. The brightness and L<SUP>*</SUP> value in all papers were slightly increased by accelerated aging under gaseous HCHO and TVOCs, but highly decreased by conditions under gases NO₂ and SO₂. The optical properties of paper records were most vulnerable in acid-based paper and high stable in filter paper and traditional Hanji by air pollutant degradation. The aging treatments under mixed gas pollutants including NO₂ resulted in decrease of physical, mechanical and optical properties of paper, so it was supposed that the concentration of NO₂ gas would be strictly controlled for optimum indoor air quality management in domestic storage centers for paper records.

        Plant genetic engineering has led to the production of plant-derived mAb (mAbP), which provides a safe and economically feasible alternative to the current antibody expression systems. PCR and Reverse Transcription-PCR analyses showed existence of heavy and light chain genes of mAb with or without KEDL and their transcription in plant, respectively. Western blot showed that the expression levels of mAbP SO57 with KDEL were significantly higher than that without KDEL. Flow cytometry analysis showed that the Fc domains of both purified mAbP and mammalian-derived mAb have similar binding activity to the FcγRI receptor. High-performance liquid chromatography analysis showed that the mAbP SO57 with KDEL had glycan profile with both oligomannose and golgi type, whereas the mAbP SO57 without KDEL had only golgi type glycans. Neutralizing analysis with rabies virus CVS-11 showed the similar neutralizing activity between mAbP SO57 with and without KDEL.         SOD2와 NDPK2 유전자 집적에 의한 페튜니아의 아황산가스 저항성 증진

        유전자 집적에 의해 아황산가스 저항성 증진 페튜니아를 개발하고자 비생물적스트레스 저항성 유전자로 널리 알려진 SOD2 와 NDPK2유전자가 각각 도입된 SOD2 형질전환 계통 SOD2-2-1-1-35(T₄)[S(T₄)]와 NDPK2 형질전환 계통 NDPK2-7-1(T₂)[N7-1(T₂)]간 상호교잡을 실시하여 교잡 제1세대를 획득하였다. 교잡 제1세대 중 SOD2와 NDPK2유전자가 모두 집적된 개체 획득율은 32.1-73.0%이었고, SOD2와 NDPK2유전자가 모두 집적된 개체는 SOD2 또는 NDPK2유전자가 단독 도입된 개체에 비해 아황산가스 30ppm처리 피해율이 2.6-5.1배 낮아 아황산가스 저항성이 증진되었음을 확인하였다. 또한 아황산가스 저항성이 증진된 교잡 제1세대를 자가수분에 의해 세대진전 시켜 획득한 교잡 제2세대의 아황산가스 저항성 검정을 통하여 교잡 제1세대에서의 아황산가스 저항성이 후대에서도 안정적으로 발현함을 확인하였다. 또한, 광합성 효율 증진 조사를 통해 아황산가스 저항성이 증진된 SOD2와 NDPK2유전자 집적 후대가 아황산 가스에 대한 스트레스를 덜 받고 있음을 확인하였고, RT-qPCR분석을 통해 SOD2와 NDPK2유전자 집적 후대의 아황산가스 저항성 증진이 SOD2와 NDPK2 유전자 집적에 의한 효과임을 확인하였다. The aim of this study was to develop a transgenic petunia with enhanced resistance to sulfur dioxide (SO₂) gas by stacking two genes, SOD2 and NDPK2, which are both known to confer resistance to abiotic stresses. The first-generation hybrids (TF1) were obtained through reciprocal crosses between an SOD2-transgenic line SOD2-2-1-1-35(T₄)[S(T₄)] and an NDPK2- transgenic line NDPK2-7-1(T₂)[N7-1(T₂)]. Approximately 32.1-73.0% of the first-generation hybrids (TF₁) carried both SOD2 and NDPK2 genes. These hybrids showed 2.6 and 5.1 times less damage than hybrids carrying only SOD2 or NDPK2 genes, respectively, when they were treated with SO₂ gas at 30 ppm. This confirmed that the heterozygous hybrids were more resistant to SO₂ than the hybrids carrying either one of the resistance genes. Second-generation hybrids (TF₂) were obtained by selfing the TF₁ individuals. We confirmed the expression of the stacked genes in the TF₂ hybrids by phenotypic observation of their response to SO₂ gas at 30 ppm as well as using RT-qPCR and photosynthetic efficiency.

        표면근전도와 적외선체열촬영을 이용한 요통의 평가

        목적 : 요통환자에서 요통의 유무와 강도를 평가하고자 표면근전도를 이용하여 근육 활동성의 차이를 검증하였고 적외선체열촬영을 이용하여 제표면 온도의 변화를 측정하였다. 방법 : 요통환자군 16명과 대조군 16명에 대하여 요부에 부착한 표면전극에서 얻은 근전도 신호와 적외선체열촬영 결과를 비교하였다. 교란요인의 영향을 배제하기 위하여 연령과 성별 그리고 비만도에 따라 정상 대조군과 요통환자군을 짝짓기하여 각각 16명씩 선정하였다. 결과 : 정적표면근전도 실시 결과 우측의 경우 중앙값이 요통환자군에서 198.1 ㎶, 정상대조군에서는 161.3 ㎶로 통계적으로 유의한 차이가 있었고(p<0.01), 좌측은 요통환자군에서 194.2 ㎶, 정상대조군에서 180.5 ㎶로 유의한 차이는 없었다. 동적표면근전도 실시 결과 재신전시와 굴곡시 근전위의 비가 우측의 경우 요통환자군에서 중앙값이 1.12였고 정상 대조군은 1.39로서 두 군간에 유의한 차이가 있었고(p=0.027), 좌측의 경우도 요통환자군에서는 1.08이었고 정상 대조군에서는 1.21로 유의한 차이가 있었다(p=0.036). 적외선체열촬영결과 ΔT가 0.5 ℃ 이상의 차이가 있을 때 이상이 있는 것으로 정하면, 요통환자군에서는 16명 모두 이상자로 판정되었고, 정상 대조군에서는 16명중 3명이 이상자로 판정되어 민감도는 100 %, 특이도는 81.3 % 였다. 표면근전도를 이용한 요통의 진단은 민감도가 78.6 %이고 특이도가 72.2 %였으며, 적외선체열촬영은 민감도가 100 %였고 특이도가 87.5 %였다. 결론 : 이러한 결과로 볼 때 표면근전도 검사법과 적외선체열촬영은 요통을 간접적으로 평가하는데 도움이 되는 검사법이며, 적외선체열촬영이 표면근전도검사법에 비하여 보다 정확한 검사법으로 판단된다. Objectives : We studied the efficacy of surface electromyography(SEMG) and digital infrared theimographic imaging (DITI) in the assessment of low back pain both(LBP) Methods : We compared electromygraphic signals from electrodes placed in the lumbar area and the digital theimographic images in 16 LBPpatients and 16 control subjects. The LBP patients and the control subjects were matched for age, gender, and body mass index(BMI) to adjust for any confounding effects Results : In the static analyses of SEMG, median value was 198.1 ㎶ for the controls on the right side, and they were 194.2 ㎶ and 180.5㎶ on left side respectively. There was a statistically significant difference between the two groups on the right side(p<0.01), but not on the left side. In the dynamic analyses of the SEMG, the median values of the extension per flexion ratio of right side were 1.12 for LBP patients and 1.39 for controls. and those on the left side were 1.08 and 1.21 respectively. There were statistically significant differences in both sodes(p<0.05). The sensitivity and specificity were 100% and 81.3% in DITI, 78.6% and 72.2% in SEMG. Conclusions : These results indicate that SEMG and DITI are useful methods for indirect assessments of LBP, and that DITI is more sensitive and specific than SEMG.

      • 남자대학생의 흡연태도와 행동에 관한 연구

        There have been so many researches about problems of cigarettes and relation of smoking and heath since 1950's and generally known that smoking is one of the worst factor of increasing disease and mortality. No matter what such many proofs about various problems of smoking have been researched, smoking rate of current male collage students has not been decreased. Besides, most of the researches about cigarette use and its addiction have been focused on the adolescent or adult women smokers or adult male, not male collage student. Therefore, the subject of this study is about smoking attitude and smoking behavior of male collage students by general features. Smoking attitude was analyzed by modifying 'Guidelines for the content of tobacco smoking surveys of the general population'(WHO, 1982). Two hundred of male collage students participated to the distributed questionnaire for one week in May. The smoking attitude was categorized into duration of smoking and number of cigarette per day(CPD), and nicotine dependency, investigated by Fagerstrom Tolerance Questionnaire(FTQ) (Fagerstorm & Schneider, 1989). All statistics were performed by procedure of SAS according to number, percentage, mean, standard deviation, and ANOVA. The results are as follows: 1. Smoking attitude level was mean 25.73±5.15 marks, ranging from 13 to 39 marks of the total 40 marks. 2. Duration of cigarettes, number of cigarette per day, and nicotine dependency were mean 5.14±2.84 years, 12.88±6.85 pieces, and 4.78±1.69, respectively. 3. Smoking attitude showed significance according to general features such as marital state and allowance, but the significance according to marital state is not valuable because the numerical difference is so big between married one and single that they cannot be compared with each other. 4. Duration of smoking was significant according to general features such as age and allowance. 5. Smoking amount showed significance the factors of age only. 6. ll1ere was no significance in nicotine dependency according to general features. As a result, smoking attitude of male collage student in this study shows moderate level. Smoking attitude and duration is significant according to allowance, and smoking amount is significant according to age. Nicotine dependency according to general features is not significant but comparing with the preceding study about adult male, male collage students in this study are respectively more nicotine-dependent than adult male smokers. In conclusion, the right allowance usage strategies and effective heath education focused on the young are needed. In addition, helpful programs and information spreading strategies are necessary for establishing higher level of smoking attitude. At last, systemic regulations in daily living for ""anti-nico-dependent"" and perspective reforming of male university student are needed for reducing nicotine dependency.

      • 일반논문 : 통신자료제공제도(전기통신사업법 제83조 제3항)의 문제점과 개선방안 -ISP에 대한 수사기관의 통신자료제공 요청과 이용자의 정보인권 보장을 중심으로

        이 연구에서는, 수사기관이 이용자의 인적사항 등을 요청하면 전기통신사업자가 이에 응할 수있도록 한 ‘통신자료제공제도(전기통신사업법 제83조 제3항)’의 문제점과 개선방안에 대해 논한다. 이 제도에 대한 논란은 2010년부터 본격화되기 시작했으며, 2012년에 있었던 헌법재판소 결정과 고등법원 판결로 인해 사회적 이슈로 떠올랐다. 통신자료제공제도의 활용 건수는 매년 큰 폭으로 증가하는 추세이다. 이런 현상의 본질적인 이유는 ISP(인터넷서비스제공자)가 수사기관의 요청에 대해 예외 없이 통신자료를 제공하고 있다는 사실에 있다. 현행법상으로 통신자료제공 여부를 결정하는 주체는 법원이 아닌 ISP다. 그러나 ISP는 이러한 결정을 할 역량이 부족할 뿐더러 거절 시 받게 될 수사기관으로부터의 압박에서 자유롭지 못하다. 이 때문에 통신자료제공제도는 법문상 ‘재량적’으로 규정되어 있음에도 불구하고 사실상 ‘강제적’으로 운용되고 있다. ‘정보의 유통에 대한 개인의 통제권’을 그 주요 내용으로 하는 정보인권은 새로운 영역의 권리를 보편적 권리로 격상시킨 ‘새로운 인권담론’이며, 동시에 기존의 인권담론과 그 맥락을 같이하고 있는 ‘기본적 인권’이다. 특히 정보와 관련된 권리들이 기존의 권력관계와 맞물리며 작동할 수밖에 없다는 점에서 정보인권을 소수자 문제와 연결지어 생각할 필요가 있다. 통신자료제공제도에서 쟁점이 되는 정보인권은 익명표현의 자유와 개인정보자기결정권이다. 우선 통신자료제공제도는 2012년 위헌 결정이 내려진 ‘제한적 본인확인제(일명 인터넷 실명제)’와 마찬가지로 익명 표현의 자유를 침해한다. 이는 익명 표현의 자유라는 이용자의 개인적 권리를 침해하는 것에서 더 나아가 이용자의 자유로운 의사표현을 바탕으로 여론을 형성하는 ISP의 언론의 자유 또한 제한하는 결과를 낳는다. 그 외에도 통신자료제공제도는 개인정보자기결정권을 근본적으로 침해한다. ISP가 제공하는 통신자료는 이용자의 개인정보에 해당한다. 그러나 이를 제공함에 있어 정보주체인 이용자의 동의를 요구하는 등의 사전개입절차가 없고, 사후통지절차 또한 미비하다. 민감한 개인정보에 대하여 그 어떤 통제장치도 마련되고 있지 않은 것은 기타 유사한 제도와 비교해 보았을 때도 불합리한 것으로 평가된다. 그러므로 통신자료제공제도는 폐지 또는 개정되어야 한다. 우선 ‘수사기관의 통신자료 취득행위’를 대상으로 헌법소원심판을 청구함으로써, 부수적 규범통제에 의해 통신자료제공제도 자체에 대한 위헌결정을 받아내는 방법이 있다. 이와 달리 입법론적으로는 이 제도를 삭제하거나 수정하는 방법도 생각해볼 수 있다. 우리가 상정하는 새로운 입법안은 영장주의를 기본원칙으로 채택하고, 이용자의 사전개입절차 및 사후통지절차를 마련하는 것을 그 내용으로 한다. 통신자료제공제도는 어디까지나 ‘통신비밀 누설 금지 원칙’의 예외에 해당한다. 그럼에도 지금과 같이 주객이 전도된 상태가 방치되고 있는 것에는, 정보인권 침해에 대해 미온적인 태도를 보이고 있는 국가의 잘못이 크다. 국가가 나서지 않는다면 이 제도의 잠정적 피해자인 국민들이라도 나서야 한다. 정보인권이야 말로 새로운 민주주의를 위하여 다시 한 번 쟁취해 내야 할 우리의 ‘인권’이기 때문이다. This paper deals with the problems and improvement plans of the Communication Data Provision System(Telecommunications Business Act §83-3) which allows ISP (Internet Service Provider) to provide users` personal data when requested by investigation agencies. The controversy over this system kicked into high gear in 2010 and became a main social issue after the decision of the Constitutional Court and the ruling of the High Court in 2012. The use of the Communication Data Provision System has considerably increased year by year. This trend largely originates from the reality that ISP provides investigation agencies with users` personal data with few exceptions. Under current law, it is not the Court but ISP that decides if providing users` personal data is approved or not. However, ISP does not have enough ability to make such decision; moreover it cannot avoid the pressure that investigation agencies may exert especially when ISP refuses to provide data. Because of this, although the law leaves the provision of user`s personal data ‘to discretion’, the Communication Data Provision System is more likely to be operated ‘by force’. The discourse about information human rights, which defines the individual`s control over the distribution of information as its core content, has contributed to newly raising the status of information human rights up to the stream of the universal rights. Also it has served as the discourse about ‘fundamental human rights’ which extend the realm of the previous human rights discourse. Especially given that information-related rights are not free from the existing power relations, we have to further consider minority-related issues with regard to information human rights. The freedom of anonymous speech and the right to informational self-determination are the two issues mainly discussed about information human rights with regard to the Communication Data Provision System. First, the Communication Data Provision System can infringe the freedom of anonymous speech, similar to the Restrictive Identification System(the so-called Internet Real-name System) which was ruled unconstitutional by the Constitutional Court in 2012. This can breach not only the user`s individual rights, the freedom of anonymous speech, but also ISP`s freedom of speech that makes ISP shape public opinion based on users` free expression of opinion. In addition, the Communication Data Provision System fundamentally infringes the right to informational self-determination. The communication data provided by the ISP is the user`s personal information. However, there is no proceeding for users` prior intervention, such as requesting process for users` consent, nor is there any follow-up proceeding to notify the users. Given the other similar systems, the absence of control system whatsoever on such sensitive personal information can be problematic. Therefore, this paper argues that the Communication Data Provision System should be abolished or revised. The first alternative to the present system is to file a petition in an attempt to get a decision of unconstitutionality on the Communication Data Provision system from the Constitutional Court. By filing a constitutional appeal against the investigation agency`s ‘acquisition’ of communication data, the system can be found unconstitutional through incidental norm control(Incidenter-Kontrolle). Secondly, the statement that this system should be deleted or revised can be supported by the principle of legislation. The new legislation bill projected in this study includes warrant requirement as a basic principle as well as user`s prior intervention proceedings and follow-up notice proceedings. The Communication Data Provision System has been exempted from the basic principle, ‘prohibition of communication secrets` leakage’. The main reason why this preposterous situation still exist is closely related to the lukewarm attitude of the government toward the information of information right. If the government does not take any measures, then at least the citizens, the potential victims of this system, should do so. Information human rights is the ‘human rights’ that we should strive again to achieve for a new democracy.

      • HSS STI-CMP적용을 위한이중 패드의 최적화

        As the device geometry shrinks to the deep submicron region, chemical mechanical polishing(CMP) planarization become a more essential technique of advanced ULSI process. Also, CMP process was required for the global planarization of inter-metal dielectric(IMD), inter-level dielectric(ILD) layers and interconnections with free-defect. Especially, the complete global planarization of IMD, ILD and interconnections can be achieved only with the CMP process. However, as the IMD and ILD layer gets thinner, several problems were found in the CMP process. It does have various problems such as dishing effect, torn oxide defects and nitride residues in oxide. So, it leads to severe circuit failure, which affects yield. In this paper, we studied the characteristics of polishing pad, which can apply STI-CMP process for global planarization of multilevel interconnection structure. Also, we investigated the effects of different sets of polishing pad, such as soft and hard pad. As an experimental result, hard pad showed center-fast type, and soft pad showed edge-fast type. Totally, the defect level has shown little difference, however, the counts of scratch was defected less than 2 on JR111 pad. Through the above result, we can select optimum polishing pad, so we can expect the improvement of throughput and device yield.

        The average indoor concentration of the library's TSP was 0.03 ng/m³. TAB in library and lounge in college of engineering showed higher concentrations in corridors than indoors. The correlation coefficient between TAB and humidity is 0.985, so it is judged that there is a significant need for humidity control in indoor air quality management to reduce the concentration of total suspended bacteria(p<0.01).
