      • The Effect of pH and Various Cations on the GTP Hydrolysis of Rice Heterotrimeric G-protein α Subunit Expressed in Escherichia Coli

        Previously, we reported the biochemical properties of RGA1 that is expressed in Escherichia coli (Seo et al., 1997). The activities of RGA1 that hydrolyzes and binds guanine nucleotide were dependent on the $MgCl_2$ concentration. The steady state rate constant ($k_{cat}$) for GTP hydrolysis of RGA1 at 2 mM $MgCl_2$ was $0.0075{\pm}0.0001\;min^{-1}$. Here, we examined the effects of pH and cations on the GTPase activity. The optimum pH at 2 mM $MgCl_2$ was approximately 6.0; whereas, the pH at 2 mM $NH_4Cl$ was approximately 4.0. The result from the cation dependence on the GTPase (guanosine 5'-triphosphatase) activity of RGA1 under the same condition showed that the GTP hydrolysis rate ($k_{cat}=0.0353\;min^{-1}$) under the condition of 2mM $NH_4Cl$ at pH 4.0 was the highest. It corresponded to about 3.24-fold of the $k_{cat}$ value of $0.0109\;min^{-1}$ in the presence of 2 mM $MgCl_2$ at pH 6.0.

        통어적 기능을 고려한 명사토의 형태론적 구조 분석과 분류

        This paper critically reviews some previous analyses of Korean Noun inflectional affixes(N-infl). And this paper is a work for reflecting morphologic and syntactic properties of N-infl. I analyze Korean Noun and N-infl into morphologic structure. And I introduce the feature structure and principle of HPSG for its syntactic function discussion. I explain that the N-infl unites the Korean noun stem and other inflectional affixes as a agglutination and linguistic information of Korean inflectional affixes unite the linguistic information of Korean noun stem and other inflectional affixes as the principle of 'unification'.         An Optimality Theoretic Approach to Semi-vowel [j] Jo, Hak-haeng and Seo, Jeong-min(Chonsun Univ.) There have been three kinds of arguments about the status of semi-vowel [j] in Korean within the Korean syllable structure. The three theories, Nucleus Hypothesis, Onset Hypothesis, and Tautosyllabic Hypothesis, consider the semi-vowel [j] in Korean as part of nucleus, onset, and both respectively. Each theory, however, has provided conflicting accounts in reference to analogous evidence. Thus, in this paper we argue that the permission of semi-vowel [j] of Seoul dialect in Korean can be given a functionally grounded treatment by means of *ONSET/X subhierarchy(Smith, 2003: 3) that maintains its universally fixed ranking based on the sonority scale, if the *ONSET/X is defined to be sensitive to the moracity of the segments. And then according to Optimality Theory(Prince & Smolensky, 1993) we will analyse semi-vowel [j] of Seoul and Chonnam dialect in Korean as nucleus. And then the result will be extended to the semi-vowel [j] in English.

        허웅의 ‘훈민정음’에 대한 인식과 영향관계 : 19세기 말∼20세기 전반기, ‘국어학’ 학술네트워크 고찰을 위하여

        ‘훈민정음’은 일제강점기 이후 일본 제국에 저항할 수 있는, 민족적 자부심이 담긴 대표적인 유산으로 여겨왔다. 그리고 그러한 인식은 19세기 말∼20세기 초의 지식인, 특히 국어학자들의 훈민정음에 대한 연구성과와 직접 연결되어 있다. 특히 ‘허웅’은 20세기 전반기의 국어학 연구나 언어에 대한 인식 등을 현재까지 잇는 중간 역할을 하였다는 측면에서 ‘허웅’의 훈민정음 인식에 대해 주목할 필요가 있다. 이 연구는 앞선 연구에서 주로 다루어 온 주시경, 권덕규, 김윤경, 최현배, 홍기문 등의 연구 성과를 바탕으로 허웅의 ‘훈민정음’에 대한 인식이나 연구에 대해 고찰하고자 한다. 그것은 허웅(1974가, 나)에서 볼 수 있듯이 그의 말과 글의 관계나, 한글전용과 같은 언어 정책에 대한 입장 등은 대부분 ‘훈민정음’ 특히 ‘어제서문’의 해석과 관련하여 설명되고 있어서 그의 ‘훈민정음’에 대한 인식을 검토할 필요성이 있기 때문이다. 한편 허웅은 학문에 대한 태도는 실증적이고 엄정하다는 평가를 받고 있고 스스로도 그러한 관점에서 연구하고자 하였다. 그러나 훈민정음 서문의 해석에서는 앞선연구의 전통을 그대로 이어받아 실증적이고 비판적인 태도를 찾아보기 힘들었다. 이 연구에서는 그러한 허웅의 관점이 허웅의 학문적 태도에서 출발했다기보다는 훈민정음에 대한 우리의 인식이 그만큼 맹목적인 부분이 있었기 때문이었음을 강조하고자 하였다. 허웅의 훈민정음에 대한 인식은 『한글과 민족 문화』(1974ㄱ)에 선명히 나타나 있는데, 이외에도 『우리말과 글의 내일을 위하여』(1974), 『우리말과 글에 쏟아진 사랑』(1979), 「세종의 언어 정책과 국어 순화 정신」 (1980), 『이삭을 줍는 마음으로』(1981) 등에서도 그의 훈민정음에 대한 인식을 엿볼 수 있다. 이 연구에서는 위의 자료를 바탕으로 허웅의 훈민정음에 대한 인식을 고찰하고자 한다. Since the flowering period, ‘Hunminjeongeum’ has been understood as a representative heritage showing the pride of the people who oppose the Western modernity or resist the Japanese empire. In this process, it was natural that the research related to the Hunminjeongeum of intellectuals, especially Korean scholars, became the basis. So far, in the previous study, discussions about Hunminjeongeum such as Ju Si Kyung, Kwon Duk Kyu, Kim Yoon Kyung, Choi Hyun Bae, and Hong Gi Moon have been discussed to some extent. However, there is a need for this research in that the review of the ‘Hunminjeongeum’ by Mr. Huh has not been done yet. As we can see in Huh Woong(1974, I) in particular, the relationship between his words and writings, and the position of language policy, such as Hangul, are mostly explained in relation to the contents of ‘Hunminjeongeum’, especially ‘Preface’, which makes it more necessary to examine his perception of ‘Hunminjeongeum’. On the other hand, as discussed in Seo Min-jeong(2011), “Hunminjeongeum” and “Preface” were translated into the language of the 15th century (say) and translated into the language of the 20th century in the first half of the 20th century. The material that reveals Huh Woong’s awareness of Hunminjeongeum is “Hangul and National Culture”(1974). Other than that, for the sake of Korean and Writing tomorrow (1974), Korean and Writing Love(1979), Sejong’s Language Policy and Korean Refinement Spirit(1980), Isaac’s Mind, etc.(1981), and the Korean and Korean Cultures(1974, Sejong University, and the Korean People’s Association(1974), The Korean language policy is also discussed, and the teacher’s recognition of the Hunminjeongeum can be said to be followed by the Korean language policy. Based on the above data, I would like to examine the perception of Hunminjeongeum by Mr. Huh. In this study, we found that the attitude of Huh Hung"s scholarship showed a different pattern in the interpretation of the preface of Hunminjeongeum. And it was examined that such aspect did not overcome the limitations of the situation of the times rather than the problem of attitude of Heung. Based on these considerations, we should recognize that it is time to reinterpret and evaluate Hunminjeongeum.

        Dimethylnitrosamine 유발 급성 간 손상 흰쥐에서 ^(99m)Tc-Lactosylated Serum Albumin을 이용한 간 기능의 평가

        목적: ^(99m)Tc-lactosylated serum albumin (^(99m)Tc-LSA)은 간세포에 특이적으로 결합하는 간수용체 영상용 방사성의약품으로 새로이 합성되었다. 간섬유화를 유발하는 dimethylnitrosamine (DMN)을 투여한 간 손상 휜쥐 모델에서 ^(99m)Tc-LSa의 역동학적인 간섭취를 조사하고 간효소치의 변화와 조직학적 소견을 비교하여, LSA의 간섭취가 간기능의 변화를 반영하는지를 연구하였다. 대상 및 방법: SD계 흰쥐에 DMN를 27 mg/kg으로 복강 내 주사하여 급성 간손상을 유도하고 대조군과 비교하였다. DMN을 주사한 흰쥐를 3일(DMN-3), 8일(DMN-8), 21일(DMN-21)에 ^(99m)Tc=LSA (1,665 mg/kg) 29 MBq를 정맥 주사하여, 30분 동안 동적 영상을 획득하고 간과 신장부위에 관심영역을 설정하여 간과 심장부위의 시간방사능 곡선을 얻었다. 간기능 평가를 위해 시간방사능 곡선을 이용하여 간섭취지수와 혈중제거지수를 구하였고 곡선 최적화를 시행하였다. DMN 투여군과 대조군의 간효소치의 변화와 간조직의 광학현미경 소견을 비교하였다. 결과: 대조군에서는 ^(99m)Tc-LSA가 빠르게 간에 섭취되고 혈중에서 제거되었으나 DMN을 처리한 군에서는 간섭취가 낮았다. 간섭취지수의 비교에서 대조군에 비해 DMN 처리군에서 유의하게 간섭취지수가 낮았다(DMN-3: 0.842, DMN-8: 0.898, DMN-21: 0.91, 대조군: 0.96, p<0.05). 혈중제거지수의 비교에서도 대조군에 비해 DMN 처리군에서 혈중제거지수가 유의하게 높았다(DMN-3: 0.731, DMN-8: 0.654, DMN-21: 0.604, 대조군: 0.473, p<0.05). 비선형 회귀분석에서 R_(2) 값은 0.9이상으로 좋은 일치를 보였고, 대조군에ㅓ K값이 DMN처리군에 비해 크고(DMN-3: 0.28, DMN-8: 0.41, DMN-21: 0.46, 대조군: 0.97, p<0.05), T_(1/2)값은 작았다(DMN-3: 2.5, DMN-8: 1.7, DMN-21: 1.5, 대조군: 0.7, p<0.05). 간효소치의 변화는 DMN-3군에서는 대조군에 비해 상승하였으나 DMN-8, DMN-21군에서는 간효소치의 상승이 관찰되지 않았다. 간조직 소견의 경우 DMN-3군에서 중심정맥 주위에 괴사가 관찰되었으나 DMN-8군, DMN-21군에서는 미약한 정도의 염증세포 침윤만이 관찰되었다. 결론: ^(99m)Tc-LSA 간신티그래피의 간섭취 정도는 간손상과 반비례하였으며 간섭취의 변화는 조직학적 손상이 심한 정도와 간손상후 회복되는 과정을 반영하여 주었다. ^(99m)Tc-LSA 간신티그래피가 간손상을 평가하고 간손상후 회복되는 과정을 추적하는 간수용체 영상용 방사성 의약품으로 사용될 수 있을 것으로 생각된다. Objects: ^(99m)Tc-lactosylated human serum albumin(LSA) is a newly synthesized radiopharmaceutical that binds to asialoglycoprotein receptors, which are specifically presented on the hepatocyte membrane. Hepatic uptake and blood clearance of LSA were evaluated in rat with acute hepatic injury induced by dimethylnitrosamine(DMN) and results were compared with corresponding findings of liver enzyme profile and these of histologic changes. Materials and Methods: DMN (27 mg/kg) was injected intraperitoneally in Sprague-Dawley rat to induce acute hepatic injury. At 3(DMN-3), 8(DMN-8), and 21(DMN-21) days after injection of DMN, LSA injected intravenously, and dynamic images of the liver and heart were recorded for 30 minutes. Time-activity curves of the heart and liver were generated from regions of interest drawn over liver and heart area. Degree of hepatic uptake and blood clearance of LSA were evaluated with visual interpretation and semiquantitative analysis using parameters (receptor index : LHL3 and index of blood clearance : HH3), analysis of time-activity curve was also performed with curve fitting using Prism program. Results: Visual assessment of LSA images revealed decreased hepatic uptake in DMN treated rat, compared to control group. In semiquantitative analysis, LHL3 was significantly lower in DMN treated rat group than control rat group (DMN-3:0.842, DMN-8: 0.898, DMN-21: 0.91, Control: 0.96, p<0.05), whereas HH3 was significantly higher than control rat group (DMN-3: 0.731, DMN-8: 0.654, DMN-21: 0.604, Control: 0.473, p<0.05). AST and ALT were significantly higher in DMN-3 group than those of control group. Centrilobular necrosis and infiltration of inflammatory cells were most prominent in DMN-3 group, and were decreased over time. Conclusion: The degree of hepatic uptake of LSA was inversely correlated with liver transaminase and degree of histologic liver injury in rat with acute hepatic injury.

        Production of 5,8-dihydroxy-9(Z)-octadecenoic acid from oleic acid by whole recombinant cells of Aspergillus nidulans expressing diol synthase.

        <P>The optimal conditions for the production of 5,8-dihydroxy-9(Z)-octadecenoic acid (5,8-diHOME) from oleic acid by whole recombinant Escherichia coli cells expressing diol synthase from Aspergillus nidulans were 40 °C, pH 7.5, 10 % (v/v) dimethyl sulfoxide, 35 g cells l(-1), and 12 g oleic acid l(-1) at 250 rpm in a 250 ml baffled flask. Under these conditions, whole recombinant cells produced 5.2 g 5,8-diHOME l(-1) together with 1 g l(-1) of the intermediate 8-hydroperoxy-9(Z)-octadecenoic acid (8-HPOME) after 60 min. This corresponded to a conversion yield of 43 % (w/w), a volumetric productivity of 5.2 g l(-1 )h(-1), and a specific productivity of 148 mg g cells(-1 )h(-1). This is the first report of the biotechnological production of 5,8-diHOME from oleic acid.</P>

        Process Design and Fabrication of Sample for Non‑contact Liquid Crystal Alignment with High Yield

        In this study, we report the results of an experiment for aligning liquid crystals (LCs) by irradiating ultraviolet rays that are not polarized to an alignment layer, and a simulation result on the alignment principle. In the case of irradiating un-polarized ultraviolet rays to the substrate coated with the alignment flm, there have been many experimental reports on the alignment mechanism of the LC due to the anisotropy generated in the substrate, but the theoretical approach is somewhat lacking. As a result of the experiment, the best orientation was observed at around 45°. As a result of the simulation, it was found that the maximum diference between the p-polarization and the s-polarization was within the range of similar angles there was. The theoretical approach to the alignment mechanism of LC was possible. And we have experimentally confrmed that the mechanism of liquid crystal alignment by conventional Groove theory is possible by irradiation of unpolarized UV light. The AFM analysis confrmed that the surface anisotropy that causes this orientation principle actually occurs in the unpolarized UV light. It is expected that this non-contact orientation method can realize higher production yield than the conventional contact type rubbing method, and the result of this study could contribute to the improvement of the yield.

        석연 양기훈(石然 楊基薰) 노안도 연구

        This paper focused on life and Noando(蘆雁圖) of Seokeon(石然) Yang Gi-hun(楊基薰, 1843-1911) who had worked in the late 19th and early 20th century. Yang Gi-hun was not just a court painter, but he was an actually occupational painter of noble birth from generation to generation in Pyongyang. He was also recommended to a position late in life at 49, and he started his career as a low-ranking government official. In 1905, he was promoted to Jeong-3-poom(正三品), however, he focused on paintings in several areas like Pyongyang, Seoul and Japan. Meanwhile, he was known to paint 《The Album leaf of The United States(美國風俗畵帖)》 after following Min Yeong-ik(閔泳翊, 1860-1914) to the US in 1883. But this should be considered again because it doesn’t match with his youngest son Yang Yu-jin(楊瑜鎭, 1884-?)’s birth time. Yang Gi-hun presented works with Sinjagwan(臣字款) to court as a local painter exceptionally. This Sinjagwan painting reflected the change of court paintings form of production. Thus, as a local artist, his special backgrounds and his works to a superior had important roles to represent for changed aspect of the age. Yang Gi-hun had finished short life in Seoul for 4 years and he moved to Japan in 1908. But he returned to Korea because of an injury in 1909 and Before long, died in 1911. The most famous of Yang Gi-hun’s paintings is NoAndo. He created his own form in detailed expression of paintings in Joseon Dynasty traditional style. His style is different from Jang Seung-eop(張承業, 18431897) and Seoul Painting Circles’s painters. His Noando was produced in various ways. Especially embroidery Noando based on his original works was connected with the growth of local embroidery artisan groups such as Anju district in Southern pyeong"an province. Yang Gi-hun’s Noando exerted influence on following generations of painters in Pyongyang. Yang young-jin(楊英鎭, 1881-1918), Kim Yun-Bo(金允輔, 1865-1938) and Moon Hwa-Sun(文化善) were representative painters influenced him. They also passed Yang Gi-hun’s style down to Women Gisaeng(妓生) Artists, Joo San-woll(朱山月, 1894-1982) and Ham In-sook(咸仁淑). It shows genealogy of Pyongyang painting circles in the Modern Era. Through his activities and artworks, we could examine on painters aspect and new tendency. And also I hope this paper is of some help to understand local artists’ way of existence in late 19th century.
