      • 물곰팡이 Allomyces macrogynus의 홀씨와 팡이실에 존재하는 칼슘이온결합 단백질의 검색

        45Ca overlay를 이용한 방법으로 Allomyces macrogynus의 생장과정에서 형성되는 암수배우자 홀씨, 반수체 홀씨, 그리고 이배체 홀씨등의 세가지 홀씨와 팡이실에 존재하는 칼슘이온결합단백질들을 검색하였다. 세가지의 홀씨 각각에서는 모두 5가지 종류의 칼슘이온결합단백질들이 동일하게 관찰되었다. 팡이실에서는 적어도 6개 이상의 칼슘이온결합단백질들이 관찰되었으며, 이중에 3가지는 홀씨에서 관찰된 것과 동일한 것으로 판단된다. Ca2+-dependent phenyl-sepharose를 이용하여 팡이실에서 SDS-PAGE 겔 상에서 19kDa의 분자량을 가지는 칼슘이온결합단백질을 분리하였다. 이 단백질은 등전점이나 열에 대한 안정성, 칼슘이온에 의한 전기영동상에서의 움직임의 변화 등으로 판단하건 데 calmodulin으로 추정된다. 따라서 Allomyces macrogynus의 홀씨와 팡이실에는 calmodulin을 비롯하여 다수의 칼슘이온결합단백질들이 존재하며, 이들 중에는 홀씨, 또는 팡이실에서만 관찰되는 것들도 있었다. Calcium binding proteins(CaBPs) present in 3 different types of zoospores and in mycelium of Allomyces macrogynus were screened by using 45Ca overlay method. Five different kinds of CaBPs were equally observed in gametes, haploid spores, and diploid spores, respectively. Six kinds of CaBPs were observed in fractions of DEAE-cellulose chromatography of mycelial extract. Among them 3 CaBPs were shown to be identical with those detected in zoospore extract. 19-kDa CaBP was isolated from mycelial extract by Ca2+-dependent phenyl-sepharose chromatography. The 19-kDa mCaBP showed calmodulin-like properties as determined by molecular weight, isoelectric point, and Ca2+-dependent mobility shift on SDS-PAGE. It is shown that a variety of CaBPs including calmodulin-like protein are present in zoospores and mycelium of Allomyces macrogynus and some are confined to either.

        A 6 years old Schunauzer dog (7.2kg, intact female) with history of hematuria for two days was referred to the Veterinary Medical Teaching Hospital of Chungnam National University. Complete blood cell count showed no significant abnormalities and serum chemistry profile showed increased phosphate profile. On urinanalysis, there was evidence of bacterial infection. On ultrasonographic evaluation, irregular renal contour and increased echogenecity, pelvic and uretral dilation, and thickening of urinary bladder wall were observed. Based on these finding, the dog was diagnosed as pyelonephritis. The dog was treated with antibiotics based on antimicrobial susceptibility test results, then successfully resolved after 8 weeks of treatment.

        버섯첨가된장의 생리활성 작용

        The study was carried out to evaluate the radical scavenging activity, and antimutagenic and anticancer effects in Korean soybean paste(doenjang) added with various mushrooms. Ganoderma lucidum doenjang(Gl-TD) showed significant inhibitory activity against the 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical, with an IC50 value of 245㎍/㎖. In contrast, the other doenjang varieties appeared to have weaker antioxidant activity. Four kinds of doenjang didn't have any antimutagenic activity against N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) by adding of 5mg/plate, but exhibited a strong inhibitory effect(55~70%) against aflatoxin B1(AFBI). Especially Phellinus linteus doenjang(Pl-TD) inhibited more than 70% of the mutagenicity induced by AFBI in Salmonella typhimurium TA100. On the other hand, the mushroom doenjang varieties showed relatively weak activity toward MNNG in SOS chromotest, their inhibitory rate ranging from 23% to 33%. Methanol extracts of Gl-TD and Pl-TD inhibited by 91%~92% the growth of AGS gastric cancer cells in a concentration of 100㎍/㎖.

        횡 요천골부 후피부판 (Transverse Lumbosacral Back Flap)을 이용한 천골부 욕창 치험례

        Pressure sore is an area of ischemic tissue loss resulting from prolonged or often repeated pressure, usually over a bony prominence of body. Recently, the prevalence of pressure sore is major clinical problem, due to the increased number of patients with spinal cord injuries or longterm comatous patient after traffic, industrial accidents. The surgical principles in the treatment of pressure sores are total excision of the ulcer, scartissue, bursae, complete removal of all infected bone with recontouring of bony prominences to alleviate discrete pressure points and resurfacing and padding of the defects with healthy and well vascularized tissues. The sacral sore is the most common site for the development of pressure sore in Korea. We had experienced 14 cases who had received surgical treatments for sacral sores with transverse lumbosacral back flap. We obtained the following conclusions. Advantages 1)It is quick and easy procedure 2)The flap elevation itself is relatively bloodless 3)The donor defect is in a non-weight-bearing area 4)Muscle is not sacrified(possibly of importance in the ambulatory patient) 5)Surgical options are preserved in cases of recurrence(the flap itself may be reelevated and inset in some cases of recurrence) Disadvantages 1)It is lacking the bulky padding afforded by alternative musculocutaneous flaps 2)The vascularity of the flap tip is empirically less dependable than that of a musculocutaneous flap 3)The donor defect is large, cannot usually be closed primarily, and may prove of cosmetic concern

        야뇨증 환아들의 심리사회적 특성에 대한 다기관 연구 : 행동 및 정서 문제를 중심으로

        Objectives : The aim of this study was to examine the behavioral and emotional problems associated with nocturnal enuresis in Korean children. Methods : Three hundred eighteen children with nocturnal enuresis, together with their parents, completed the Child Behavior Checklist (CBCL), Disruptive Behavior Disorder Scale according to DSM-IV (DBDS), Children's Depression Inventory (CDI), State-Trait Anxiety Inventory for Children (STAIC), and Piers-Harris Children's Self-Concept Scale (PHCSC). Ninety-three normal students were selected as the control group. Results : Compared to the normal control group, the mean scores with regard to the withdrawn, social problems, attention problems, delinquent behavior, aggressive behavior, externalizing problems and total problems profiles were significantly higher in the nocturnal enuresis group according to the CBCL results. The nocturnal enuresis group also scored significantly higher in the ADHD and ODD profiles of the DBDS. The nocturnal enuresis group was more depressed and anxious than the control group according to the results of the CDI and STAI. The mean score of the PHCSC was significantly lower in the nocturnal enuresis group when compared to the normal control group. Conclusion : The results of this study suggest that children with nocturnal enuresis in Korea have clinically relevant behavioral and emotional problems. The findings support the link between nocturnal enuresis and psychopathology in Korean children.

        운동성 산화 스트레스와 항산화비타민의 보충이 말초단핵세포의 NF-kB 활성에 미치는 영향

        Reactive oxygen species(ROS) are implicated in the pathogenesis of a wide variety of human diseases. Numerous studies indicate that ROS may serve as common intracellular molecules that contribute to the process of nuclear factor(NF)κB activation in response to a diverse stimuli. In our laboratory, we have demonstrated tat antioxidants could reverse the decline of immune function caused by exercise-induced ROS. Furthermore, it in necessary to understand a mechanism underlying ROS-dependent disorder in biological system. Recent studies have been shown that several gene expression were regulated by oxidants, antioxidants and other determinants of the intracellular reduction-oxidation(redox) state. In this process, NF-κB have been shown to play a important role. The purpose of the present investigation was to evaluate the effect of exercise-induced oxidative stress and antioxidnt supplementation on NF-κB activation in peripheral mononuclear cells. Forty male SD rats(4 weeks old) were randomly divided into noraml diet group and antioxidants(ATO) supplement group, and then ATO groups were treated with antioxidants(VE: 2001U/kg, VC: 50mg/rat, β-carotene: 300mg/kg, vitamin B6: 250㎍/100g, selenomethionine: 0.1mg/kg) for 16 weeks. After 16 weeks breeding at each condition, each group divided into two groups: Control group(CR) fed generally, Exercise group(CE) fed generally and followed by acute exercise 16 weeks later. Antioxidant Antioxidants and Exercise group(AE) fed with antioxidants and followed by acute exercise. The activation of NF-κB binding activity was increased after exhaustive exercise bout in both group. In addition, pretreatment of ATO group with the antioxidants mixture lead to the inhibition of NF-κB binding activity. This results suggest that NF-κB activation should be further studies in response to a variety of exercise.

        A 13-year-old, castrated male, Maltese-dog was admitted to the Veterinary Medical Teaching Hospital of Chungnam National University with the history fo petechiae. On physical examination, she had petechiae and ecchymoses at her back, bilateral inguinal region, and inner part of ear pinna. CBC indicated neutrophilia and severe thrombocytopenia. A few platelets, no autoagglutination of RBC, and no spherocyte were observed. Coagulation profile demonstrated normal PT, and aPTT. No significant findings was not found on serum biochemical profile and electolytes test. FNA of a splenic mass which was found on the abdominal radiograph and sonograph demonstrated extramedullary hematopoiesis, Proteinuria was observed on urinarysis. Primary immune-mediated thrombocytopenia was diagnosed by ruling out of other underlying disease, severe thrombocytopenia, and response to immunosuppressive therapy. Prednisone and antibiotics were administered orally and parenterally, respectively. After altering prednisone to azathioprine, the number of platelets was recovered to normal range and petechiae and ecchymoses were improved.

        Ar과 N₂?가스가 혼합된 분위기에서 반응성 스퍼터링 방법에 의하여 TiN 박막을 증착하였다. N₂ 가스의 농도는 화학양론적으로 TiN이 형성되는 조건에 맞도록 조절하였으며, 기판의 온도는 실온에서 부터 700℃의 범위내로 유지하였다. (111) texture 구조를 가지면서 화학양론적으로 Ti??N??인 박막은 기판의 온도가 600℃ 이상에서 형성되었고, 기판의 온도가 600℃ 이하에서는 형성된 박막은 N-과다형이었다. XRD, XPS 및 RBS 분석 결과 TiN 박막의 조성비는 기판의 온도에서 다소 의존하였으나 약 5% 이내에 불과하였다. TiN 박막의 면저항은 기판온도의 증가에 따라 감소하였고, 기판온도가 600℃에서 증착된 TiN 박막의 면저항능 14.5Ω/□ 였고, Ar―가스 분위기에서 700℃로 30초간 열처리한 후는 8.9Ω/□ 이었다. 따라서 반응성 스퍼터링방법에 의하여 형성되는 양질의 TiN 박막은 기판온도가 600℃이상이 최적조건임을 알았다. Titanium nitride (TiN) films were prepared by reactive sputter deposition in mixed gas of Ar+N₂?. The volume percentage of N₂ in the working gas was chosen so as to grow stoichiometric TiN films and the substrate temperature during film growth was set from room temperature to 700℃. stoichiometric Ti??N?? films with (111) texture were grown at temperatures over 600℃, while films prepared at temperatures below 600℃ showed N-rich TiN. The composition x and y in the Ti??N?? films determined by XPS and RBS varied within 5% with the substrate temperature. The sheet resisstance of the TiN films decreases as the substrate temperature increased. TiN film prepared at 600℃ showed 14.5Ω/□, and it decreased to 8.9Ω/□ after the sample was annealed at 700℃, 30 sec in Ar-gas ambient by RTA. By far, high quality stoichiometric TiN films by reactive sputtering in the mixed gas ambient could be prepared at substrate temperature over 600℃.

        This paper concerns IF-stage and data recovery as a basic study for home products of GPS receiver. Using a transmission simulator, the same data can be received as transmission simulator transmits. Consequently, it is considered that IF-stage and the part of data recovery operate fairly good and attaching microprocessor control and time interval counter to this system will operate better. It can be expected that providing the system with RF-stage and antenna will be complete GPS receiver.

        FC-25계 회주철의 Boro-Nitriding 복합 표면처리에 따른 표면특성과 고온산화거동에 관한 연구

        Surface properties and oxidation behavior were studied for FC-25 gray cast iron (GCI), ion nitrided CCI (N CCI), powder boronized CCI (B GCI) and powder boronized N CCI (B-N GCI). A boride (Fe2B and FeB) layer with a tooth shape was formed by boronizing GCI at 650℃. The thickness and hardness of the layer were proportional to boronizing temperature and time. The hardness was decreased as the depth of the boronized layer increased. The activation energy for boride formation was about 74 kJ/mol. The thickness of the graphite layer formed by boronizing was increased by increasing temperature and time. A silicon layer insoluble to borides accumulated beneath the boride layer. The major phases were Fe4N, Fe3N and FeN in the nitride layer formed by ion nitriding. The thickness and hardness of the layer increased with increasing nitriding temperature and time under the Fe4N decomposition temperature of 680℃. The hardness was maximum at the surface and decreased with increasing nitrided depth on B-N GCI at 700℃, above the Fe4N decomposition temperature. Hardness of B-N GCI at the layer depth of 10 μm was 1000 Hv, which was lower than that of the B GCI at the same condition. Also the layer thickness was 50 gm; 10 μm deeper than that of B GCI. The reason was that the nitrogen gas given off by the decomposition made the layer porous and facilitated the diffusion of oxygen and other reactants. The samples were oxidated at 800℃ for 48hrs under atmospheric air. Oxidation rate was in decreasing order of N GCI, GCI, B GCI and B-N GCI. Oxidation rates were greatly influenced by the diffusion of nitrogen gas formed by Fe4N decomposition over the decomposition temperature of 680℃ and the oxidation barrier of the less corrosive boride layer.
