        설악산국립공원 멸종위기 산양(Naemorhedus caudatus) 개체군 크기와 서식지 이용 현황

        본 연구에서는 설악산국립공원에 서식하는 멸종위기종 산양과 서식지의 보전 및 관리를 위해 산양의 분변 및 카메라 트랩을 이용한 개체수와 서식지 이용 분석을 2010~2014년 동안 수행하였으며 생태학적 조사 방법(흔적조사, 카메라트 랩조사)을 사용하였다. 분변 조사와 카메라트랩핑 분석 결과 각각 166개체 및 251개체의 산양이 설악산국립공원에 서식하고 있는 것으로 나타났다. 흔적조사(분변)를 통한 서식지 이용 특성 분석에서 산양은 경사도 35°~60°, 고도 600~700m, 향 북동, 수계와의 거리 0~50m, 도로와의 거리 300~600m, 활엽수림을 가장 선호하는 것으로 나타났다. 카메라트랩 조사를 통한 산양 개체군 구성, 주간(07-18시)이 56.5%, 야간(18-07시)이 43.5%로 산양의 활동성을 파악하였다. 이러한 산양의 개체수와 서식지 이용 특성 분석은 향후 그들이 살아가는 서식지의 보전과 서식지의 관리를 위한 중요한 기초 자료로 활용될 수 있을 것이다. This study was conducted investigate population size and habitat use for the conservation and management of the endangered long-tailed goral in the Seoraksan National Park using feces and camera trap during 2010 to 2014 (track survey, camera trap). As a result of feces tracking and camera trap, its population size was estimated as 160 (camera trap)~251 (feces) individuals in the Seoraksan National Park. The goral prefer 35°~60° (slope), 600~700m (elevation), NE (aspect), 0~50m (distance to stream), 300~600m (distance to road) and bread-leaved forest (forest type) according to field tracking of fecal. Based on field camera trap, we estimated the age classes of goral populations and activity of gorals during day-time (07-18 time, 56.5%) and night-time (18-07 time, 43.5%). Such analyses of population size and habitat use of the goral could be applied as important fundamental data for conservation of gorals and management of their habitats.

        The cadmium contents of environment and human body were measured for the health status of the residents in the selected area near an old zinc-mining site assuming that cadmium contamination in the environment would affect the health status of the residents and the results were as follows: 1. The cadmium contents of the environment were 0.021±0.012 mg/m3 in the air. 0.019±0.011 mg/ml in the water, 0.382±0.233 mg/g in the rice and 0.342±0.211 mg/g in the tooth. 2. The major complaints of the residents were excessive sputum, dyspnea and pains on both extremities. F드믿 complained more frequently than male in dyspnea, lumbago and pains on both extremities. 3. An assessment of the nutritional health status of residents revealed that for male 8.5% were weak, 10.1% were fat and 32.5% were anemic, while for female 17.6% were weak, 15.3% were fat and 30.1% were anemic. 4. The prevalence rate of hypertension was 152.8?(male; 84.7?. female;200.0?) and the prevalence rate of diastolic hypertension was higher than that of systolic hypertension. 5. The diagnostic variables verifing the assumption mode in this study were blood pressure(0.76, p〈0.05) and E.S.R.(0.57, p〈0.01). From the above results, the environmental cadmium contamination affects the health status of the residents in the area near an old zinc-mining site to a certain extent, but not to a great extent(R2=0.55).

      • 유도지도자 운동지도행동의 체계적 관찰 분석

        The purpose of this paper is to observe and record Coaching behaviors of Judo Coaches and compare the difference between Judo Coaches by field. This study observed and recorded the coaching behaviors of 12 coaches (private hall 1 : 3, secondary school coach : 3, college coach : 3, national team : 3) complying with the standard of research subject and agreeing on the research intent of Judo coaches working for private hall, secondary and tertiary schools and national team located in Kyungin Region as of 2001 year by Coaching behaviors Observational Recording System (CBORS), and observational reliability coefficient was Scott's pl coefficient, over .90. As a in-depth interview to Judo Coaches' sports behaviors, the interview questionnaire used by Grossman (1990) and open type interview questionnaire made out based on precedented research was used. As for the collected data, percentage and frequency by the field of CBORDS coaching behaviors was compared by the field of Judo Coaches through systematic observational analysis, and the collected data through in-depth interview was analyzed using inductive category analysis. The authenticity of in-depth interview was confirmed by diverse analysis through members' review and expert groups' consultation. The observatory analysis and conclusion to Judo Coach Behaviors through the above method and procedure is as follows. First, there is difference in learning behaviors between Judo Coaches according to coaching field. National team coaches show significant difference to practicing time, middle and high school coaches show significant difference in coaching time, and college coaches show difference in operation time, and social sports coaches show significant difference in stand-by time. Second, there is difference in interactional behaviors among Judo Coaches according to coaching field. National team coaches was indicated to perform interactions by simplifying the experimented as individuals rather than diversified interactions at interactions with trainees. Third, there is difference in coaching behaviors among coaches by coaching field. At the coaching behavior of sub-areas of coaching behavior areas, social sports coaches was the highest in coaching time ratio, while there is any remarkable difference between national team and middle and high school coaches. At the demonstrating behavior of sub-areas of coaching behavior areas, national team coaches were the highest in coaching time ratio. At the social behavior of sub-areas of coaching behavior areas, social sports coaches was the highest in coaching time ratio. Middle and high school coaches were indicated to use trainee's voluntary depressible physical punishment for the effective progress of classes. And social sports coaches and national team coaches were indicated to use praise as social behavior. At the encouraging behavior of sub-areas of coaching behavior areas, college coaches were the highest. Especially, coaches with less experienced in coaching performed pressing behavior during coaching hour, but they were aware that coaching behaviors to be ultimately pursued are encouraging behaviors. At the technical feed-back behavior of sub-areas of coaching behavior areas, national team coaches were the highest. Al1 coaches provided positive feed-back for trainees, especially corrective feedback has been provided by national team coaches. At non-coaching behavior of sub-areas of coaching behavior areas, national team coaches were the lowest, and the absence status of coaches by outside figures during classes. suggests the need of more responsible and independent classes.

      • 안모유형에 따른 악관절융기와 전치의 경사도에 관한 연구

        The purpose of this study was to evaluate the difference and relationship between the slope of articular eminence and the inclination of upper and lower incisor teeth, which are related to the functional stability of occlusion in mandibular protrusion, according to Rickett's facial types, by the use of lateral cephalogram and SAM2 articulator in 68-adult normal occlusion without tooth missing, orthodontic treatment and occlusal equilibration. The results of this study were as follows. 1. The angle of articular eminence slope to occlusal plane in brachyfacial type was steeper than that in dolichofacial type, but the angle of articular eminence slope to SN plane and FH plane was not different between facial types. 2. The upper incisor axis in dolichofacial type was steeper than that in brachyfacial type, but lingual surface slope of upper incisor was not different between facial types. 3. In all samples there was a positive correlation between the angle of articular eminence slope and the lingual surface slope of upper incisor to SN plane and FH plane, and in mesofacial type there was a positive correlation between the angle of articular eminence slope and the lingual surface slope of upper incisor to SN plane, FH plane and occlusal plane, and in brachyfacial type there was a positive correlation between the angle of articular eminence slope and the lingual surface slope of upper incisor to occlusal plane. 4. In all samples there was a positive correlation between the angle of articular eminence slope and the angle of Dc-Gn to SN plane, and in mesofacial type there was a positive correlation between the angle of articular eminence slope and the angle of Dc-Gn to occlusal plane, and in brachyfacial type there was a positive correlation between the angle of articular eminence slope and the angle of Dc-Gn to SN plane and FH plane. 5. In all samples there was a positive correlation between the angle of Dc-Gn and the lingual surface slope of upper incisor to SN plane and FH plane, and in mesofacial type there was a positive correlation between the angle of Dc-Gn and the lingual surface slope of upper incisor to SN plane. 6. In all samples and facial types there was a negative correlation between MP to 1 axis and condylar incisal angle.

        Novel Trimeric Complex for Efficient Uptake of Plasmid Vector into HepG2 Cells

        Viral and :ion-viral vectors have been used in the delivery of genetic materials into animal cells and tissues, with each approach having pros and cons. Non-viral vectors have many useful merits such as easy preparation, low immunity and size tolerance of a transgene when compared to those of viral vectors. Delivery specificity may be achieved by complex formation between receptor ligands and a non-viral vector. In the present study, non-viral vector systems are investigated in an effort to find a practical delivery means for gene therapy, Receptor-ligand interaction between transferrin-receptor and transferrin was utilized for efficient gene transfer into cancer cells. A plasmid vector, pcDNA3 (LacZ) was ligated with a small duplexed oligo fragment in which a Biotin-VN^(TM) phosphoramidite was placed in the middle of the oligo. The plasmid vector labeled by biotin was then conjugated with biotin-labeled transferrin via streptavidin. This trimeric conjugates were delivered to a hepatoma cell line, HepG2. The delivery efficiency of the trimeric conjugate was 2-fold higher than that of cationic liposomes used for transfection of a plasmid vector. These results demonstrate that a plasmid vector can be efficiently transferred into cells by forming a trimeric complex of plasmid vector-linker-ligand.

        상온보관이 가능하며 사용이 편리한 DNA Size-Marker와 저분자량의 DNA 계측을 위한 Size Marker용 플라스미드 개발

        기존의 수입된 핵산 size markers는 dry ice에 넣어져서 운반되므로 매우 고가이다. 따라서 상온에서 보관될 수 있는 방법을 모색하던 중, 많은 물질의 경우 건조상태를 유지할 때 높은 안정성을 유지한다는 점에 착안했다. DNA는 매우 안정한 물질이므로 그 가능성은 더욱 높다고 가정했다. DNA size markers에 첨가되는 요소를 모두 혼합한 다음 동결건조를 시도해 보았다. 상온과 빛, 상온과 빛 차단 그리고 4℃와 빛 차단의 조건으로 60일간 방치한 후 건조 전 부피의 증류수로 건조체를 이용하여 전기영동 양상을 비교한 결과 DNA size markers의 양 및 질에서 큰 변화가 없었다. ΦX174 phage DNA는 double strand로 분기가 쉽지않으며 phage의 대량배양도 용이하지 않다. 이러한 문제점을 해결하기 위해 기존의 플라스미드를 변조하여 ΦX174 HaeIII 절단체와 유사한 DNA 분리 양상을 나타내도록 하였다. 개발된 새로운 플라스미드는 column chromatography를 사용하여 분리한 후 제한효소에 의해 효과적으로 절단되었다. 종합하면, 새로운 보관방법과 플라스미드의 개발은 국내에서 양질의 ΦX174 DNA size markers를 적정한 원가로서 개발할 수 있음을 나타낸다. Molecular biotechnology is a broad research and development principle encompassing basic research as well as applied industrial developments. Biotechnology is expected to become one of the most important research and development sectors in the next century, but the domestic support industry for the field is severely underdeveloped and at best in its infancy. DNA size marker is frequently employed during research in a purpose of extrapolating the relative size of a DNA fragment or measuring the amount of DNA loaded a sample. However, DNA size marker is not developed for commercialization domestically. Further, liquid suspension containing DNA size marker needs to be kept at a low temperature during storage and shipping. In an attempt to maintain the DNA samples at room temperature for extended period of time, lyophilization of DNA with addition of a nuclease inhibitor was studied. Hae Ⅲ digest of ΦX174 phage DNA has been a useful size marker for low molecualr weight DNA molecules, but the double strand RF form of the phage DNA requires laborious procedures for cell culture and purification. A new plasmid was constructed to generate band patterns which can substitute the Hae Ⅲ digest of ΦX174 phage DNA for a comparable purpose. Gel staining dye was also added to the lyophilized DNA to provide additional convenience such that DNA size marker was ready for use by simple reconstitution of distilled water. DNA size marker appears to be advantageous for development as one can judge the quality of the product by direct observation.

        Specific Gene Silencing by Single Stranded Large Circular Antisense Molecules

        I report that single-stranded antisense as a part of large circular (LC-) genomic DNA of recombinant M13 phage exhibits enhanced stability, sequence specific antisense activity, and no need for target site search. A cDNA fragment (708 bp) of rat TNF-α was inserted into a phagemid vector, and TNF-α antisense molecules (TNFα-LCAS) were produced as single-stranded circular DNA. When introduced into a rat monocyte/macrophage cell line. WRT7/P2, TNFα-LCAS was able to ablate LPS-induced TNF-α mRNA to completion. The antisense effect of TNFα-LCAS was shown to be sequence-specific because expressions of three control genes (β-actin. GAPDH and IL-1β) were not significantly altered by the antisense treatment. Further, TNFα-LCAS was found to be highly efficacious as only 0.1 ㎍ (0.24 nM) of TNFα-LCAS was sufficient to block TNF-α expression in 1×10^(5) WRT7/P2 cells. I have also observed specific antisense activity in reduction of NF-kB gene expression. The results suggest that an antisense sequence as a part of single-stranded circular genomic DNA has a specific antisense activity.

        재조합 아데노 바이러스 생산을 위한 효과적인 Shuttle 벡터의 개발과 검증

        Recombinant adenovirus is an attractive vehicle to deliver a foreign gene for the virus can infect many cell types including non-dividing cells. Production of a recombinant adenovirus requires co-transfection of a shuttle vector which harbors a gene of interest and a rescue vector. Adenoviral shuttle vectors currently in use contain limited numbers of cloning sites for insertion of a foreign gene. They also lack a promoter and a signaling sequence for the poly(A) tail. Therefore, we set out to construct two adenoviral shuttle vectors, pAd-YC and pAd-YC2, in an effort to facilitate cloning and expression of foreign genes. pAd-YC contains the CMV promoter and multiple cloning sites but without a poly(A) signal sequence. pAd-YC2 contains the CMV promoter, the multiple cloning sites and the poly(A) signal sequence. The functionality of pAd-YC and pAd-YC2 was tested by cloning and expression of a chimeric Fcγreceptor cDNA and the β-galactosidase gene. Fcγreceptor expression was confirmed by binding of opsonized erythrocytes to COS-1 infectants, and β-galactosidase expression was assayed for X-gal staining. The results demonstrate the functional efficacy of the two shuttle vectors. We have since successfully constructed several recombinant adenoviruses by employing the two versatile shuttle vectors.

      • 중부지역 옥수수 파종기에 따른 생육단계 및 수량성 변화

        Maize hybrids were planted on April 1, April 25, May 20 and June 15 in 2012, 2013 at the experimental paddy and upland fields in Suwon city , respectively to know changes of growth stage and yield according to different planting time. Maize planting on April 1 was not recommended because total growing days at planting on April 1 was not different with that at planting on April 25 due to many days from planting to emergence over 20 days at planting on April 1. Total growing days were not much different among three planting time after April 25 because days from silking to maturity was lengthened though days from planting to silking was shortened at later planting time. Stalk weight was decreased as planting time was delayed, particularly at planting times after May 20 at paddy field by excess water injury during early growth stage. But, grain yield was not low as in May and June. It is needed to introduce early maturity maize variety as Yanganok at late planting for higher grain yield and enough time for soil preparation for next winter crops

        DNA Chip using Single Stranded Large Circular DNA : Low Background and Stronger Signal Intensity

        Massive identification of differentially expressed patterns has been used as a tool to detect genes that are involved in disease related process. We employed circular single stranded sense molecules as probe DNA for a DNA chip. The circular single stranded DNAs derived from 1,152 unigene cDNA clones were purified in a high throughput mode from the culture supernatant of bacterial transformants containing recombinant phagemids and arrayed onto silanized slide glasses. The DNA chip was examined for its utility in detection of differential expression profile by using cDNA hybridization. Hybridization of the single stranded probe DNA were performed with Cy3- or Cy5-labeled target cDNA preparations at 60℃. Dot scanning performed with the hybridized slide showed 29 up-regulated and 6 down-regulated genes in a cancerous liver tissue when compared to those of adjacent noncancerous liver tissue. These results indicate that the circular single stranded sense molecules can be employed as probe DNA of arrays in order to obtain a precious panel of differentially expressed genes.
