        Antinociceptive Effect of Glyasperin F Isolated from Glycyrrhiza inflata in Mice

        Antinociceptive effect of glyasperin F isolated from Glycyrrhiza inflata extract (GIE) in ICR mice was studied. Oral administration of GIE (1-100 mg/kg) caused a dose-dependent reduction in acetic acid-induced writhing responses. To identify the active antinociceptive compound from the GIE, sub-fractions were obtained from the EtOAc layer of GIE by using a medium pressure liquid chromatography. From the sub-fractions obtained, the sub-fraction, which, when administered orally (10 mg/kg) showed an antinociceptive effect in both the writhing test and second phase of the formalin test was identified as glyasperin F using NMR and MS analyses. Finally, the antinociceptive effect of glyasperin F in mouse models of pain was confirmed. Orally administered glyasperin F (0.1-10mg/kg) showed a dose-dependent antinociceptive effect in both the writhing test and second phase of the formalin test. Taken together, glyasperin F isolated from the GIE may be used as a leading compound for further studies on pain and as a new drug derived from natural products for pain therapy.

        2,4-bis (p-hydroxyphenyl)-2-butenal (HPB242) induces apoptosis via modulating E7 expression and inhibition of PI3K/Akt pathway in SiHa human cervical cancer cells.

        <P>The Maillard reaction is a chemical reaction occurring between an amino acid and a reducing sugar, usually requiring thermal processing. Maillard reaction products (MRPs) have antioxidant, antimutagenic, and antibacterial effects, and although 2,4-bis (p-hydroxyphenyl)-2-butenal (HPB242), a fructose-tyrosine MRP, appears to inhibit proliferation of cancer cells, its mechanism of action has not been studied in detail. We found that HPB242 treatment modulated expression of cyclins and tumor suppressor genes in SiHa human cervical cancer cell lines: cyclins and phospho-pRB were downregulated, whereas the expression of CDK inhibitors and p53 was enhanced. HPB242 induced apoptosis dose-dependently by suppressing E7 expression and leading to sub-G1 cell-cycle arrest in SiHa cell lines; treatment also led to the proteolytic cleavage of caspase-3, -9, and poly (ADP-ribose) polymerase. Moreover, HPB242 upregulated Fas expression, altered expressions of pro- and antiapoptotic factors, and also inhibited nuclear translocation of nuclear factor κB and phosphorylation of IκB. HPB242 treatment decreased phosphatidyl inositol-3 kinase and p-Akt expression levels, demonstrating that this survival pathway may also be inhibited by HPB242. Cumulatively, HPB242 promotes apoptosis by influencing E7 expression, inducing cell-cycle arrest at sub-G1 phase, and promoting both intrinsic (mitochondrial) and extrinsic (Fas-dependent) apoptosis in SiHa human cervical cancer cells.</P>

      • Purge & Trap-GC를 이용한 의약품 필름코팅 정제 중 잔류용제에 관한 연구

        의약품은 약물을 생체에 적풋하기 위하여 유효성분의 효과가 언제나 일정하게 확보되고 사응에 편리하도록 만들어지는 것이므로 유효썽분 이외에 약효에 영향을 주지 않는 성분이 첨가되는 경운가 많다. 이 때 사용되는 용매들은 제피의 광택 및 건쪼시간의 단축 등을 위하여 휘발점이 낮을 용매들이 주로 사용되어진다. 본 연구는 의약품 필름코팅정제 중 잔류용매 4종(chlorofonr benzen, trichloro ethylen, 1,4-dioxane)에 대한 변형된 pirge & trap-GC 장치를 이용한 동시분석방법을 개발하였으며, 각 표준품의 RSD 값은 chloroform 3.03%, benzen 3.17%, trichloroethylen 3.69% and 1,4-dioxane 3.41%였다. 또한 시중 유통중인 의약품 50종에 대하여 잔류웅매 양을 측정하였으며, 검출되는 잔류용매는 한 건도 없었다. This study nras carried out to develope the analytical method for the mixture of chlorefonn, benzen, trichloroethylen and 1,4-dioxane simultaneously and determine the remainingorgauic solvents in coating tablets by Purge & Trap-GC. The results were as follouFs ; 1. Chloroform, benzen, trio:tloroethylen and 1,4-dioxane separated by tenax #5 trap by HP-624GC column by terrlperature programming. The peaks were separated completely at retentiontime of 6.88min for chloroform, 8.21min for benzen, 10.38miu for trichloroethylen and 11.95minfor 1,4-dioxane. 2. Standard RSD were individually chloroform 3.03%, benzen 3.17%, trichloroethylen 3.69%and 1,4-diorane 3.41%. 3. 60 samples were not detrcted chloroform, benzen, trichloroethylen and 1,4-dioxane.

      • 철도시스템분야 실무적용을 위한 RAM Guidance 개발에 관한 연구

        철도시스템 기술 고도화 및 복잡성의 증대와 더불어 발주기관으로부터 시스템의 성능보증과 안전성 확보가 요구되고 있는 실정이다. 또한 국내 철도안전법 강화로 인하여 시스템 보증활동의 중요성이 증가되고 있다. 본 논문에서는 국제철도관련 규격을 바탕으로 철도시스템의 RAM 업무에 대한 절차를 정의하고, 해당 철도 프로젝트에서 RAM 업무를 원활히 수행할 수 있도록 관련된 국제 규격 및 유사시스템 적용 사례에 기반하여 철도시스템 개발 특성을 고려한 RAM 업무 수행 지침(Guidance)을 개발하였다. Performance and safety assurances for the railway system are required by the client because of the high degree of technology and increased complexity. In addition, as a result of reinforced domestic railway safety laws, the importance of system assurance activity has been growing steadily. This paper defines the task of the RAM process based on international railway industry standards for RAMS engineers to work efficiently and develops the RAM guidance considering the railway system characteristics based on domestic standards and application cases of railway systems.

      • 서울의 Penicillinase Producing Neisseria Gonorrhoeae 발생빈도(1994)

        Background : In recent years gonorrhoea has been panendemic and remains one of the most common Sexually Transmitted Diseases in the world, especially in developing countries. Objective & Method : For the detection of a more effective therapeutic regimen and assessing the prevalence of Penicillinase Producing Neisseria Gonorrhoeae(PPNG), Ive have been trying to study the patients who have visited the VD Clinic of Choong-Ku Public Health Center in Seoul since 1980 by means of the chromogenic cephalosporin method. Results : In 1994, 168 strains of N. gonorrhoeae were isolated, among which 109 (64,9% ) were PPNG. Conclustion : Our results suggests that the prevalence of PPNG in Seoul is still increasing.

      • High-throughput screening of protein variations in soybean seeds.

        The protein in soybean seeds accounts for approximately 40% of the dry seed weight. Two major storage proteins, 7S and 11S, constitute 70-80% of the total storage proteins in the seeds. In this study, the variation of total soluble protein extracts from 1152 soybean landraces that have been collected from South Korea were studied using high-throughput screening method with HT Protein Express Labchip (Caliper Life Sciences, Inc.). Seven distinct protein band patterns - four protein sub-units of 11S and three sub-units of 7S, were taken into account and their presence or absence were analyzed. Among the 1152 landraces, 525 genotypes were identified as lacking lipoxygenase, 255 lacking α1 subunit, 680 lacking α subunit, 169 lacking β subunit, 140 lacking acidic, 114 lacking Kunitz Trypsin Inhibitor (KTi) and 199 lacking basic protein patterns. The high-throughput protein analysis is helpful in screening a large number of populations with less time and minimum labor. The selected genotypes with low amounts or lacking of anti-nutritional factors such as trypsin inhibitor, lipoxygenase and α subunit would be used for future breeding purpose of quality improvement in soybean protein.

        Among the hematologic complications of solid tumors, leukoerythroblastosis, DIO and MAHA have been known to be clinically significant According to the recent increasing incidence of cancer, these hematologic complications have been reported frequently. However, the simutaneous developement of such manifestations is rare. Recently, we have experienced a patient with gastric adenocarcinoma in which case severe bleeding tendency were observed and laboratory studies including bone marrow examination demonstrated the findings of leukoerythroblastosis, DIC and MAHA. She died of upper gastrointestinal bleeding despite the supportive care such as vlood transfusions. We should give attention to the findings of peripheral golld smear for the patient with malignancy and it is mandatory for therapeutic plan to perform bone marrow examination if the above mentioned hemorrhagic complications are suspected.

        당일 라식, 라섹 수술과 기존 수술법의 수술 후 결과 비교

        Purpose: To evaluate the postoperative results of one day laser-assisted in-situ keratomileusis (LASIK) or laser-assisted sub-epithelial keratectomy (LASEK) procedures, which were performed on the same day as preoperative examinations, including fundus examinations after dilating the pupil. Methods: This study included 226 LASIK patients (226 eyes) and 201 LASEK patients (201 eyes) who underwent surgery with Visumax and EX500 from January to December in 2016. We divided the patients into two groups. The one-day surgery group (one-day group) underwent surgery on the same day as preoperative examinations, including dilated fundus examinations. The scheduled surgery group (scheduled group) underwent surgery on the scheduled day after the preoperative examinations. In the one-day group, the surgery was usually performed 2–5 hours after instillation of the pupil dilating eye drops. Results: Among LASIK patients, the one-day group included 109 patients and the scheduled group included 117 patients. The postoperative myopic errors were 0.06 ± 0.37 diopters (D) and 0.07 ± 0.36 D, respectively (p = 0.91). The postoperative astigmatism was -0.38 ± 0.24 D and -0.37 ± 0.24 D, respectively (p = 0.77). The postoperative uncorrected visual acuity was -0.05 ± 0.03 logMAR and -0.06 ± 0.03 logMAR, respectively (p = 0.13). Among LASEK patients, the one-day group included 107 patients and the scheduled group included 94 patients. The postoperative myopic error was 0.18 ± 0.52 D and 0.22 ± 0.54 D, respectively (p = 0.95). The postoperative astigmatism was -0.48 ± 0.30 D and -0.46 ± 0.29 D, respectively (p = 0.14). The postoperative uncorrected visual acuity was -0.05 ± 0.03 logMAR and -0.06 ± 0.03 logMAR, respectively (p = 0.33). Conclusions: The postoperative results of the one-day LASIK and LASEK patients, whose surgery was performed on the same day as the preoperative examinations, were not significantly different from those using the conventional method.         수돗물 불소화지역과 비불소화 지역의 공직자 및 일반 주민의 수돗물 불소화사업에 대한 인식도 연구

        For the purpose of establishing basic data for developing approariate strategies to resolve problems of community water fluoridation decision making, this study was conducted to detect the knowledge and attitudes about community water fluoridation among Koreans. In this survey, the questionnaires were used to personally interview 1,562 citizens in 14 fluoridated areas and 18 non-fluoridated areas and 420 public offcials with postage in 30 fluoridated areas and 40 non-fluoridated areas. This survey areas and persons were collected by multiple stratified systematic and proportional sampling. In this survey, the questionnaires of 1,539 citizens and of 275 public officials were analysed by SAS program. Overall, 44.2 percent of citizens had heard about community water fluoridation, while 99.6 percent of public officials had heard about community water fluoridation. Only 32.9 percent of the respondents of citizens had precise information on the purpose of community water fluoridation, but for public officials, that was 96.0 percent. 54.6 percent of the respondents of citizens who knew about community water fluoridation learned the information from mass media (television and newspater etc.). 53.5 percent of the respondents of citizens hope to implement and to continue commnuity water fluoridation, while 71.6 percent of public officials hoped to implement and to continue water fluoridation. Based on these finding it is recommended that the education and promotion activities should be done on the purpose, the implementation or not and the meaning of community water fluoridation through mass media.
