        Analysis of Lignans in Acanthopanax sessiliflorus Fruits and Their Fermented Wine by HPLC

        High performance liquid chromatography (HPLC) was used for the determination of lignans, eleutherosides B and E, in Acanthopanax sessiliflorus fruits and their fermented wine. The lignans were quantified by a reversed-phase system using a gradient of H2O and acetonitrile as a mobile phase within 20 min. The analysis was successfully carried out within 20 min. The contents of eleutherosides Band E as main active principles of Acanthopanax species were measured in A. sessiliflorus fruits (1.15 and 8.49 μg/mg, respectively), their fermented wine (0.45 and 1.33 μg/mg, respectively) and wine residues (no detection).

        Caries Prevention by Use of the Computer Program on Diet Control for Adolescent

        Objective: In order to utilize dietary control, one of ways to prevent dental caries, more easily, the sugar content and viscosity of food were measured to obtain cariogenic index. Using this index, this study is to contribute to having proper eating habits through improvement of eating habits and to the prevention of dental caries through the use of computation program for dietary control. Methods: A 5-day dietary log from 105 students in the first year of high school, who are residing in Cheonan, and the result of survey was entered into the existing computation program for dietary control to examine calorie intake, 3 major nutrients and deficiency of clean food intake count, cariogenic index; S-PHP index and Snyder test were performed to examine the correlation of each category with oral health. Results: The ratio of average daily intake of 3 major nutrients was 62.4:19.9:17.7 respectively, and the average daily intake count of clean food was 2.44±1.71, which was appropriate. The average cariogenic index, 152.89±65.45 min. Since cariogenic index (r=−0.535), S-PHP index (r=−0.338) and Snyder test (r=−0.365) decreased as clean food intake count increased, they showed a relatively high correlation (p<0.01). Conclusion: Computation program for dietary control enables analyzing adolescents’ dietary habits by category and leads them to have proper eating habits through improvements in eating habits, which will result in dental caries prevention and oral health improvements; therefore, utilization of the program for preventive care in clinical environment is recommended.

        폐 젤리충진 통신케이블로부터 고순도 구리회수를 위한 대형화 방안 연구

        폐 젤리충진 통신케이블로부터 고순도 구리회수를 위하여 실리콘계 및 톨루엔계 합성 열매유와 폐식용유를 사용하여 적용성을 평가하였다. 실리콘계 오일의 경우 모든 조건에서 분리효율이 낮은 반면, 톨루엔계 오일 및 폐식용유의 경우 처리온도 및 시간을 조절함에 따라 높은 분리효율을 얻을 수 있었다. 폐식용유를 사용하는 50 kg급 밀폐형 대용량 설비를 이용하여 300 ℃에서 60 min 이상 처리할 경우 99.2% 이상의 고순도 구리와이어를 100% 회수할 수 있었다. We examined and compared the feasibility of vegetable oils with synthetic thermal conductive oils to recover highly purified copper metal from waste jelly-filled communication cables. While polydimethylsiloxane shows relatively poor separation effi-ciency under entire operating conditions, dibenzyltoluene and waste vegetable oil show the high separation efficiency if the appropriate operating temperature and time were given. By running 50 kg-class equipment with waste vegetable oils, we ob-tained 100% copper metal recovery with 99.2% purity at 300 ℃ for 60 min.

      • 피프로닐 오염도 조사를 위한 분석법 정립

        2017년 유럽에서 피프로닐에 오염된 계란이 유통된데 이어 국내에서 생산된 일부 계란에서도 살충제가 검출되었다. 피프로닐은 진드기 방제 목적으로 사용되는 살충제로 sulfide, sulfone, amide, sulfoxide의 대사산물을 생성한다. 피프로닐은 물에 1.9 mg/L 정도 용해되고, 아세톤에 545.9g/L가 용해되는 성질이 있어 시료 채취시 아세톤을 사용한다. 농장시설 등에 부착된 피프로닐을 제거하기 위해 5% 탄산나트륨(Na2CO3)과 15% 과산화수소(H2O2)를 권장하고 있다. 이는 피프로닐을 피프로닐설폰으로 산화시킨 다음 반복적으로 사용하여 제거한다. 이러한 제거 작업을 통해 피프로닐이 제거된 정도를 조사할 필요가 있고, 이를 위한 분석법 정립이 필요하다. 시료를 250 mL 용기에 넣고 1% 함유 아세토니트릴 100 mL을 주가하여 10분간 격렬하게 진탕한 후, 3500 rpm으로 원심분리를 하였다. 추출액 1 mL을 0.2 um 멤브레인필터로 여과하여 시험용액으로 하였다. 칼럼은 C18(2.0 mm I.d×50mm, 3 um)을 사용하였고, 이동상은 0.1% 포름산 함유 메탄올과 0.1% 포름산 및 5 mM 아세트산암모늄이 함유된 물을 사용하였다. 분석조건으로 피프로닐의 선구이온은 437 m/z, 생성이온은 368, 255, 290 m/z, positive 조건으로 분석하였고, 피프로닐 설폰의 선구이온은 451 m/z, 생성이온은 415, 282, 244 m/z negative 조건으로 분석하였다. 분석법의 정량한계는 피프로닐 0.92 ug/kg, 피프로닐설폰 0.70 ug/kg이었다. 피프로닐과 피프로닐 설폰을 각각 1, 5, 10, 50, 100 ug/kg 수준으로 검량선을 작성한 결과, r2는 모두 0.9999이었다. 10 ug/kg과 50 ug/kg 수준의 3반복 회수율 실험결과, 10ug/kg 수준에서 피프로닐은 74.7±0.56%, 피프로닐 설폰은 88.9± 0.27%로 나타났다. 50 ug/kg 수준에서는 피프로닐은 78.6±0.11%, 피프로닐 설폰은 95.2±0.02%로 유효회수율 범위(70~120%)에 포함되었다. 머무름 시간은 피프로닐 5.3 mim, 피프로닐 설폰은 5.7min에서 검출되었다. 교차확인 목적으로 LC/MS/MS로 분석한 시료를 GC/μ-ECD로 분석을 하였다. 분석조건은 주입구 온도 250℃, (5%-Phenyl)-methylpolysiloxane이 충진된 30 m×0.25 mm×0.25 um길이의 칼럼이다. 오븐온도는 130℃에서 300℃까지 승온 조건으로 분석하였고, 머무름시간은 피프로닐 11.64 min, 피프로닐 설폰12.79 min으로 LC/MS/MS와 GC/μ-ECD 모두 표준물질과 시료의 결과가 일치함을 알 수 있었다. 정립된 분석법은 피프로닐 오염도조사를 위한 분석법으로 활용할 수 있을 것으로 판단된다.

      • 포도 추출물들의 자유 라디칼 소거 작용 및 마우스 대식세포주의 염증 발현 매개 인자들에 대한 생성 억제 효과

        국내에서 생산된 거봉 및 캠벨 포도의 씨, 줄기 및 껍질추출물에 대하여 항산화 작용, 염증 관련 인자 생성에 미치는 활성 및 암세포 성장에 대한 영향 등을 resveratrol과 비교하여 평가하였다. 그 결과 포도 추출물 중 거봉줄기, 캠벨줄기, 캠벨씨 및 거봉씨 추출물들이 항산화 능력을 나타내었고 그 중 거봉씨 추출물은 vitamin C와 효력이 유사하게 나타나 항산화 효능이 우수함을 알 수 있었다. 또한 마우스 대식세포주인 RAW 264.7 cell을 이용하여 포도 추출물들의 LPS 처리에 의한 PGE₂ 및 NO 생성을 저해 여부를 확인한 결과, 거봉줄기, 거봉씨, 및 캠벨씨 추출물이 50 ㎍/mL에서 PGE₂ 및 NO 생성을 50% 가량 저해하는 효능을 나타내었다. 또한 사람 폐암 및 대장암 세포주를 이용하여 포도 추출물들이 암세포 성장 저해 효과를 나타내는지를 확인하였는데 거봉줄기 및 씨 추출물 50㎍/mL에서 30% 정도의 암세포 성장 저해 작용을 나타내었다. Antioxidant and anti-inflammatory potentials of various grape extracts were evaluated. Extracts from Kyho seed, Kyho stem, and Campbell seed showed potent l,l-diphenyl-2-picrylhydrazyl (DPPH.) free radical scavenging activities compared to resveratrol (IC_(50) 16.9, 21.5, 21.9, 34.6 ug/mL, respectively), among which, antioxidant effect of Kyho seed extract were similar to that of vitamin C (IC_(50)=12.2㎍/mL). These extracts also exhibited inhibitory activities on lipopolysaccharide (LPS)-induced prostaglandin E₂ production and nitrite formation in mouse macrophage RAW 264.7 cells at 50㎍/mL. Kyho stem and seed extracts showed growth inhibitory activities in human lung and colon cancer cells. These results suggest the potential roles of grape extracts as antioxidants and anti-inflammatory agents.

      • 딸기 와인 개발을 위한 최적 발효 조건

        Optimal fermentation condition for development of new strawberry wine were investigated. Commercial yeast, Fermivin was good for brewing of strawberry wine and its optimal addition concentration was also 1%. Maximal ethanol content of strawberry wine was obtained when raw strawberry juice(24 brix) was fermented by 1% Fermivin at 25℃ for 7 days and its antioxidant activity was 78.5%. 새로운 딸기 와인을 개발하기 위한 최적 발효조건을 연구하였다. 시판효모인 페미빈이 딸기와인 제조에 제일 좋았고 1%가 최적 첨가 농도이었다. 딸기와인의 최대 에탄올 생성량은 생딸기주스 (24brix)를 1% 페미빈으로 25℃에서 7일 발효시켰을 때 얻어졌고 이 발효액의 항산화활성은 78.5% 이었다.

        Fimasartan attenuates renal ischemia-reperfusion injury by modulating inflammation-related apoptosis

        Fimasartan, a new angiotensin II receptor antagonist, reduces myocyte damage and stabilizes atherosclerotic plaque through its anti-inflammatory effect in animal studies. We investigated the protective effects of pretreatment with fimasartan on ischemia-reperfusion injury (IRI) in a mouse model of ischemic renal damage. C57BL/6 mice were pretreated with or without 5 (IR-F5) or 10 (IR-F10) mg/kg/day fimasartan for 3 days. Renal ischemia was induced by clamping bilateral renal vascular pedicles for 30 min. Histology, pro-inflammatory cytokines, and apoptosis assays were evaluated 24 h after IRI. Compared to the untreated group, blood urea nitrogen and serum creatinine levels were significantly lower in the IR-F10 group. IR-F10 kidneys showed less tubular necrosis and interstitial fibrosis than untreated kidneys. The expression of F4/80, a macrophage infiltration marker, and tumor necrosis factor $(TNF)-{\alpha}$, decreased in the IR-F10 group. High-dose fimasartan treatment attenuated the upregulation of $TNF-{\alpha}$, interleukin $(IL)-1{\beta}$, and IL-6 in ischemic kidneys. Fewer TUNEL positive cells were observed in IR-F10 compared to control mice. Fimasartan caused a significant decrease in caspase-3 activity and the level of Bax, and increased the Bcl-2 level. Fimasartan preserved renal function and tubular architecture from IRI in a mouse ischemic renal injury model. Fimasartan also attenuated upregulation of inflammatory cytokines and decreased apoptosis of renal tubular cells. Our results suggest that fimasartan inhibited the process of tubular injury by preventing apoptosis induced by the inflammatory pathway.

        Whole-genome resequencing analysis of 20 Micro-pigs

        Background Miniature pigs have been increasingly used as mammalian model animals for biomedical research because of their similarity to human beings in terms of their metabolic features and proportional organ sizes. However, despite their importance, there is a severe lack of genome-wide studies on miniature pigs. Objective In this study, we performed whole-genome sequencing analysis of 20 Micro-pigs obtained from Medi Kinetics to elucidate their genomic characteristics. Results Approximately 595 gigabase pairs (Gb) of sequence reads were generated to be mapped to the swine reference genome assembly (Sus scrofa 10.2); on average, the sequence reads covered 99.15% of the reference genome at an average of 9.6-fold coverage. We detected a total of 19,518,548 SNPs, of which 8.7% were found to be novel. With further annotation of all of the SNPs, we retrieved 144,507 nonsynonymous SNPs (nsSNPs); of these, 5968 were found in all 20 individuals used in this study. SIFT prediction for these SNPs identified that 812 nsSNPs in 402 genes were deleterious. Among these 402 genes, we identified some genes that could potentially affect traits of interest in Micro-pigs, such as RHEB and FRAS1. Furthermore, we performed runs of homozygosity analysis to locate potential selection signatures in the genome, detecting several loci that might be involved in phenotypic characteristics in Micro-pigs, such as MSTN, GDF5, and GDF11. Conclusion In this study, we identified numerous nsSNPs that could be used as candidate genetic markers with involvement in traits of interest. Furthermore, we detected putative selection footprints that might be associated with recent selection applied to miniature pigs.
