        강제환우와 보존기간이 난질에 미치는 영향

        An experiment was conducted to investigate the effects of forced molting and egg storage time on the various egg qualities. A total of 240 ISA Brown layers (60 wk of age) were employed as the unmolted treatment (Control). Two hundred and forty ISA Brown layers, molted at the age of 55 wk, were used as a forced molting treatment (T1), and the same number and strain of layers, molted at the age of 70 wk. were also used as the another forced molting treatment (T2). A total of 120 eggs were sampled from each treatment, and divided into six sets, 20 eggs per set. These six sets were stored for 1, 3, 6, 9, 12, and 15 days at 18℃ temperature, respectively. Eggs from T1 were collected from laying hens at the age of 68 wk, which started molting at 60 wk of age and achieved 50% egg production at 63 wk of age. Eggs from T2 were collected from hens at 82 wk of age, which started molting at 70 wk of age and achieved 50% egg production at 78 wk of age. The eggshell strength of T1 was significantly (p<0.05) higher than the Control and T2, and the storing periods did not affect the eggshell strength at all. Neither the forced molting nor the storing periods did not exert any consistent effect on the egg weight, eggshell thickness, eggshell color and egg yolk color. The albumin heights of T1 and T2 were significantly (p<0.05) lower than the Control, and it was remarkably reduced gradually as the storage periods increased in all three treatments. The Haugh unit showed very similar trends as the albumin height, indicating that both albumin height and Haugh unit were very much related to each other. In conclusion, the forced molting improves the eggshell strength, but decreases the albumin height and Haugh unit. The storage of eggs also decreases the albumin height and Haugh unit regardless of molting.

        A Rapid and Simple Method for DNA Preparation of Magnaporthe oryzae from Single Rice Blast Lesions for PCR-Based Molecular Analysis

        Rice blast is one of the most destructive diseases of rice worldwide, and the causative agent is the filamentous ascomycete Magnaporthe oryzae. With the successful cloning of more and more avirulence genes from M. oryzae, the direct extraction of M. oryzae genomic DNA from infected rice tissue would be useful alternative for rapid monitoring of changes of avirulence genes without isolation and cultivation of the pathogen. In this study, a fast, low-cost and reliable method for DNA preparation of M. oryzae from a small piece of infected single rice leaf or neck lesion was established. This single step method only required 10 min for DNA preparation and conventional chemical reagents commonly found in the laboratory. The AvrPik and AvrPi9 genes were successfully amplified with the prepared DNA. The expected DNA fragments from 570 bp to 1,139 bp could be amplified even three months after DNA preparation. This method was also suitable for DNA preparation from M. oryzae strains stored on the filter paper. All together these results indicate that the DNA preparation method established in this study is reliable, and could meet the basic needs for polymerase chain reaction-based analysis of M. oryzae.

      • Multiple Sexual Partners as a Potential Independent Risk Factor for Cervical Cancer: a Meta-analysis of Epidemiological Studies

        It's known that having multiple sexual partners is one of the risk factors of human papillomavirus (HPV) infection which is a major cause of cervical cancer. However, it is not clear whether the number of sexual partners is an independent risk factor for cervical cancer. We identified relevant studies by searching the databases of MEDLINE, PubMed and ScienceDirect published in English from January 1980 to January 2014. We analyzed those studies by combining the study-specific odds ratios (ORs) using random-effects models. Forty-one studies were included in this meta-analysis. We observed that the number of sexual partners was associated with the occurrence of non-malignant cervical disease (OR=1.82, 95%CI 1.63-2.00) and invasive cervical carcinoma (OR=1.77, 95%CI 1.50-2.05). Subgroup analyses revealed that the association remained significant after controlling for HPV infection (OR=1.52, 95%CI 1.21-1.83 for non-malignant disease; OR=1.53, 95%CI 1.30-1.76 for invasive cervical carcinoma). We found that there was a non-linear relation of the number of sexual partners with both non-malignant cervical disease and invasive cervical carcinoma. The risk of both malignant and non-malignant disease is relatively stable in women with more than 4-7 sexual partners. Furthermore, the frequency-risk of disease remained significant after controlling for HPV infection.The study suggested that h aving multiple sexual partners, with or without HPV infection, is a potential risk factor of cervical cancer.

        Silencing of long noncoding RNA PVT1 inhibits podocyte damage and apoptosis in diabetic nephropathy by upregulating FOXA1

        The number of patients with diabetic nephropathy (DN) is still on the rise worldwide, and this requires the development of new therapeutic strategies. Recent reports have highlighted genetic factors in the treatment of DN. Herein, we aimed to study the roles of long noncoding RNA (lncRNA) plasmacytoma variant translocation 1 (PVT1) and histone 3 lysine 27 trimethylation (H3K27me3) in DN. A model of DN was established by inducing diabetes in mice with streptozotocin. Mouse podocyte clone 5 (MPC5) podocytes and primary podocytes were cultured in normal and high glucose media to observe cell morphology and to quantify PVT1 expression. The roles of PVT1 and enhancer of zeste homolog 2 (EZH2) were validated via loss-of-function and gain-of-function in vitro experiments to identify the interactions among PVT1, EZH2, and forkhead box A1 (FOXA1). The podocyte damage and apoptosis due to PVT1 and FOXA1 were verified with in vivo experiments. PVT1 was highly expressed in MPC5 and primary podocytes in DN patients and in cultures grown in high glucose medium. A large number of CpG (C-phosphate-G) island sites were predicted at the FOXA1 promoter region, where PVT1 recruited EZH2 to promote the recruitment of H3K27me3. The silencing of PVT1 or the overexpression of FOXA1 relieved the damage and inhibited the apoptosis of podocytes in DN, as was evidenced by the upregulated expression of synaptopodin and podocin, higher expression of Bcl-2, and lower expression of Bax and cleaved caspase-3. The key findings of this study collectively indicate that the suppression of lncRNA PVT1 exerts inhibitory effects on podocyte damage and apoptosis via FOXA1 in DN, which is of clinical significance.

      • MicroRNA Expression Profile Analysis Reveals Diagnostic Biomarker for Human Prostate Cancer

        Prostate cancer is a highly prevalent disease in older men of the western world. MicroRNAs (miRNAs) are small RNA molecules that regulate gene expression via posttranscriptional inhibition of protein synthesis. To identify the diagnostic potential of miRNAs in prostate cancer, we downloaded the miRNA expression profile of prostate cancer from the GEO database and analysed the differentially expressed miRNAs (DE-miRNAs) in prostate cancerous tissue compared to non-cancerous tissue. Then, the targets of these DE-miRNAs were extracted from the database and mapped to the STRING and KEGG databases for network construction and pathway enrichment analysis. We identified a total of 16 miRNAs that showed a significant differential expression in cancer samples. A total of 9 target genes corresponding to 3 DE-miRNAs were obtained. After network and pathway enrichment analysis, we finally demonstrated that miR-20 appears to play an important role in the regulation of prostate cancer onset. MiR-20 as single biomarker or in combination could be useful in the diagnosis of prostate cancer. We anticipate our study could provide the groundwork for further experiments.

        Influence of Kluyveromyces marxianus on proteins, peptides, and amino acids in Lactobacillus-fermented milk

        With increasing application of yeast in fermented milk, in order to study the effect of yeast on milk protein during the fermentation process, the effects of the presence of Kluyveromyces marxianus in milk fermented by Streptococcus thermophilus and Lactobacillus bulgaricus were investigated. After fermentation, the amino acid, protein, and peptide contents were analyzed by ultra-performance liquid chromatography, two-dimensional gel electrophoresis, and liquid chromatography–mass spectrometry, respectively. After the addition of K. marxianus for fermentation, 25 protein spots changed significantly. These were mostly caseins and bovine serum proteins, and the content of total free amino acids increased by 16.30%; ten types of bioactive peptides were identified. Furthermore, the number of peptide types in milk fermented by K. marxianus increased significantly compared with milk fermented by Lactobacillus. K. marxianus is considered to promote proteometabolism in milk when added with Lactobacillus, generate flavor compounds, and improve the digestion and absorption character of milk.

        β-Xylosidase and β-mannosidase in combination improved growth performance and altered microbial profiles in weanling pigs fed a corn-soybean meal-based diet

        Objective: In this study, two glycosidases (XMosidases), β-xylosidase and β-mannosidase, were investigated on their in vitro hydrolysis activities of feed and on the improvement of growth performance in vivo in weanling pigs. Methods: Enzyme activities of XMosidases in vitro were evaluated in test tubes and simulation of gastric and small intestinal digestion, respectively, in the presence of NSPase. In vivo study was performed in 108 weaned piglets in a 28-d treatment. Pigs were allotted to one of three dietary treatments with six replicate pens in each treatment. The three treatment groups were as follows: i) Control (basal diet); ii) CE (basal diets+CE); iii) CE-Xmosidases (basal diets+ CE+β-xylosidase at 800 U/kg and β-mannosidase at 40 U/kg). CE was complex enzymes (amylase, protease, xylanase, and mannanase). Results: In vitro XMosidases displayed significant activities on hydrolysis of corn and soybean meal in the presence of non-starch polysaccharide degrading enzymes (xylanase and β-mannanase). In vitro simulation of gastric and small intestinal digestion by XMosidases showed XMosidases achieved 67.89%±0.22% of dry matter digestibility and 63.12%±0.21% of energy digestibility at 40°C for 5 hrs. In weanling pigs, additional XMosidases to CE in feed improved average daily gain, feed conversion rate (p<0.05), and apparent total tract digestibility of crude protein (p = 0.01) and dry matter (p = 0.02). XMosidases also altered the gut bacterial diversity and composition by increasing the proportion of beneficial bacteria. Conclusion: Addition of a complex enzyme supplementation (contained xylanase, β-mannanase, protease and amylase), XMosidases (β-xylosidase and β-mannosidase) can further improve the growth performance and nutrient digestion of young pigs.

        Centrifuge Model Tests and Numerical Simulations of the Landslide Evolution Process

        Centrifuge model tests and numerical simulations were performed to study the landslide evolution process and failure mechanism. A TLJ-500 geotechnical centrifuge was used for the experiments and landslide deformation and stress was monitored using high-precision differential displacement sensors and earth pressure micro-sensors. Discrete element numerical simulations were performed using PFC2D based on the experimental results. The findings show that the landslide evolution process can be divided into three stages: 1) compaction and consolidation; 2) uniform deformation; and 3) accelerated deformation and failure. The numerical simulation results verify the distinct stage characteristics of the landslide evolution process. According to the migration of microscopic soil mass particles within the landslide, stage 3) can be further divided into a deformation development stage and instability and failure stage. The simulation displacement monitoring curves and displacement map show distinct deformation characteristics and displacement indicators from stages 2) to 3) and from the deformation development stage to the instability and failure stage. The experimental and numerical results reveal the landslide failure mechanism: the upper part of the landslide thrusts and slides; the middle part squeezes; the lower part collapses; and shear plane penetration leads to landslide failure.
