Suppression of the Ly6 Antigens Expression on P815 Mastocytoma Cells by Expressing Antisense RNA

Ly6항원은 T세포, B세포, 대식세포와 같은 여러 면역기능을 담당하는 세포의 표면에 g1ycosy1 phosphatidylinositol결합형태로 발현된다. 활성화된 대식세포와 T 림파구는 보다 증가된 Ly6항원의 발현을 보여준다. 여러 다른 면역세포에서 이 항원의 정확한 기능을 연구하기 위하여, antisense RNA 기법을 이용하여 Ly6항원의 발현이 억제된 세포를 얻고자 시도하였다. DBA/2생쥐(H-2^(d))에서 기원된 mastocytoma인 P8l5세포를 pNeoSRαⅡ-Ly6(-)백터, 또는 pFRSVSRα-Ly6(-)와 pSV2neo plasmid DNA로 유전자 전달을 행하였다. 15개의 G4l8에 대한 저항성을 갖는 clone을 얻었으며, 이들을 flew cytometry분석과 Southern blot분석을 하였다. Southern blot으로 분석한 6개의 clone들 모두가 Ly6 cDNA를 갖고 있으며, 또한 neomycin resistance유전자를 갖고 있음이 밝혀졌다. 모든 클를을 FACS분석법으로 Ly6항원의 발현을 조사한 결과, 한 클론이 Ly6항원의 발현이 현저히 억제된 것으로 밝혀졌다. 이 클론은 CTL에 대한 민감성과 allo-MHC반응을 유도하는 능력을 비교하는데 이용될 것이다.

<P>The possible beneficial effects of chronic low-dose irradiation (LDR) and its mechanism of action in a variety of pathophysiological processes such as cancer are a subject of intense investigation. While animal studies involving long-term exposure to LDR have yielded encouraging results, the influence of LDR at the cellular level has been less well defined. We reasoned that since natural killer (NK) cells constitute an early responder to exogenous stress, NK cells may reveal sentinel alterations in function upon exposure to LDR. When purified NK cells received LDR at 4.2 mGy/h for a total of 0.2 Gy <I>in vitro</I>, no significant difference in cell viability was observed. Likewise, no functional changes were detected in LDR-exposed NK cells, demonstrating that LDR alone was insufficient to generate changes at the cellular level. Nonetheless, significant augmentation of cytotoxic, but not proliferative, function was detected when NK cells were stimulated with low-dose IL-2 prior to irradiation. This enhancement of NK cytotoxicity was not due to alterations in NK-activating receptors, NK1.1, NKG2D, CD69 and 2B4, or changes in the rate of early or late apoptosis. Therefore, LDR, in the presence of suboptimal cytokine levels, can facilitate anti-tumor cytotoxicity of NK cells without influencing cellular proliferation or apoptosis. Whether these results translate to <I>in vivo</I> consequences remains to be seen; however, our data provide initial evidence that exposure to LDR can lead to subtle immune-enhancing effects on NK cells and may explain, in part, the functional basis underlying, diverse beneficial effects seen in the animals chronically exposed to LDR.</P>

The purpose of this study is to investigate the status of the experimental and practical education in high school general science. The present status and reasonable management of the experimental and practical education in high school general science were evaluated by the questionaires. To do this, 146 science teachers in Korea are administered questionaires. The frequency of the experimental and practical items in the seven kind of general science text books were investigated. The problems and the reasonable management for experimental and practical education were proposed in this paper.
Review of Soil Structure Quantification from Soil Images

Soil structure plays an important role in ecological system, since it controls transport and storage of air, gas, nutrients and solutions. The study of soil structure requires an understanding of the interrelations and interactions between the diverse soil components at various levels of organization. Investigations of the spatial distribution of pore/particle arrangements and the geometry of soil pore space can provide important information regarding ecological or crop system. Because of conveniences in image analyses and accuracy, these investigations have been thrived for a long time. Image analyses from soil sections through impregnated blocks of undisturbed soil (2 dimensional image analyses) or from 3 dimensional scanned soils by computer tomography allow quantitative assessment of the pore space. Image analysis techniques can be used to classify pore types and quantify pore structure without inaccurate or hard labor in laboratory. In this paper, the last 50 years of the soil image analyses have been presented and measurements on various soil scales were introduced, as well. In addition to history of image analyses, a couple of examples for soil image analyses were displayed. The discussion was made on the applications of image analyses and techniques to quantify pore/soil structure.
