        This study was aimed to establish the system of centers and to offer the viewpoint to studies and policies of urban spatial structure while analyzing their characteristics in Busan. The results of this study were summarized as followed : According to the result about establishment of the system of centers, 2 Centers, 4 Sub-Centers and 5 low-lanking centers were identified. This is a little different with 2 Centers, 6 Sub-Centers, 2 District-Centers written in 「Busan Master Plan toward 2011's」. In other words, the system of centers in Busan was represented more subdevided and abundant than that of 「Busan Master Plan toward 2011's」 and these centers had both polycentric and multi-nuclei characteristics. With premise that the urban structure is being changed on the basis of the change of a period, society and economy in the future to establish effective system of centers, it is advisable that the establishment of the system of centers is to readjust by stage in the process of planning.

      • 삼일열 말라리아의 감염시 혈중 지질변화

        Background : Alteration in plasma lipid levels during malaria attacks was studied to evaluate the diagnstic values in vivax malaria. Methods : The plasma levels of total cholesterol, triglycerides, HDL-cholesterol (HDL-c), and LDL-cholesterol (LDL-c) were analyzed and compared in 32 patients with vivax malaria at presentation, in 10 patients after 17-days of treatment with anti-malaria drug, and in 40 control individuals. Interrelation of lipid profile with other parameters including parasitemia level, platelet count, hemoglobin and WBC counts were analysed. Results : In patients with malaria, serum total cholesterol, triglycerides, HDL-c and LDL-c concentrations were significantly lower than those of control subjects. None of lipid profile showed any correlation with the parasitemia level. After treatment, HDL-c was significantly elevated. Conclusion : These results suggest that lipid profile, especially decreased of HDL-c, may be a valuable information in the diagnosis of the malaria. (Korean J Infect Dis 33:58∼61, 2001)

      • 과학 수업에 대한 초등학교 아동의 감성 상태와 기억 유형과의 관계

        In this study, we examined their memories on science instruction and categorized them, and explored relationships between their emotional aspects and memory types. The study involved each three classes of the fifth- and sixth-grade students respectively who learned science topics through traditional ways. The amount of immediate memory on the instruction was highest in semantic one. In degrees of persistence of each memory type, which measured immediately and after one-week, the semantic and emotional memories decreased, whereas the episodic one increased. The more positive emotion the children had in science lesson, the more were total amounts of memories. In the positive emotional states, the proportion of semantic memory was higher than in the negative ones. On the other hand, episodic memory showed opposite trend. Boys and more episodic memories, whereas girls hand more semantic memories. The findings are discussed, including implications for science teaching.

        CD40 Agonists Alter the Pancreatic Cancer Microenvironment by Shifting the Macrophage Phenotype toward M1 and Suppress Human Pancreatic Cancer in Organotypic Slice Cultures

        Background/Aims: CD40 agonists are thought to generate antitumor effects on pancreatic cancer via macrophages and T cells. We aimed to investigate the role of CD40 agonists in the differentiation of macrophages and treatment of human pancreatic adenocarcinoma. Methods: Immunohistochemistry was performed on paraffin-embedded surgical blocks from patients with pancreatic cancers to evaluate macrophage phenotypes and their relationship with survival. The effects of CD40 agonists on macrophage phenotypes and human pancreatic cancer were evaluated utilizing cell cocultures and organotypic slice cultures. Results: CD163+ (predominant in M2 macrophages) and FOXP3+ (predominant in regulatory T cells) expression levels in the tumors were significantly lower in patients with stage IB pancreatic cancer than in those with stage II or III disease (p=0.002 and p=0.003, respectively). Patients with high CD163+ expression had shorter overall survival than those with low CD163+ expression (p=0.002). In vitro treatment of THP-1 macrophages with a CD40 agonist led to an increase in HLA-DR+ (predominant in M1 macrophages) and a decrease in CD163+ expression in THP-1 cells. Cell cocultures showed that CD40 agonists facilitate the suppression of PANC-1 human pancreatic cancer cells by THP-1 macrophages. Organotypic slice cultures showed that CD40 agonists alter the pancreatic cancer microenvironment by shifting the macrophage phenotype toward M1 (increase HLA-DR+ and decrease CD163+ expression), decreasing the abundance of regulatory T cells, and increasing tumor cell apoptosis. Conclusions: CD163 is related to advanced human pancreatic cancer stages and shorter overall survival. CD40 agonists alter macrophage phenotype polarization to favor the M1 phenotype and suppress human pancreatic cancer.

        Growth hormone-releasing peptide-biotin conjugate stimulates myocytes differentiation through insulin-like growth factor-1 and collagen type I

        Based on the potential beneficial effects of growth hormone releasing peptide (GHRP)-6 on muscle functions, a newly synthesized GHRP-6-biotin conjugate was tested on cultured myoblast cells. Increased expression of myogenic marker proteins was observed in GHRP-6-biotin conjugate-treated cells. Additionally, increased expression levels of insulin-like growth factor-1 and collagen type I were observed. Furthermore, GHRP-6-biotin conjugate-treated cells showed increased metabolic activity, as indicated by increased concentrations of energy metabolites, such as ATP and lactate, and increased enzymatic activity of lactate dehydrogenase and creatine kinase. Finally, binding protein analysis suggested few candidate proteins, including desmin, actin, and zinc finger protein 691 as potential targets for GHRP6-biotin conjugate action. These results suggest that the newly synthesized GHRP-6-biotin conjugate has myogenic stimulating activity through, at least in part, by stimulating collagen type I synthesis and several key proteins. Practical applications of the GHRP-6-biotin conjugate could include improving muscle condition. [BMB Reports 2015; 48(9): 501-506]

        BDNF Enhancement of Neuronal Differentiation with Adipose-Derived Stem Cells(ADSCs)

        Adipose-derived stem cells(ADSCs) are multipotent cells that can be manipulated to differentiate into different cell lines and are considered a potential cell source for regenerative medicine and tissue engineering. The brain derived neurotrophic factor (BDNF) is known to stimulate stem cells to differentiate into neuronal cells as well as promote plasticity. The purpose of this study was to evaluate the effects of BDNF on the neuronal differentiation of ADSCs. The proliferation of the ADSCs was increased by BDNF in a dose-dependent manner. The expression of neuronal markers including Nestin and microtubule-associated protein(MAP2) were highly upregulated by BDNF. In addition, BDNF considerably suppressed the expression of glial fibrillary acidic protein(GFAP), a preventative glial cell or astrocyte marker. Using immunocytochemistry, we confirmed the results by real time-PCR. An increased production of Nestin and MAP2 by BDNF, but not GFAP was observed. These data show that supplementation of the ADSCs with BDNF effectively maintained cell survival, proliferation and differentiation of these cells into neuronal precursor cells. In conclusion, specific neuronal differentiation of ADSCs after treatment with BDNF increased the generation of a mature neuronal phenotype.

        Organotypic slice cultures of pancreatic ductal adenocarcinoma preserve the tumor microenvironment and provide a platform for drug response

        <P><B>Abstract</B></P> <P><B>Background</B></P> <P> <I>/Objective</I>: The conventional models currently used to evaluate various anti-tumor therapeutic agents are not sufficient for representing human pancreatic ductal adenocarcinoma (PDA), which has a unique tumor microenvironment. We aimed to produce an organotypic slice culture model from human PDA that resembles the <I>in vivo</I> situation and to evaluate the responses of PDA slices to established cytotoxic drugs.</P> <P><B>Methods</B></P> <P>PDA tissues were obtained from 10 patients who underwent pancreatic resection. The tissues were sliced by a vibratome, and the tumor slices were then cultured. The viability of tumor slices during slice culture was evaluated using H&E and immunohistochemical staining, and stromal cells were demonstrated. The effects of cytotoxic drugs on PDA cell lines and slices were analyzed.</P> <P><B>Results</B></P> <P>Tumor slices maintained their surface areas and tissue viability for at least five days during culture. Preserved proliferation and apoptosis in tumor slices were observed by the expression of Ki-67 and cleaved caspase-3. Stromal cells including macrophages (CD68<SUP>+</SUP> and CD163<SUP>+</SUP>), T cells (CD3<SUP>+</SUP>, CD8<SUP>+</SUP>, and FOXP3<SUP>+</SUP>), and myeloid cells (CD11b<SUP>+</SUP>) were present throughout the culture period. Staurosporine, gemcitabine, and cisplatin treatment of PDA cell lines and tumor slices exerted proportional cytotoxic effects in terms of MTT viability, tumor cell number, and Ki-67 and cleaved caspase-3 expression.</P> <P><B>Conclusions</B></P> <P>Organotypic human PDA slice cultures preserved their viability and tumor microenvironment for at least five days during slice culture. PDA slice culture appears to be a feasible preclinical test model to assess the response to anti-tumor agents.</P>
