Objectives: To examine the effect of diazoxide on hypoxia-induced soluble fms-like tyrosin kinase-1 (sFlt-1) release in JEG-3 choriocarcinoma cells. Methods: Cells were cultured under normoxia (20% O2) or hypoxia (1% O2), and expression of sFlt-1 mRNA and protein release was determined by quantitative real-time reverse-transcriptase polymerase chain reaction (qRT-PCR) assays and enzyme-linked immunosorbent assay (ELISA). Results: Tumor necrosis factor-alpha (TNF-α) as well as hypoxia stimulated sFlt-1 release and diazoxide inhibited both of them. The selective inhibitor of mitochondrial adenosine triphosphat (ATP)-sensitive K+ channel opener (KATP) 5-hydroxydecanoate (5-HD) completely reversed the diazoxide-induced inhibition of hypoxia-stimulated sFlt-1 release. qRT-PCR and Western blot analyses showed that diazoxide up-regulated the heme oxygenase-1 (HO-1) expression. In addition, the HO-1 inducer cobalt protoporphyrin (CoPP) and the metabolic product of HO-1 bilirubin mimicked diazoxide to inhibit sFlt-1 release and reactive oxygen species (ROS) production under hypoxia, whereas the HO-1 inhibitor zinc protoporphyrin IX (ZnPP IX) antagonized the effect of diazoxide. In cells transfected with the HO-1 siRNA, diazoxide did not exert any effect on sFlt-1 release and ROS production under hypoxia. Conclusion: These results, taken together, strongly suggest that up-regulation of the HO-1 expression is the crucial mechanism responsible for the diazoxide-induced inhibition of the sFlt-1 release and ROS production under hypoxia.

The purpose of this study was to evaluate the relationship between variable factors and clinical results following the operative treatment of the tibial plateau fractures. Materials and Methods: The clinical and radiological analysis was performed on 29 cases of the tibial plateau fractures who had been treated with operative treatment and followed up for more than 1 year from January 1991 to December 1997. The analysis of clinical results was performed dividing into age, cause of injury, fracture type of Schatzker classification, associated soft tissue injury and method of operative treatment. Results: According to Schatzker classification, 2 cases(6.9%) were type I, 11 cases (37.9%) were type II, 1 case(3.5%) was type III, 5 cases(17.2%) were type IV, 4 cases(13.8%) were type V, and 6 cases(20.7%) were type VI. In all cases, bony unions were obtained. According to Blokker evaluation, 23 cases(79.3%) of 29 cases were acceptable. Conclusion: We could expect good clinical results if early knee joint mobilization following minimal invasive open reduction and internal fixation could be obtained. Bad clinical results were related with young age group under 30, more than Schatzker classification typeIV of high energy trauma and associated injury of anterior cruciated ligment or meniscus.

Artificial teeth are manufactured through a CAD/CAM technology for disc blocks using zirconia powder, which has excellent aesthetics, strength, and biocompatibility. The milling (CAM) process of the ceramic disc block generally requires two steps as follows; a step processing for forming steps for fixing jigs on the upper and lower outer peripheral surfaces of the disk block, and a vertical surface processing for flatness of the upper and lower surfaces. The above two processes require additional processing and waiting time, and cause material loss and lower quality due to various impact, which ultimately leads to an increase in manufacturing costs. In this study, a research was conducted to exclude the above additional process by mounting a resin assembly to a disk block. The assembly was manufactured using 3D printing equipment and found to be easily combined during the milling process using the so-called Lego method and the ring-groove method, enabling an automated process. By skipping the aforementioned two process steps, effects such as shortening manufacturing time, reducing material loss, improving processability and durability, increasing productivity, and finally reducing manufacturing cost were confirmed.

Mycobacterium avium subsp. paratuberculosis is the pathogen of paratuberculosis called Johne`s disease. Johne`s disease is hardly eliminated because of its long latent period and continuous dissemination, so it is found in ruminants worldwide and can cause substantial economic losses in cattle. It has been reported in many studies on the distribution of Johne`s disease in some provinces of Korea that not many, but noticeable numbers of infected cows have been detected since the first detection in 1984. The aims of this study was to investigate the prevalence of Johne`s disease obtained from slaughtered cattle in central area of Gyeongnam province, Korea. In this study, the ELISA serum antibody test and PCR were employed on a total of 240 blood and ileac substrate samples from slaughtered cattle in two slaughtering and wholesale centers in Gyeongsangnam-do Livestock Veterinary Research Institute Central Branch. Out of the entire 240 blood samples, three (1.3%) were positive by ELISA, while five (2.1%) were suspected cattle. But ileac substrate samples, eight (3.3%) were positive by PCR. By breeds, positive rates of ELISA and PCR in Korean native cattle were 1.3% and 3.5%, respectively, but no positive cows were found in dairy cattle. By provinces, sero-positive rates of Gyeongnam and Gyeongbuk were 1.6% and 1.3%, respectively. And PCR positive rates of Gyeongnam, Gyeongbuk and other provinces were 2.4%, 5.0% and 2.8%, respectively. These results indicate that it requires the nationwide monitoring test and measure to deal with subclinically infected slaughtering cows.

Railway vehicles is composed of mechanical, electrical, electronic, communication, and a variety of systems, failure and also shows a variety of forms, many of the parts are repaired or re-use, is difficult to consider the public interest, such as the safety of the train station and passenger service. Recently “all of the equipment is not to increase the failure rate over time, but failures is often occur at random, regardless of the time” because it is established theory is that in developed railway companies overseas, I have applied the Condition-Based Maintenance(CBM) that know the need for changes in the maintenance system through monitoring the state of the machine and equipment. SMRT is trying to be based on Condition-Based Maintenance(CBM) doing train failure data analysis and risk analysis of target component selection to match to these changes.
