        Effects of load height application and pre-bucklingdeflections on lateral buckling of thin-walled beams

        Based on a non-linear model taking into account flexural-torsional couplings, analytical solutions are derived for lateral buckling of simply supported I beams under some representative load cases. Aclosed form is established for lateral buckling moments. It accounts for bending distribution, load height application and pre-buckling deflections. Coefficients C1 and C2 affected to these parameters are then derived. Regard to well known linear stability solutions, these coefficients are not constant but depend on another coefficient k1 that represents the pre-buckling deflection effects. In numerical simulations, shell elements are used in mesh process. The buckling loads are achieved from solutions of eigenvalue problem and by bifurcations observed on non linear equilibrium paths. It is proved that both the buckling loads derived from linear stability and eigenvalue problem lead to poor results, especially for I sections with large flanges for which the behaviour is predominated by pre-buckling deflection and the coefficient k1 is large. The proposed solutions are in good agreement with numerical bifurcations observed on non linear equilibrium paths.

        Vibrations of long repetitive structures by a double scale asymptotic method

        In this paper, an asymptotic two-scale method is developed for solving vibration problem of long periodic structures. Such eigenmodes appear as a slow modulations of a periodic one. For those, the present method splits the vibration problem into two small problems at each order. The first one is a periodic problem and is posed on a few basic cells. The second is an amplitude equation to be satisfied by the envelope of the eigenmode. In this way, one can avoid the discretisation of the whole structure. Applying the Floquet method, the boundary conditions of the global problem are determined for any order of the asymptotic expansions.

        Pre-buckling deflection effects on stability of thin-walled beams with open sections

        The paper investigates beam lateral buckling stability according to linear and non-linear models. Closed form solutions for single-symmetric cross sections are first derived according to a non-linear model considering flexural-torsional coupling and pre-buckling deformation effects. The closed form solutions are compared to a beam finite element developed in large torsion. Effects of pre-buckling deflection and gradient moment on beam stability are not well known in the literature. The strength of singly symmetric I-beams under gradient moments is particularly investigated. Beams with T and I cross-sections are considered in the study. It is concluded that pre-buckling deflections effects are important for I-section with large flanges and analytical solutions are possible. For beams with T-sections, lateral buckling resistance depends not only on pre-buckling deflection but also on cross section shape, load distribution and buckling modes. Effects of prebuckling deflections are important only when the largest flange is under compressive stresses and positive gradient moments. For negative gradient moments, all available solutions fail and overestimate the beam strength. Numerical solutions are more powerful. Other load cases are investigated as the stability of continuous beams. Under arbitrary loads, all available solutions fail, and recourse to finite element simulation is more efficient.

        Sustained calcium signalling and caspase-3 activation involve NMDA receptors in thymocytes in contact with dendritic cells

        L-glutamate, the major excitatory neurotransmitter, also has a role in non-neuronal tissues and modulates immune responses. Whether NMDA receptor (NMDAR) signalling is involved in T-cell development is unknown. In this study, we show that mouse thymocytes expressed an array of glutamate receptors, including NMDARs subunits. Sustained calcium (Ca<SUP>2+</SUP>) signals and caspase-3 activation in thymocytes were induced by interaction with antigen-pulsed dendritic cells (DCs) and were inhibited by NMDAR antagonists MK801 and memantine. NMDARs were transiently activated, triggered the sustained Ca<SUP>2+</SUP> signal and were corecruited with the PDZ-domain adaptor postsynaptic density (PSD)-95 to thymocyte-DC contact zones. Although T-cell receptor (TCR) activation was sufficient for relocalization of NMDAR and PSD-95 at the contact zone, NMDAR could be activated only in a synaptic context. In these T-DC contacts, thymocyte activation occurred in the absence of exogenous glutamate, indicating that DCs could be a physiological source of glutamate. DCs expressed glutamate, glutamate-specific vesicular glutamate transporters and were capable of fast glutamate release through a Ca<SUP>2+</SUP>-dependent mechanism. We suggest that glutamate released by DCs could elicit focal responses through NMDAR-signalling in T cells undergoing apoptosis. Thus, synapses between T and DCs could provide a functional platform for coupling TCR activation and NMDAR signalling, which might reflect on T-cell development and modulation of the immune response.
