Longhorned tick, Haemaphysalis longicornis (Ixodida: Ixodidae) is one of the vectors of severe fever with thrombocytopenia syndrome virus (SFTSV) in human. The tick occurs in mostly grass fields, and the use of pyrethroid insecticides induced pest resistance and environmental residual toxicity. Particularly the use of chemicals near residential areas where persons live become a big issue, so environmentally safe control agents need to be considered. Here in this work, our interest was given to the selection of highly virulent fungi against H. longicornis. A total of 101 fungal pathogens collected from mountainous areas were assayed by a dipping the nymph stage of ticks into a conidial suspension (1×107conidia/ml). Interestingly among several species, one species showed high virulence and mycosis were observed in 7-15 days. Most of the selected isolates produced a large amount of conidia in Italian millet, rice and millet with thermotolerance at 40-45°C. Based on these results, we selected a couple of isolates with high virulence against H. longicornis and they could be used for the control of the ticks after the set-up of a practical application strategy in fields by optimizing fungal colonization in soil and phyllosphere.
The Effect of Gamma Irradiation on the Development of Harmonia axyridis (Col.: Coccinellidae)

The multicolored Asian ladybird beetle, Harmonia axyridis (Pallas) (Coleoptera: Coccinellidae), is common to a wide range of natural and agricultural habitats. Applications of gamma irradiation minimized the losses of stored food and the death or failure of emergence in larval and pupal stages. On the other hand, degrade toxin waste as one of alternative to chemical pesticide for both quarantine and sprout control purposes of storage crop pest. So, we have investigated whether gamma irradiation exposed to eggs, lava, pupa and adults of H. axyridis. It may be affected the emergence, fertility, fecundity, development period and sex ratio of H. axyridis. Some changes of physiological characteristics may be applied to more efficient agents as biological control of several aphids. Insects were exposed to gamma irradiation from 0 to 500 Gy of 60Co depended on their developmental stages. The results showed that the first instar, eggs, third instar, pupae, and adults were more sensitive in order of irradiation dose. And fecundity and fertility of female adults were significantly decreased with increasing gamma irradiation dose at all tested individuals.

The multicolored Asian ladybird beetle, Harmonia axyridis, which demonstrates typical genetic polymorphism in its elytra color patterns. Early studies have color polymorphism in terms of geographical clines while a few investigated temporal populations in Coccinellidae. Nevertheless, note that geographical and temporal morph variation does not always correspond to what is expected from thermal and industrial adaption theories. A recent study of transformation and RNAi of the ladybird beetle, however, there is yet no evidence to indicate the variation is genetic or environmental factors. Here we describe a relatively new molecular fingerprinting technique, amplified fragment length polymorphism (AFLP). Because we think that color polymorphism in Coccinellidae is affected by genetic polymorphism. In total 38 markers were scored from which some markers were polymorphic. Supsequent UPGMA cluster analysis revealed 3 major group of Harmonia axyridis populations. But for strains that are more genetically similar, different primer combinations may be need to generate enough polymorphic marker.

Myeloid differentiation factor 88 (MyD88) is an intracellular adaptor protein involved in Toll signaling pathway. In this study, we monitored the response of 4 key genes of the insect immune system against Beauveria bassiana JEF-007 in Tenebrio molitor using RT-PCR. TmGPR, antimicrobial peptide Tenecin 1 and Tenecin 2 were up-regulated after fungal infection. To better understand the roles of Toll signaling pathway in mealworm immune system, TmGRP and TmMyD88 was knocked down by RNAi silencing. Target gene expressions were decreased at 2 days post-dsRNA injection, and dramatically reduced at 6 days post-dsRNA injection. Therefore, mealworms were compromised by B. bassiana JEF-007 at 6 days post-dsRNA injection. Silencing of the TmMyD88 and TmGRP resulted in reducing the resistance of the host to fungal infection. However, only dsTmMyD88 showed significant difference with dsEGFP by statistical analysis, which may be due to partial gene knock down of dsGRP. These results indicate that TmMyD88 is required in mealworms for survival against B. bassiana JEF-007.

Here in this work, our interest was given to the selection of highly virulent fungi against longhorned tick. A total of 101 fungal pathogens were assayed by dipping the nymph stage of ticks into a conidial suspension. Interestingly of the several species, one species showed high virulence and mycosis were observed in 7-15 days. Highly virulent strains were selected, and semi-field experiments were conducted. As a result, the control efficacy of the isolate was over 80% at 30 days of treatment. This work suggests that entomopathogenic fungi could be used to effectively control longhorend ticks.
Evaluation of Entomopathogenic Fungi for the Control of Hard Tick, Haemaphysalis longicornis

The hard tick, Haemaphysalis longicornis (Ixodida: Ixodidae) is one of the vectors of severe fever with thrombocytopenia syndrome virus (SFTSV) causing in humans, however little consideration was given to the biological control of the pest. Therefore, this study provides a screening method to select entomopathogenic fungi, having high virulence against H. longicornis. The virulence test was conducted by dipping method with a conidial suspension (1×107 conidia/ml). As a result, cadavers of H. longicornis infected with entomopathogenic fungi were obtained by this method. Based on this results, we selected several isolates having high virulence to H. longicornis. We suggest that, selected isolates in this study can be used for the control of H. longicornis.

The hard tick, Haemaphysalis longicornis (Ixodida: Ixodidae) is one of the vectors of severe fever with thrombocytopeniasyndrome virus (SFTSV) causing in humans, however little consideration was given to the biological control of the pest.We evaluated three bioassay methods, such as spray, dipping and dropping. As a result, the dipping method was themost efficient way to measure the fungal virulence. Finally, we constructed a Haemaphysalis longicornis pathogenic fungallibrary to further facilitate the resources to be used as potential biological control agents. A conidial suspension of 1×107conidia/ml was exposed to the ticks by dipping. We confirmed that some isolates were highly virulent to the ticks. Thisresults indicated that some fungi could be used to effectively control the Haemaphysalis longicornis.
