Numerical simulation is carried out for flow characteristics in a plenum fan and the influence of the diameter ratio of the rotating vaneless diffuser on the performance of plenum fan is analyzed. The diameter ratio of the rotating vaneless diffuser employed is from 1.03 to 1.3. The research results show that the rotating vaneless diffuser is able to enhance the performance of plenum fan. It is found that there is significant improvement in static pressure and efficiency at the diameter ratio of 1.05 at high flow coefficients, while the optimal diameter ratio is 1.2 at rated and low flow coefficient.

A method of optimization design for the blade profile of a centrifugal impeller by controlling velocity distribution is presented, and a plenum fan is successfully designed. This method is based on the inner flow calculation inside the centrifugal impeller, and is related to the distribution of relative velocity. The results show that after optimization, the boundary layer separation on the suction surface has been inhibited and the stability of plenum fan is improved. The flow at the impeller outlet is also studied, and the jet-wake pattern at the impeller outlet is improved obviously by optimization. The calculation result shows that the static pressure and static pressure efficiency can be increased by 15.4% and 21.4% respectively.

Currently, enzymatic synthesis of lactulose, a synthetic prebiotic disaccharide, is commonly performed with glycosyl hydrolases. In this work, a new type of lactulose-producing biocatalyst was developed by displaying β-galactosidase from Bacillus stearothermophilus IAM11001 (Bs-β-Gal) on the surface of Bacillus subtilis 168 spores. Localization of β-Gal on the spore surface as a fusion to CotX was verified by western blot analysis, immunofluorescence microscopy, and flow cytometry. The optimum pH and temperature for the resulting spore-displayed β-Gal was 6.0 and 75℃, respectively. Under optimal conditions, it showed maximum activity of 0.42 U/mg spores (dry weight). Moreover, the spore-displayed CotX-β-Gal was employed as a whole cell biocatalyst to produce lactulose, yielding 8.8 g/l from 200 g/l lactose and 100 g/l fructose. Reusability tests showed that the spore-displayed CotX-β-Gal retained around 30.3% of its initial activity after eight successive conversion cycles. These results suggest that the CotX-mediated spore-displayed β-Gal may provide a promising strategy for lactulose production.

Two model proteins of an enhanced green fluorescent protein (EGFP) and a tetrameric β- Galactosidase (β-Gal) from Bacillus stearothermophilus IAM11001 were fused to the C-terminus of the crust proteins Y (CotY) and Z (CotZ) from B. subtilis 168. Surface-expression of the resulting fusion proteins CotY-EGFP, CotZ-EGFP, CotY-β-Gal, and CotZ-β-Gal were then evaluated. Fluorescence intensity analysis of the transformed strains indicated that the CotY-EGFP and CotZ-EGFP fusion proteins were successfully displayed on spores. β-Gal was anchored on the surface of B. subtilis spores, confirmed based on western blotting and immunofluorescence microscopy. Results of a β-Gal assay showed that when fused to CotY and CotZ, the obtained enzyme activities of spore-displayed β-Gals were 0.12 and 0.25 U/mg of spores (dry weight), respectively. CotY and CotZ were, thus, shown to be suitable candidate carriers for display of multimeric enzymes on the spore surface.

Intervertebral disc degeneration (IDD) is characterized by excessive apoptosis of nucleus pulposus (NP) cells and hyperactive extracellular matrix (ECM) catabolism. Our previous studies revealed the relationship between human islet amyloid polypeptide (hIAPP) and NP cell apoptosis. However, the role of hIAPP aggregates in IDD has not yet been investigated. This study aimed to determine whether the accumulation of hIAPP aggregates promotes IDD progression. The aggregation of hIAPP increased in human NP tissues during IDD. The deposition of hIAPP aggravated the compression-induced IDD that promoted NP cell apoptosis and ECM degradation via IL-1β/IL-1Ra signaling in an ex vivo rat disc model. Moreover, neutralizing IL-1β augmented the protective effects of hIAPP overexpression by decreasing hIAPP aggregation in human NP cells. These results suggest that the aggregation of hIAPP promotes NP cell apoptosis and ECM degradation ex vivo and in vitro by disrupting the balance of IL-1β/IL-1Ra signaling.

Low back pain (LBP) is a major musculoskeletal disorder and the socioeconomic problem with a high prevalence that mainly involves intervertebral disc (IVD) degeneration, characterized by progressive nucleus pulposus (NP) cell death and the development of an inflammatory microenvironment in NP tissue. Excessively accumulated cytosolic DNA acts as a damage-associated molecular pattern (DAMP) that is monitored by the cGAS-STING axis to trigger the immune response in many degenerative diseases. NLRP3 inflammasome-dependent pyroptosis is a type of inflammatory programmed death that promotes a chronic inflammatory response and tissue degeneration. However, the relationship between the cGAS-STING axis and NLRP3 inflammasome-induced pyroptosis in the pathogenesis of IVD degeneration remains unclear. Here, we used magnetic resonance imaging (MRI) and histopathology to demonstrate that cGAS, STING, and NLRP3 are associated with the degree of IVD degeneration. Oxidative stress induced cGAS-STING axis activation and NLRP3 inflammasome-mediated pyroptosis in a STING-dependent manner in human NP cells. Interestingly, the canonical morphological and functional characteristics of mitochondrial permeability transition pore (mPTP) opening with the cytosolic escape of mitochondrial DNA (mtDNA) were observed in human NP cells under oxidative stress. Furthermore, the administration of a specific pharmacological inhibitor of mPTP and self-mtDNA cytosolic leakage effectively reduced NLRP3 inflammasome-mediated pyroptotic NP cell death and microenvironmental inflammation in vitro and degenerative progression in a rat disc needle puncture model. Collectively, these data highlight the critical roles of the cGAS-STING-NLRP3 axis and pyroptosis in the progression of IVD degeneration and provide promising therapeutic approaches for discogenic LBP.
