        Acute and Chronic Effects of Nanoplastics on the Water Flea Moina macrocopa

        Here, upon acute (96 h) and chronic (14 days) exposure, ingestion of polystyrene NPs (100 nm) and physiological, biochemical, and cholinergic modulations were analyzed in the water flea Moina macrocopa exposed to different concentrations (0.001, 0.01, 0.1, 1, 5, 10, 50, 100, and 500 μg l-1). Exposed NPs were observed in the internal organs (e.g., digestive tract and foregut) of the water flea. Chronic exposure to the relatively high concentrations resulted in significant decreases in survival, body length, and the total number of molts, whereas reproduction parameter was not affected. Significant increase in oxidative stress biomarker (malondialdehyde) and decrease in the intracellular content of endogenous antioxidant (glutathione) and enzymatic activity of antioxidant enzymes (glutathione peroxidase, glutathione reductase, catalase, superoxide dismutase, and glutathione S-transferase) were detected in response to relatively high concentrations of NPs. Transcriptional expression of the hsp70 gene was increased in response to relatively high concentrations of NPs, whereas acetylcholinesterase activity was lowered by the same concentrations of NPs. Taken together, NPs exposure would be a significant modulator on physiological and biochemical metabolism of water flea.

        Mitigating off-target distribution and enhancing cytotoxicity in breast cancer cells with alpha-ketoglutaric acid-modified Fe/Mg-CA nanoparticles

        In this work, pH-sensitive alpha-ketoglutaric acid-modified Fe/Mg-carbonate apatite (α-KAM-Fe/Mg-CA) NPs were introduced and found to be capable of promoting the selective delivery of cancer-killing drug doxorubicin (DOX) in breast cancer cells, while simultaneously mitigating DOX toxicity on normal cells.As part of the characterization and evaluation of α-KAM-Fe/Mg-CA NPs to target breast cancer cells, a series of assessments were performed, which included size measurements, morphological analysis, FTIR, cytotoxicity assessment, hemolysis, drug binding, cellular uptake, and pH-responsive drug release tests. Liquid chromatography-mass spectrometry was used to conduct the protein corona analysis of α-KAM-Fe/Mg-CA using 10% FBS (fetal bovine serum) and mice plasma. Furthermore, to investigate the distribution of DOX-loaded α-KAM-Fe/Mg-CA NPs in major tissues and the tumor, a biodistribution investigation was conducted in mammary tumor-induced Balb/c mouse models 24 h after the intravenous administration of DOX-loaded α-KAM-Fe/Mg-CA NPs.The in vitro pH-dependent release of DOX over time demonstrated that α-KAM-Fe/Mg-CA NPs were pH-responsive and degraded rapidly at acidic pH levels. When compared to free DOX, the DOX-loaded α-KAM-Fe/Mg-CA NPs demonstrated a potent antiproliferative effect on breast cancer cells. Confocal microscopy confirmed the effective internalization of DOX-loaded α-KAM-Fe/Mg-CA NPs in breast cancer cells. The protein corona analysis revealed an affinity for dysopsonins (serum albumin, apolipoproteins) and transport proteins that may assist in extending their blood circulation period. Furthermore, biodistribution data of DOX-loaded α-KAM-Fe/Mg-CA NPs in the mammary tumor-induced Balb/c mouse model indicated extended circulation in the bloodstream, reduced non-target distribution in major tissues, and increased drug accumulation in the tumor.The results obtained suggest that α-KAM-Fe/Mg-CA NPs may emerge as a prospective candidate for delivering therapeutic cargos to treat malignant mammary tumors.

        Nutrient Uptake and Productivity as Affected by Nitrogen and Potassium Application Levels in Maize/Sweet Potato Intercropping System

        Field experiment was conducted during 1993-94 season to determine the pattern of nutrient uptake and productivity of maize/sweet potato intercropping system. Four levels of nitrogen (0, 50, 100 and 150kg N ${ha}_{-1}$) and four levels of potassium (0, 40, 80 and 120kg $K_2$O ${ha}_{-1}$) formed treatment variables. Plants were sampled periodically to determine dry matter and tissue concentrations of N and K in the individual plant components of intercropped maize and sweet potato. Nitrogen and potassium fertilizer did not interact significantly to nutrient uptake by any plant parts of intercropped maize and sweet potato. But application of N fertilizer independently enhanced N uptake in all the plant parts of maize and sweet potato. The uptake of N in leaf, leaf sheath, stem, husk, and cob of maize increased upto 90 days after planting (DAP) but grain continued to accumulate N till its maturity. Sweet potato exhibited a wide variation in N uptake pattern. Sweet potato leaf shared the maximum uptake of N at 50 DAP which rapidly increased at 70 DAP and then declined. Declination of N uptake by petiole and stem were observed after 120 DAP whereas N uptake by tuber increased slowly upto 90 DAP and then rapidly till harvest. Rate of applied K had very little effect on the uptake patterns in different components of intercropped maize. Pattern of K uptake by leaf, petiole and stem of sweet potato showed almost similar trend to N uptake. But uptake of K by tuber increased almost linearly with the K application. Pattern of N and K uptake by grain and tuber paralleled the grain yield of maize and sweet potato respectively. Intercropped productivity of maize and sweet potato found to be better by the application of 100kg N and 120 kg $K_2$O ${ha}_{-1}$

        Current Status and Technologies for Treating Groundwater Arsenic Pollution in Bangladesh

        Arsenic (As) contamination in groundwater is one of the main problems in Bangladesh. As toxicity causes serious human health problems such as edema, skin cancer, bladder cancer, lung cancer, hyperkeratosis, premature birth, and black foot disease. As contamination in groundwater mainly originates from the geological characteristics of the area due to the influence of anthropogenic activities. Since most of the people in Bangladesh rely on tube well for drinking water, it is necessary to investigate the current status of As pollution and identify the treatment technologies that can be used to provide arsenic-free drinking water in water-scarce areas. A total of 92 papers were reviewed in this study to present a complete overview of the recent status of groundwater As contamination in Bangladesh and different low-cost remediation technologies. A method for evaluating the relative feasibility of different treatment technologies was also utilized to determine the most appropriate technologies for groundwater As treatment in Bangladesh. The districts with the highest groundwater As contamination include Brahamanbariya, Tangail, Barisal, Pabna, Patuakhali, Kurigram, Magura, and Faridpur, with concentrations exceeding 0.05 mg/L. Only six districts had relatively low groundwater arsenic concentrations (0.01 mg/L), including Kushtia, Khagrachari, Jessore, Dinajpur, Meherpur, and Munshiganj. There were a number of technologies used for treating As in water, but aerated electrocoagulation, Mg-Fe-based hydrotalcite-like compound, and electro-chemical As remediation (ECAR) reactor were found to be the most feasible treatment methods for As. Overall, the investment, operational, and maintenance costs, availability of materials, and expertise requirements should be considered when selecting the most appropriate treatment method for As in water.

        Physiological effects of biocide on marine bivalve blue mussels in context prevent macrofouling

        Background: Mussels are stubborn organisms attached to solid substrata by means of byssus threads. The abundance of marine mussel Mytilus edulis in marine facilities like power stations was reason to select among fouling animals. Methods: Mortality patterns as well as physiological behavior (oxygen consumption, foot activity, and byssus thread production) of two different size groups (14- and 25-mm shell length) of M. edulis were studied at different hydrogen peroxide concentrations ($1-4mg\;l^{-1}$). Results: Studied mussels showed progressive reduction in physiological activities as the hydrogen peroxide concentration increased. Mussel mortality was tested in 30 days exposure, and 14 mm mussels reached the highest percentage of 90% while 25 mm mussels reached 81%. Produced data was echoed by Chick-Watson model extracted equation. Conclusions: This study points that, while it could affect the mussel mortality moderately in its low concentrations, hydrogen peroxide has a strong influence on mussels' physiological activities related to colonization. Therefore, hydrogen peroxide can be an alternative for preventing mussel colonization on facilities of marine environment.

      • Pharmacological study of 'treenoponchomul'-an Ayurvedic diuretic preparation

        The pharmacological effects of an Ayurvedic diuretic drug 'Treenoponchomul' (TPM) was investigated in animal model. The pharmacological actions of the test drug along with that of the components thereof, on the Central Nervous System (CNS) were studied. The drug under study TPM showed little effect on the CNS, the same can not be said about the components. The most prominent CNS depressant effect was observed with Saccharum officinarum Linn. (EE) in that it lowered the spontaneous motor activity as well the exploratory -behavior of the animals. An exploration retarding effect of moderate degree, was evident with Imperata cylindrica Beauv. (UU), and Phragmites maxima Blatter & McCann (NN). Although the test drug did not alter the normal locomotor and/ or exploratory behavior of the treated animals, it did significantly (p<0.01) lower the locomotion of the amphetamine induced hyperactive animals. TPM along with its components (especially Desmostachya bipinnata Stapf. Root, KU), significantly reduced the gastro-intestinal motility of the treated animals (p<0.01). The test drug and its components lowered the body weight of the treated animals, on being administered chronically (30 days), with EE being the only exception.

        Combination strategy of multi-layered surface camouflage using hyperbranched polyethylene glycol and immunosuppressive drugs for the prevention of immune reactions against transplanted porcine islets

        <P><B>Abstract</B></P> <P>This study suggests a novel method of stabilizing fragile porcine islets to prevent the dissociation after isolation and reducing immune cell invasion in a combination therapy of ‘surface camouflaging’ and immunosuppressive drugs (FK506, Rapamycin, MR-1, anti-CD19 mAb, and Clodrosome<SUP>®</SUP>) to effectively alleviate overall immune reactions against xenotransplanted porcine islets. The surface camouflage of pancreatic islets using biocompatible materials improved stabilization of pancreatic islet and prevented the infiltration of immune cells. Firstly, the surface of porcine islets was camouflaged by SH-6-arm-PEG-lipid and gelatin-catechol (artificial extracellular matrix) in order to stabilize the fragile isolated islets. Secondly, three different PEG layers (6-arm-PEG-SH, 6-arm-PEG-catechol, and linear PEG-SH) were chemically conjugated onto the surface of the stabilized porcine islets. Both artificial extracellular matrix (artificial ECM) and PEGylation effectively covered the surface of porcine islets without increasing the size of the whole islet. In addition, the viability and functionality of the islets were not affected by this multi-layer surface modification. The multi-layer modification significantly reduced the attachment of human serum albumin, fibronectin, and immunoglobulin G in comparison to the control collagen surface. The combination effect of multi-layer PEGylation and cocktailed immunosuppressive drugs on the survival time of the transplanted islets was assessed in a xenogeneic porcine-to-mouse model. The median survival time (MST) of ‘artificial ECM + PEGylation' group was 4-fold increased compared to that of control group. In addition, the MST of ‘artificial ECM + PEGylation + drug' group was 2.16-fold increased, compared to the ‘control + drug’ group. In conclusion, we proposed a novel porcine islet transplantation protocol using surface multi-layer modification and cocktailed immunosuppressive drugs, for stabilization and immunoprotection against xenogeneic immune reactions.</P>

        Dose- and age-specific antioxidant responses of the mysid crustacean <i>Neomysis awatschensis</i> to metal exposure

        <P><B>Abstract</B></P> <P>Waterborne metals can adversely affect an organism’s innate defenses through oxidative stress. In the present study, the marine mysid <I>Neomysis awatchensis</I> was exposed to sublethal concentrations (1/10 and 1/5 of the median LC50s) of As, Cd, Cu, Pb, and Zn for 48 or 96 h at the juvenile and adult developmental stages, and the dose- and age-specific antioxidant defense system responses were characterized. Metal accumulation and modulation of four key antioxidant biomarkers, malondialdehyde (MDA), glutathione (GSH), superoxide dismutase, and catalase, were measured in juvenile and adult mysids. Based on LC50, level of metal toxicity was dependent on metal concentration, developmental stage, and the exposure duration. Intracellular MDA content was increased in the As-, Cu-, Pb-, and Zn-exposed juvenile mysids after exposure for 48 and/or 96 h, while increases in MDA content were observed in adult mysids following Cu and Zn exposure. Interestingly, GSH content was differentially modulated, where intracellular GSH levels decreased in juvenile mysids following Cd, Cu, Pb, and Zn exposure, but significantly increased in metal-exposed adult mysids, except for Pb exposure. The catalase and superoxide dismutase activities displayed similar stage-specific increases or decreases as also observed for the different GSH levels, suggesting that the susceptibility to and defense against metal-induced oxidative stress differed based on stage. Modulations in MDA and GSH content and enzymatic activity of the antioxidant defense system indicate that mysid antioxidant defense system factors are intimately connected during control of oxidative imbalances with different capacities at different developmental stages.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Evidence on the age-specific bioconcentration of five metals in the marine mysid was obtained. </LI> <LI> Age-specific relationship between metal dynamics and ecotoxicity between ages was observed. </LI> <LI> Oxidative stress was induced by metals and caused distinctive age-specific antioxidant response. </LI> </UL> </P>
