Pullorum disease and fowl typhoid are septicemic avian diseases transmitting through egg by transovarian infections. A series of tests has been performing in breeding flocks of chickens and test plans for proper inspection have been modified by government veterinary institute to control of such diseases.To improve inspection plans, different test methods were compared using fowl typhoid positive samples from a poultry farm located in Jeonbuk state in 2012. Based on first inspection, 11 samples among total 200 samples were positive by rapid slide agglutination (RSA) test and 7 samples among RSA positive samples were finally diagnosed as Salmonella Gallinarum infection by ELISA, bacterial isolation, PCR,and histopathologic examination. In the second inspection, 20 samples among total 100 samples were positive by RSA test. Among RSA positive ones, 19 samples were positive by ELISA, S. Gallinarm were successfully isolated in 3 samples, and 16 samples were positive by PCR in the cecal tonsils where were not successful for bacterial isolation. Based on histopathologic examination, severe inflammation in the 13 cecal tonsils and infiltration of lymphocytes and heterophils in the 11 livers were observed.Therefore, we suggest that bacterial isolation, PCR, and histopathologic examination methods in the third inspection need to be further used in various tissues for correct diagnosis and for final eradication of pullorum disease and fowl typhoid in breeding flocks of chickens.

Chicken anemia virus (CAV) has been recognized as an immunosuppressive agent and plays role as an etiological agent of multifactorial diseases in chicken. In this study, we investigated distribution of CAV antibody by ELISA and the virus gene by PCR in poultry farms in Jeongeup, Jeonbuk province. In the test using ELISA kit, 41 (95.3%) of 43 flocks and 88.6% of the individual chickens were positive, respectively. By PCR, 90.9% of the broiler breeders and 75.0% of White-semi breeders were found positive, respectively. All hatchery was negative by PCR. Of the clinical cases from 49 poultry flocks, 87.5% of flocks and 54.7% for each samples were found positive by ELISA, respectively. By PCR test, 21 (42.9%) of 49 flocks were positive. Major clinical signs of the infected flocks were growth retardation, femoral subcutaneous bleeding, depression, limping, and continuing selection. The genetic analysis of separate N genes of CAV showed highly homologous each other. The nucleotide sequence of field isolates had homology ranged from 99.9% to 97.5% with Chinese strains, and 99.9% to 99.6% with Japanese strain. Phylogenetic analysis based on the N gene of CAV isolates showed the closely relation with Chinese strains. The results of this survey could be used as basic data for development of vaccine.

The purpose of this study was to investigate the seroprevalence of paratuberculosis in Korean cattle in western Jeonbuk area. From February 2012 to January 2013, blood samples were collected from 2,606 Korean cattle of 263 farms. As a result, 60 (2.3%) heads of 46 (17.5%) farms were positive on the ELISA test for paratuberculosis. Based on regional analysis, 18 (19.6%) out of 92 farms and 24 (2.5%) out of 941 heads in Iksan area, 28 (22.0%) out of 127 farms and 36 (2.8%) out of 1,291 heads in Kimje area were positive but samples from Gunsan area were all negative. According to scale breeding, small scale (below 50 heads) breeding showed the most high prevalence rate compared to middle (50 to 99 heads) or large (over 100 heads) scale breeding. To clarify the relation between number of heads and paratuberculosis prevalence, some additional analysis would be required in further, though.

Avian adenoviruses are diverse group of pathogens and recently hydropericardium hepatitis syndrome(HHS) is an important, emerged disease of poultry. Particularly 2-3 weeks old age broilers increased mortality ranging from 20-30% and brown native chicken 3-7 weeks sudden onset with mortality 20-50%, typically development secondary infection. The infection chicken shows liver enlarged, pale and straw-colored fluid is present in the sac surrounding the heart. Histopathological positive samples have necrotic foci and basophilic intranuclear inclusion bodies in the hepatocytes and polymerase chain reaction(PCR) was useful for detect the fowl adenovirus(FAV) associated with HHS.

The strategy for infectious bursal disease (IBD) control and its success rate under field conditions depends on hygiene management, IBD field pressure, level, and variation in maternally derived IBD antibodies. This study investigated the level of IBD-specific antibody by ELISA and the prevalence of IBD virus by PCR in broilers, white-semi broilers, and Korean native chickens raised in Jeongeup, Jeonbuk. IBD-specific maternally derived antibodies were measured from 698 chickens and the mean titers of maternal antibodies were 3,572±1,402 in broilers, 1,262±762 in white-semi broilers, and 1,932±912 in Korean native chickens. At 2 weeks after vaccination, the geometric mean antibody titers of broiler, white-semi broiler, and Korean native chicken were 582±427, 3255±1,080, and 1,023±499, respectively. According to sequence analysis of the variable virion protein 2 gene, 4 isolates were found to be very virulent IBDV, 9 isolates classical virulent, and 2 isolates intermediate plus vaccine strain.

SBovine tuberculosis is a chronic bacterial disease of animals and humans caused by Mycobacterium bovis. Besides the classical intradermal tuberculin test, a number of blood and serum tests have been used. The purpose of this study was to establish seroprevalence of M. bovis. The sera were screened using the ELISA technique. A total seroprevalence of 65.8% in positive cattle, suspect 36.0%, negative 5.9% in TB-infected herds by PPD and dairy cattle is 3.0%, Hanwoo is 1.6% in TB-free herds. The deer of seroprevalence is 55.0% in TB-infected herd and 7.7% in TB-free herds.

Infectious bronchitis virus (IBV) causes an acute and highly contagious viral disease of chicken that is great economic losses to the poultry industry worldwide. Among the IBV structural proteins, the high rate spike glycoprotein S1 gene mutation and antigenic variant strains have been reported in many countries. During the years 2012∼2014, 10 IBV strains were isolated from infected chicken farms distributed in provinces of Jeonbuk. Analysis of the S1 gene sequences amplified from 10 isolated strains with QX strains showed nucleotide homologies ranging from 96.5 to 95.4%. Phylogenetic analysis revealed that all strains were clustered into QX-like groups. This study suggests that QX-like IBVs are circulating in commercial chicken farms in Jeonbuk. Therefore, the continuing survellance is significantly important for prevention and control of BIV infection.

Highly pathogenic avian influenza(HPAI) is one of the most important zoonoses in the around of world. That occurred in the domestic ducks of 34 olds day in Jeonbuk of Korea on April 2008. All of infected ducks were died. We could found several clinical signs such as facial swelling with cloudy eyes, conjunctivitis, and neurological sign. In the autopsy, moderate congestion in the kidney, multifocal hemorrhagic foci in pancreas, and mild swelling in brain were observed. Gliosis in brain and severe congestion in kidney were also shown in histopathological findings. Specific band for H5 antibody was detected in polymerase chain reaction (PCR).

There are several immunosuppressive viral diseases in chickens such as avian adenovirus (AAV), chicken anemia virus (CAV), infectious bursal disease (IBD) and Marek`s disease (MD). In this study, we have investigated two broiler chicken farms suffered from high mortality in Jeonbuk in July to August 2009. Clinically high fever and growth retardation were observed in the diseased chicken. In necropsy, the hemorrhages in thigh leg and thymus, hemorrhages and enlargement of liver, kidney and proventriculus, and yellowish fluid in heart were seen. Histologically, necrotic foci and basophilic intranuclear inclusion bodies of hepatocytes, hemorrhages and infiltrated lymphocytes in kidney and proventriculus were observed. By using polymerase chain reaction (PCR), the genes of avian adenovirus, CAV and ND virus were detected in specimens. We suggested that these coinfection cases with high mortality were due to primarily infection of immunosuppressive diseases such as avian adenovirus, CAV, followed by secondary infection of Newcastle disease (ND) virus.

A case of aspergillosis was observed in a flock of 5∼10 days old commercial domestic ducks in Jeongeup. The clinical signs were depression and breathing by gasping or rapid opening of the bill. Grossly, lung was attached to pleura with white cheesy plaques. Numerous nodules was found in lung. Histopathologically, severe granulomatous pneumonia with intralesional fungal hypha were observed in the lung. Aspergillus spp. colonies was isolated from fungal culture and identified by polymer chain reaction.
