유전적으로 고혈압을 유발시킨 쥐(spontaneously hypertensive rat, SHR)의 심근세포와 장간막 동맥의 평활근세포에서 근장그물(sarcoplasmic reticulum, SR) 칼슘 조절 단백질의 발현 level의 변화와 그 기능의 이상 여부를 조사하였다. 고혈압군은 수축기압이 평균 202 ±7.9 mmHg인 SHR을 이용하였고, 대조군인 정상혈압군으로는 이와 연령이 동일하면서 139 ±3.9 mmHg의 혈압을 갖는 Wistar-Kyoto (WKY) 쥐를 설정하였다.
심장근의 이완과 수축시 Ca2+을 근장그물내로 유입(uptake)하거나 근장그물 내부로부터 세포질로 유리(release)시키는 근장그물 막의 중요한 Ca2+ 조절 단백질의 mRNA와 단백질의 상대적 발현 level을 확인하고자, competitive RT-PCR과 western blot analysis를 각각 실시하였다. 심근의 이완에 관여하는 SR Ca2+-ATPase (SERCA2a) 와 phospholamban은 서로 상반된 형태로 발현되었다. 즉 정상혈압군에 비해 SHR에서 SR Ca2+-ATPase의 mRNA와 protein level은 감소된 반면, phospholamban의 경우 증가되었다. Ca2+ uptake의 maximum rate 역시 SHR에서 감소되어 나타났는데, 이와 같은 결과들은 고혈압에서 심근 이완이 저하된 원인으로 생각된다. 또한 심근의 수축을 조절하는 중요한 단백질인 ryanodine receptor는 SHR에서 mRNA level과 [3H] ryanodine binding이 감소되었다.
혈압 상승으로 인한 심근 및 말초혈관 저항 항진은 말초 소동맥을 구성하고 있는 작은 장간막 동맥에 가장 큰 영향을 미치게 되므로 본 연구에서는 심근 기능이상과 더불어 장간막 동맥의 유전자 발현 이상도 살펴보았다. SHR 장간막 동맥의 RT-PCR 결과 정상혈압군에 비해 SR Ca2+-ATPase (SERCA3)는 mRNA level이 감소하였고, phospholamban은 증가되어 심근과 유사한 결과를 보였으며, IP3-receptor는 변화가 없었다. 이와 같은 결과는 고혈압과 관련하여 혈관내 평활근세포의 칼슘 조절기능 이상이 전체적인 peripheral resistance를 증가시키는 기전 중의 하나임을 보여준다.
고혈압 쥐에서의 실험결과는 심근과 장간막 동맥과 같은 작은 혈관평활근에서 근이완과 수축이 감소되었으며, 이는 칼슘조절 단백질들의 발현 및 기능 이상과 밀접하게 관련되어있음을 알 수 있다.

We have investigated the alterations in the expression levels and functional changes of sarcoplasmic reticulum(SR) Ca²+ regulatory proteins in heart and mesenteric arteries from spontaneously hypertensive rats(SHR). SHR(systolic BP, 202 ± 7.9 mmHg) were used as a model of hypertension and age-matched Wistar-Kyoto (WKY) rats(systolic BP, 139 ± 3.9) were served as a normotensive control. The relative expression levels of mRNA and protein regulating the Ca²+ transport(Ca²+ uptake and release) were determined by the competitive RT-PCR using the gene-specific oligonucleotide primers and western blot analysis, respectively. There was an inverse relationship on the changes of expression levels of SR Ca²+ - ATPase and phospholamban. SR Ca2+-ATPase mRNA and protein levels were decreased, whereas the expression levels of phospholamban were increased in SHR hearts. Ca²+ uptake rates of cardiac SR from SHR were compared to those obtained from the normotensive WKY rats. Maximum rates of Ca²+ uptake were decreased in the SHR. Furthermore, the mRNA level and [3H] ryanodine binding of the ryanodine receptor, a major factor in determining cardiac contraction rate, were decreased in SHR hearts. In SHR mesenteric arteries, mRNA levels of SR Ca2+-ATPase were decreased, while those of phospholamban were increased. But, the mRNA level of IP3 receptor showed no changes. These results may provide an explanation for the decreased function of the relaxation and contraction in cardiac and mesenteric arterial smooth muscle, and for the elevated total peripheral resistance associated with hypertensive rats.
