Identification and Functional Analysis of cis-Regulatory Elements Controlling Nonhair Cell-Position Preferential Expression of the WEREWOLF in Arabidopsis thaliana Kim, Yo Han Advisor: Prof. Song, Sang Kee, Ph.D. Department of Integrative Biological Sciences, Graduate School of Chosun University Tissue-specific gene expression in plants plays a fundamental role in regulating cell differentiation and organ development. Understanding the functions of cis-regulatory elements that mediate such expression is therefore essential for elucidating the molecular mechanisms underlying transcriptional regulation. In the model plant Arabidopsis thaliana, root epidermal cells differentiate into either hair cells (H-cell) or non-hair cells (N-cells) depending on their positional relationship with the underlying cortical cells. This positional differentiation is governed by a complex gene regulatory network. Among the key regulators, the R2R3-MYB transcription factor WEREWOLF (WER) plays a central role in root epidermal cell fate determination by regulating the expression of downstream genes such as GLABRA2 (GL2) and CAPRICE (CPC), thereby promoting the formation of N-cells. In addition, the WER promoter is known to be preferentially expressed in epidermal cells located at the N-position as well as in the lateral root cap in Arabidopsis roots. Previous studies have analyzed the T-DNA insertion sites and transcriptional activity of WER alleles and examined the expression pattern of WER through genetic crosses with the GL2p:GUS reporter line, thereby confirming the positional pattern of N-cell differentiation. These studies also suggested the minimal promoter region required for N-position-specific expression and highlighted the importance of the WER terminator in regulating WER transcription. Based on these findings, the present study aimed to identify the cis-regulatory elements responsible for N-position preferential expression of the WER promoter. To identify the promoter regions responsible for positional expression, a series of progressive promoter deletions and internal substitution constructs were generated and analyzed using GUS reporter assays. These analyses revealed that the region between -420 and -346 bp upstream of the translation start site represents a critical promoter region required for N-position preferential expression. In particular, the region between −405 and −361 bp was identified as a functionally important distal promoter interval contributing to preferential WER expression. Further functional analyses using synthetic promoter constructs containing tandem repeats of four candidate core sequences (-420/-391, -405/-376, -390/-361, and -375/-346 bp) demonstrated that the WER promoter contains at least two N-position-specific expression elements (NE1 and NE2) that mediate preferential expression in N-position epidermal cells, as well as a root cap expression element (RCE) involved in root cap-specific expression. In addition, to identify promoter regions involved in basal transcriptional activity of the WER promoter in the presence of the WER terminator (TWER), overlapping deletion constructs were generated and analyzed for GUS expression. The results indicated that removal of specific promoter segments results in the loss of reporter gene expression, suggesting that this region plays an important role in basal transcriptional regulation in combination with the TWER sequence. Furthermore, constructs containing TWER and WER genomic DNA were introduced into the wer mutant background, and complementation analyses were performed by crossing these lines with the GL2p:GUS reporter. These experiments demonstrated that even a very low level of WER expression was sufficient to effectively restore the wer mutant phenotype. Finally, to investigate the evolutionary conservation of the regulatory elements identified in the WER promoter, promoter sequences of WER-like (WRL) genes from Brassicaceae species were comparatively analyzed. Sequence alignment revealed that key promoter regions associated with N-position preferential expression are partially conserved among selected Brassicaceae species. These findings suggest that the cis-regulatory mechanisms controlling positional gene expression in root epidermal cells may be evolutionarily conserved within the Brassicaceae lineage. Taken together, this study functionally identified cis-regulatory elements that regulate positional expression of the WER gene in the root epidermis of Arabidopsis thaliana. Furthermore, the results suggest that these regulatory elements cooperatively contribute to the establishment of N-position specific gene expression. These findings provide new insights into the regulatory architecture underlying positional gene expression and contribute to a better understanding of the transcriptional mechanisms governing root epidermal cell fate determination in plants.
