The rate of environmental DNA (eDNA) degradation in aquatic environments has been extensively studied across various academic disciplines, including ecology, environmental science, and molecular biology. Tracking the eDNA degradation process reveals how biological traces disappear over time in water, allowing inferences about when and where specific organisms were present. Research on human-derived DNA in aquatic systems remains limited. However, mechanisms identified in ecological studies—such as the effects of temperature, pH, ultraviolet (UV) radiation, salinity, and microbial activity— offer important insights into potential forensic applications. The detection of human eDNA holds promise for forensic contexts such as missing person searches and submerged body recovery, highlighting the need for further research in this emerging field. This review addresses this need by comprehensively examining published studies on eDNA degradation in aquatic organisms and summarizing the environmental factors that influence degradation rates. Key factors include temperature, pH, UV radiation, and bacterial activity, which are known to affect degradation patterns primarily through complex interactions rather than independent effects. Additional studies have also identified dissolved oxygen, biomass, and DNA fragment size as contributing variables. By synthesizing existing findings from an environmental perspective, this paper aims to establish a conceptual foundation for future studies on human eDNA detection in aquatic environments and to provide a scientific basis for future forensic applications. In particular, this review organizes environmental influences into three mechanistic categories—chemical, biological, and physical—to provide a structured framework for interpreting human-derived eDNA signals in aquatic forensic investigations. This mechanistic classification not only synthesizes environmental degradation pathways but also provides a practical interpretive foundation for evaluating human-derived eDNA in forensic investigations involving aquatic environments.
