    알레르기 질환은 전 세계적으로 높은 유병률을 나타내는 대표적인 만성 염증성 질환으로, 아토피 피부염, 천식, 알레르기 비염 및 아나필락시스 등을 포함한다. 이러한 질환들은 환자의 삶의 질을 현저히 저하시킬 뿐만 아니라 사회·경제적 부담을 크게 증가시키고 있으나, 현재 사용되고 있는 항히스타민제, 스테로이드 및 생물학적 제제를 포함한 치료법은 장기 사용에 따른 부작용과 제한적인 치료 효능 등의 한계를 가지고 있다. 따라서 알레르기 염증 반응을 보다 근본적으로 조절할 수 있는 새로운 치료 전략이 요구되고 있다. 비만세포는 IgE 수용체인 FcεRI-매개 탈과립 반응과 염증성 매개 물질 분비를 통해 알레르기 반응을 유도하는 핵심 면역세포로 알려져 있다. 특히, FcεRI 활성에 의한 Lyn, Fyn 및 Syk 중심의 초기 신호전달 경로는 비교적 잘 규명되어 있으나, 지속적인 염증 반응에 관여하는 비만세포 활성 조절 기전에 대해서는 아직 충분히 알려져 있지 않다. 이에 본 연구에서는 식물유래 플라보노이드인 isobavachin의 항알레르기 효능과 작용 기전을 규명하고, 이를 기반으로 비만세포 활성 과정에서 Lck signaling의 조절 양상을 분석하여 알레르기 염증 조절을 위한 새로운 신호전달 기반 치료 전략의 가능성을 제시하고자 하였다. 먼저, IgE/Ag로 활성화된 비만세포와 수동피부아나필락시스(passive cutaneous anaphylaxis, PCA) 모델에서 isobavachin은 β-hexosaminidase 분비, PGD2 및 LTC4 생성, 그리고 염증성 cytokine 발현을 유의적으로 억제하였다. 또한, isobavachin은 SHP-1 활성 증가를 통해 FcεRI-매개의 Fyn, Lyn, Syk 및 Lck 활성화를 억제여 비만세포 활성을 억제하였다. 다음으로, DNCB와 house dust mite extract를 이용한 아토피 피부염 동물모델에서 isobavachin의 경구 투여는 피부 병변, 귀 두께 증가, 염증세포 침윤 및 비만세포 축적을 유의적으로 감소시켰다. 또한 혈청 IgE 수준과 림프절 비대를 감소시켰으며, Th2 관련 cytokine 발현을 억제하였다. 아울러 피부조직 및 배액림프절에서 Lck signaling의 조절과 연관된 면역조절 효과를 확인하였다. 마지막으로, IgE/Ag에 의한 비만세포 활성에서 Lck 조절 기전을 분석한 결과, Lck는 Syk과 비교하여 보다 지속적인 활성 양상을 나타내었으며, 비만세포의 지속적인 염증 반응 조절에 관여할 가능성을 확인하였다. 또한 ERK 및 SHP-1과의 상호작용을 통해서 Lck 활성이 조절을 확인하였으며, TAOK3 결핍은 Lck signaling과 비만세포 활성을 감소시켰다. 이러한 결과는 TAOK3가 SHP-1–Lck 조절축과 연관되어 비만세포 활성 조절에 관여할 가능성을 시사한다. 종합하면, 본 연구는 isobavachin의 항알레르기 및 항아토피 효능과 그 작용 기전을 규명하였으며, 비만세포 활성화를 매개하는 중심 신호물질인 Lck 조절기전을 확인하였다. 나아가 Lck signaling의 조절이 알레르기 염증 반응을 제어하기 위한 새로운 치료 표적 및 신호전달 기반 치료 전략으로 활용될 수 있음을 시사하였다.

    Allergic diseases are among the most common chronic inflammatory disorders, affecting more than 30% of the global population , and include atopic dermatitis, asthma, allergic rhinitis, and anaphylaxis. Although current therapeutic approaches, including antihistamines, corticosteroids, and biologic agents, are widely used, their clinical application remains limited by adverse effects associated with long-term use and by insufficient therapeutic efficacy. Therefore, the development of novel therapeutic strategies that more effectively regulate allergic inflammatory responses is warranted. Mast cells are key effector cells that initiate allergic reactions through FcεRI-mediated degranulation and the release of inflammatory mediators. While the early signaling events mediated by Lyn, Fyn, and spleen tyrosine kinase (Syk) have been extensively characterized, the regulatory mechanisms responsible for sustaining mast cell activation and allertic inflammatory responses remain incompletely understood. The present study investigated the anti-allergic effects and underlying mechanisms of the plant-derived flavonoid isobavachin and, based on these findings, characterized the regulation of proximal kinase signaling including Lck, during mast cell activation, to identify potential therapeutic targets for allergic diseases. First, in IgE/Ag-stimulated mast cells and a passive cutaneous anaphylaxis (PCA) mouse model, isobavachin significantly suppressed β-hexosaminidase release, prostaglandin D2 (PGD2) and leukotriene C4 (LTC4) production, and inflammatory cytokine expression. Mechanistically, isobavachin reduced the phosphorylation of Fyn, Lyn, Syk, and Lck during FcεRI-mediated signaling and increased SHP-1 activation. Genetic knockdown of SHP-1 reversed these effects, indicating that the inhibitory action of isobavachin is mediated, at least in part, through SHP-1-dependent negative regulation of these proximal kinases. Next, in an atopic dermatitis mouse model induced by DNCB and house dust mite extract, oral administration of isobavachin markedly ameliorated skin lesions and significantly reduced ear thickness, inflammatory cell infiltration, and mast cell accumulation, with therapeutic efficacy comparable to that of the JAK inhibitor tofacitinib. Isobavachin also suppressed Th2- and Th1-associated cytokine expression and reduced Lck phosphorylation in inflammatory cells from both mast cell- and T cell-containing tissues, indicating that Lck-associated signaling is modulated across multiple immune cell types involved in allergic inflammation. Finally, comparative analysis of Lck and classical signaling molecules revealed that, unlike Syk, Lck exhibited a relatively sustained activation profile and was associated with ERK- and SHP-1-dependent regulatory mechanisms. Furthermore, TAOK3 deficiency attenuated Lck signaling and impaired mast cell activation, suggesting. that a TAOK3-SHP-1-Lck signaling axis represents a novel mechanism governing sustained mast cell activation and allergic inflammatory responses of mast cells. Taken together, these findings demonstrates the anti-allergic effects of isobavachin and suggests that modulation of SHP-1-dependent proximal kinase signaling, including Lck-signaling pathway, may represent a promising therapeutic strategy for allergic inflammatory diseases.
