장내 미생물군 (gut microbiota)은 신경전달물질 생합성, 효소 활성 조절, 비타민 항상성 유지 및 약물 대사 등 다양한 숙주 대사 과정에 깊이 관여하는 것으로 알려져 있다. 그러나 장내 미생물과 숙주 간 상호작용에 대한 정량적 분석과 미생물 유래 대사산물의 구체적인 기여도는 아직 명확히 규명되지 않아, 이들의 건강 및 질병 관련 기전적 역할에 대한 이해에 한계가 존재한다. 본 연구에서는 실시간 이차원 핵자기공명 (2D NMR) 분석과 생체 내 공생 모델을 통합하여, 장내 미생물 유래 대사산물의 동역학을 정량적으로 분석하고, 특히 파킨슨병 (Parkinson’s Disease, PD)과 관련된 도파민 대사 및 신경생리학적 결과를 규명하고자 하였다.
모델 생물로 예쁜꼬마선충 (Caenorhabditis elegans)을 사용하여, 살아있는 세균 배양 시스템과 세균-숙주 공생 시스템 내에서 L-dopa 대사를 실시간으로 모니터링하였다. Enterococcus faecalis의 다양한 substrain은 tyrosine decarboxylase (TyrDC) 유전자를 공통적으로 보유하고 있음에도 불구하고, 생체 내에서 서로 다른 수준의 도파민을 생성하는 것으로 나타났다. 이러한 대사적 이질성은 alpha-synuclein 응집 및 운동 행동 장애 등 PD 병리와 관련된 주요 특징들과 상관관계를 보였다. 이 결과는, 단순히 대사 유전자의 존재나 발현만으로는 실제 대사 활성의 예측이 불가능하며, 생체 내에서의 직접적이고 정량적인 평가가 필요함을 강조한다.
장내 미생물 유래 비타민 B6의 활성형인 pyridoxal 5'-phosphate (PLP)가 숙주 도파민 조절에 미치는 영향을 분석하였다. 박테리아 유래 PLP 생합성 유전자 결손 균주를 이용한 결과, 박테리아 유래 PLP 감소가 도파민 항상성 저하, 효소 활성 장애 및 운동 기능 손상을 초래함을 확인하였다. 이러한 결함은 pyridoxal (PL) 보충을 통해 회복될 수 있었다. 마우스 모델에서도 PLP 결핍 박테리아의 군집화는 전신 비타민 B6 농도 감소, 흑질 (substantia nigra) 내 도파민성 뉴런의 기능 손상, 운동 성능 저하를 유발하여, 박테리아 PLP 생산이 장-뇌 축 (gut-brain axis)을 통해 신경생리에 직접적인 영향을 미친다는 사실을 입증하였다.
종합적으로, 본 연구는 미생물 유래 대사산물, 즉 도파민 및 PLP가 숙주 신경생리 조절에 핵심적으로 기여하며, 도파민 기능, 운동 행동, 신경퇴행성 질환 병태생리와 밀접하게 연결되어 있음을 규명하였다. 또한 본 연구는 숙주-장내 미생물 간 대사 상호작용을 정량적으로 분석할 수 있는 기반을 제공하고, 파킨슨병 및 관련 신경질환 치료를 위한 미생물 대사 활성 조절 전략의 가능성을 제시한다.

The gut microbiota exerts a profound influence on host metabolic processes, including neurotransmitter biosynthesis, enzymatic regulation, vitamin homeostasis, and drug metabolism. While the significance of these microbial-host interactions is increasingly recognized, quantitative analyses of the metabolic contributions from the microbiota remain poorly characterized, limiting a mechanistic understanding of their role in health and disease.
In this study, an integrated experimental framework combining real-time two-dimensional nuclear magnetic resonance (2D-NMR) spectroscopy and in vivo symbiotic models was established to investigate microbiota-derived metabolite dynamics, with a particular focus on dopamine metabolism and neurophysiological outcomes relevant to Parkinson’s disease (PD).
Using Caenorhabditis elegans as a host organism, L-dopa metabolism was monitored in both live bacterial cultures and bacteria-host symbiotic systems. Distinct substrains of Enterococcus faecalis, despite harboring the tyrosine decarboxylase (TyrDC) gene, exhibited substantial variability in their capacity to convert L-dopa into dopamine in vivo. This metabolic heterogeneity among genetically similar bacteria correlated with differential α-synuclein aggregation and motor behavioral impairments, hallmark features associated with PD pathology. The results reveal that the presence or transcriptional expression of metabolic genes alone is not predictive of functional metabolic output, highlighting the necessity for direct, quantitative assessment of microbial activity in living systems.
Additionally, the impact of microbiota-derived pyridoxal-5'-phosphate (PLP), the bioactive form of vitamin B6, on host dopaminergic regulation was examined. Using bacterial PLP biosynthesis knockout mutants, it was demonstrated that a reduction in microbiota-derived PLP impaired dopamine homeostasis, disrupted enzymatic function, and led to pronounced motor dysfunction in both C. elegans and mouse models. Pyridoxal (PL) supplementation effectively rescued these deficits, emphasizing the critical role of microbial vitamin B6 metabolism in supporting host neurophysiological integrity. In mouse models, colonization with PLP-deficient bacteria resulted in systemic vitamin B6 depletion, impaired dopaminergic neuron integrity in the substantia nigra, and motor performance deficits, thereby establishing a direct gut-brain metabolic axis involving bacterial PLP production.
Collectively, the findings of this study establish that microbiota-derived metabolites both dopamine and PLP act as key regulators of host neurophysiology, modulating dopaminergic function, motor behavior, and neurodegenerative disease pathology. This study provides a quantitative framework for dissecting host-microbiota metabolic interactions and suggests microbial metabolic activity as a potential therapeutic target for modulating disease progression in parkinson’s disease and related neurological disorders.
