MEMS(Micro-Electro-Mechanical Systems)는 리처드 파인만이 1959년 There´s plenty of Room at the Bottom 강연에서 제시한 미세 세계의 가능성으로부터 시작되어, 현재는 단순한 기계적 센서를 넘어 광학, 무선 통신등 다양한 기술에 응용되고 있다. 특히, 2024년 초에 일론 머스크가 세운 뉴럴링크가 침습형 BCI(brain computer interface)를 이용해 실제 환자의 뇌에 칩을 이식하고, 생각만으로 노트북 화면의 마우스 커서를 조작하는 임상적 성공을 발표하였다. 이는 BCI에 대한 기술이 대두되고 있음을 반증한다. BCI는 인간의 뇌 뿐만 아니라 다양한 신경신호들을 외부기기(컴퓨터 등)을 활용해 연결할 수 있는 인터페이스를 말한다. 뇌 신호를 해독하여 기기를 제어하거나, 반대로 외부 신호를 뇌로 전달할 수 있다. 이 때, 사용하는 3차원 전극 중 하나가 마이크로니들이다. 이러한 3차원 마이크로니들 중 특정 세포의 선택성을 높인 옵트로드는 광학적 자극을 통해 신경신호를 측정할 수 있는 장점 덕분에 세포에서부터 신경신호 네트워크까지 다양한 분야의 신경신호 실험에 응용될 수 있다.
본 논문에서는 다양한 신경신호 실험에 응용될 수 있는 LED가 결합 가능한 SU-8 기반 광 프로브 어레이를 제안하였다. 제안된 광 프로브 어레이는 3가지 세부요소로 구성되어 있다. 첫째, Through glass silicon via(TGSV)를 이용해 전기적으로 분리시킨 구조이다. 분리된 전극 구조를 통해 각 신경신호를 측정가능하다. 둘째, 유리재흘림 구조를 이용한 초소형 유리렌즈이다. 초소형 유리렌즈는 µm 높이를 갖는 렌즈로써, 렌즈 설계를 통해 렌즈의 기하학적인 모양과 초점거리를 예측할 수 있다. 마지막은 SU-8 기반 광 프로브이다. SU-8 기반 광 프로브 어레이 구조는 반도체공정 중 노광 공정을 이용해 유리기반 렌즈와 광 프로브 어레이 구조의 결합 효율을 극대화했다. 제안한 SU-8 기반 광프로브 어레이 구조는 반도체 공정 기반 중심이기 때문에 재현성 있게 제작가능할 뿐만 아니라 모놀리식(monolithic)한 구조로 설계해 하나의 칩에 제작할 수 있다.
제안된 SU-8 기반 광 프로브 어레이의 재현성 있는 구조 구현을 위해, 2가지 반도체공정 방법을 제안하였다. 첫째는, 유리 재흘림 기반 균열 없는 유리 렌즈 제작 방법이다. 유리 재흘림 공정시 반드시 수반되는 크리스토발라이트(cristobalite)는 광학적 특성 뿐만 아니라 물리적 특성을 저해시키는 요소이다. 따라서, 화학표면처리(Plasma surface treatment)를 통해 크리스토발라이트 생성을 최대한 억제해 균열 없는 초소형 유리 렌즈 구조가 제작되도록 하였다. 표면화학처리시 사용되는 공정 조건을 확립했으며, 이를 X선 광전자 분광법(X-ray photoemission spectroscopy)를 통해 유리표면의 원자의 조성을 정량적으로 검증하였다. 또한, 유리 재흘림 공정시 질소 분위기 및 상압 과 같은 다양한 대기 환경을 변수로 유리 재흘림 공정을 최적화 하였다(제작오차율 최대 약 4.1%). 둘째는 SU-8 기반 광 프로브 어레이 제작방법이다. SU-8 공정 시, 1 mm 이상 두께를 갖는 SU-8은 i-line(365 nm) 파장의 빛이 투과율이 급격히 감소하는 문제가 발생한다. 이를 해결하기 위해 g-line(405 nm) 파장의 빛을 이용한 공정 방법을 활용하였으며, 이를 기반으로 광 프로브 어레이를 제작하고, SU-8 사진공정방법(노광, baking, develop)을 최적화하였다.
제작된 SU-8 기반 광 프로브 어레이는 광학적, 기계적, 전기적 특성을 측정하여 측정 결과를 분석하였다. 광 프로브 어레이의 광학적 특성은 컴퓨터 시뮬레이션을 통해 광 결합 효율(-12.66 dB)을 해석하고, 이에 따른 측정 결과(-14.24 dB)를 비교했다. 특히, 470 nm 파장을 갖는 LED를 SU-8 기반 광 프로브 어레이와 결합하고, 광 프로브 어레이 끝 단의 집속되는 빛의 세기를 측정하였다. 측정된 빛 세기는 약 1 mW/mm2 로 세포자극에 필요한 임계값 이상의 빛세기를 확인하였다. 전기적 특성은 전기화학적 임피던스 측정을 통해 검증하였다. SU-8 기반 광 프로브 어레이는 16개 독립된 전극이 존재하며, 임피던스는 평균 약 0.287 MΩ 로, 신경신호측정시 요구되는 2 MΩ 이하를 만족하였다. 기계적 강도는 쥐 뇌를 모사한 아가로스겔(0.6 w/v%, 1.2 w/v%)에 삽입시 발생하는 힘을 측정함으로 검증하였다. 특히, 1개의 광 프로브 어레이를 아가로스겔에 약 8.33 µm/s 속도로 총 3회이상 반복적으로 삽입했을 때에도 구조적으로 변함이 없었다. 이를 바탕으로 광 프로브 어레이를 쥐 뇌에 삽입해 구조적 안정성을 확인하였다. 이러한 점을 종합하면, 제안된 SU-8 기반 광 프로브 어레이는 표면에서부터 심부영역까지 존재하는 다양한 신경신호 측정에 응용될 수 있는 가능성을 제시한다.

MEMS(Micro-Electro-Mechanical Systems) originated from the possibilities of the microscopic world presented by Richard Feynman in his 1959 lecture “There´s plenty of room at the bottom.” Nowadays, it extends beyond simple mechanical sensors to applications in diverse technologies such as optics and wireless communications. Notably, in early 2024, Neuralink, founded by Elon Musk, announced clinical success, implanting a chip into a patient's brain using an invasive BCI (brain computer interface) and enabling the patient to manipulate a mouse cursor on a laptop screen using only their thoughts. This demonstrates the rising prominence of BCI technology. BCI refers to an interface that connects not only the human brain but also various neural signals to external devices such as computers. It can decode brain signals to control devices, conversely, transmit external signals to the brain. One type of three-dimensional electrode used for this is the microneedle. Among these 3D microneedles, optrodes, which enhance selectivity for specific cells, can be applied to neural signal experiments across diverse fields from individual cells to neural signal networks thanks to their advantage of measuring neural signals through optical stimulation.
This dissertation proposes a SU-8 based optrode compatible with LED, applicable to various neural signal experiments. The proposed SU-8 based optrode consists of three components. First, it features an electrically isolated structure achieved using Through Glass Silicon Vias (TGSV). This isolated electrode structure enables the measurement of individual neural signals. Second, it incorporates an ultra-miniature glass lens using a thermal reflow. This ultra-miniature glass lens, with a height in the micrometer range, allows prediction of its geometric shape and focal length through lens design. Finally, it is a SU-8-based optrode. The SU-8-based optrode array structure maximizes the optical coupling efficiency between the glass lens and the SU-8-based optrode array is based on semiconductor processes, enabling reproducible fabrication. Furthermore, it is designed as a monolithic structure, allowing fabrication on a single chip.
To achieve reproducible structures for the proposed SU-8- based optrode, two fabrication process methods were proposed. The first is a crack-free glass lens fabrication process based on thermal reflow. Cristobalite, which inevitably forms during the thermal reflow, is a factor that impairs both optical and physical properties. Therefore, plasma surface treatment was employed to suppress cristobalite formation as much as possible, enabling the fabrication of crack-free, ultra-miniature glass lens structures. The process conditions used for surface plasma treatment were established, and the atomic composition of the glass surface was quantitatively verified using X-ray photoemission spectroscopy (XPS). Additionally, the thermal reflow process was optimized by varying atmospheric conditions such as nitrogen atmosphere and atmospheric pressure (maximum fabrication error rate of approximately 4.1%). The second method is for fabrication of SU-8-based optrode array. During SU-8-based optrode fabrication, SU-8 layers thicker than 1 mm exhibit a problem where light transmission at the i-line wavelength (365 nm) decreases sharply. To solve this, a fabrication process using light at the g-line wavelength (405 nm) was employed. Based on this, a SU-8-based optrode array was fabricated, and the SU-8 photolithography process (exposure, baking, development) was optimized.
The fabricated SU-8 based optrode was analyzed by measuring its optical, electrical, mechanical properties. The optical properties of the optrode array were interpreted through computer simulation to determine the optical coupling efficiency (-12.66 dB), and this was compared with the measured result (-14.24 dB). Specifically, an LED with a wavelength of 470 nm was coupled to the SU-8-based oprode array, and the intensity at the tip of the optrode array was measured. The measured light intensity was approximately 1 mW/mm2, confirming it exceeded the threshold intensity required for neural stimulation. The electrical properties were verified through electrochemical impedance measurements. The SU-8-based optorde array contains 16 independent electrodes, with an average impedance of approximately 0.287 MΩ, satisfying the requirement of less than 2 MΩ for neural signal measurement. Mechanical strength was verified by measuring the force generated when inserted into agarose gel (0.6 w/v%, 1.2 w/v%). Notably, even after repeatedly inserting a single optrode into agarose gel at speed of approximately 8.33 µm/s for a total of three times, no structural changes were observed. Based on this, the optrode was inserted into a mouse brain to confirm its structural stability.
Given these results, the proposed SU-8-based optrode array compatible with LED demonstrates the potential for application in measuring diverse neural signals across regions ranging from the surface to deep areas.
