    지적장애 (Intellectual disability, ID)는 다양한 원인에 의해 발생하는 이질적인 발달장애이며, 특히 상염색체 열성 지적장애 (Autosomal recessive intellectual disability, ARID)는 유전적 기여도가 높음에도 불구하고 그 발병 기전이 충분히 규명되지 않은 경우가 많다. ARID 관련 유전자 중 하나인 TUSC3는 N-당화 및 세포 내 마그네슘 (Mg²⁺) 항상성과 연된되어 있으나, 신경계에서의 정확한 기능과 ID 병리와의 연계성은 명확히 밝혀지지 않았다. 본 연구는 TUSC3 결손이 신경세포 및 신경회로에서 어떤 분자적 및 세포적 변화를 초래하는지 규명함으로써, ARID 발병 기전을 이해하기 위한 새로운 관점을 제시하고자 하였다.
    먼저, 실시간 Mg²⁺ 이미징과 분자 및 생화학적 분석을 통하여 TUSC3가 ER 내 Mg²⁺ 유입을 조절하는 핵심 인자임을 규명하였다. 최근 밝혀진 ER 내 Mg²⁺ 수송체인 ERMA (Endoplasmic reticulum magnesium transporter A)와 TUSC3가 상호 결합하여 ER 특이적인 Mg²⁺ 수송 모듈을 형성하며, TUSC3 결손은 기저 ER Mg²⁺ 농도를 감소시키고 Mg²⁺ 재흡수 능력을 저해하였다. 이러한 기능 이상은 신경모세포주 (SH-SY5Y), 일차 배양 신경세포, 생쥐 배아섬유아세포 (MEF), 그리고 TUSC3 이형접합 돌연변이를 보유한 ARID 환자 유래 섬유아세포에서도 공통적으로 관찰되어 보존된 기전임을 확인하였다.
    TUSC3의 역할을 생체 수준에서 규명하기 위하여 TUSC3 전신 결손 마우스를 제작하였다. 본 마우스에서는 배아기 부분 치사율 증가와 함께 체중 감소가 관찰되었으며, 생후 발달 과정 전반에 걸쳐 성장 저하를 보였다. 이러한 기초적 표현형을 바탕으로 TUSC3 결손이 신경발달에 미치는 영향을 분석하기 위해 행동학적 평가를 수행한 결과, 생후 3개월령에서 공간작업기억 (Y-maze), 대상인식기억 (NOR), 스트레스 대응능력, 사회적 상호작용 등 ARID 환자에서 대표적으로 손상되는 인지 및 적응 기능 영역에서 뚜렷한 결손이 확인되었다.
    TUSC3 결손이 유발하는 분자 및 세포 수준의 변화를 규명하기 위하여 뇌 조직 및 신경세포를 분석하였다. 그 결과, ER Mg²⁺ 감소가 선택적 ER 스트레스 반응을 유도하였으며, 특히 PERK–eIF2α 경로의 만성적 활성화, 전반적인 단백질 합성 감소, GRP78과 CHOP의 증가가 두드러졌다. 신경세포 수준에서는 시냅스 단백질 (PSD-93, PSD-95, GluA1)의 발현 감소, CREB 활성 저하, 가지돌기 수 및 스파인 밀도의 감소 등 구조적 및 기능적 이상이 관찰되었다. 전기생리학적 분석에서는 TUSC3 결손이 해마 회로에서 영역 특이적으로 작용함을 확인하였는데, CA1에서는 자발적 흥분성 시냅스전류 (sEPSC)의 진폭 증가와 유발전류 (eEPSC)의 진폭 감소가 나타난 반면, CA3에서는 sEPSC 빈도 감소가 관찰되어 서로 다른 형태의 시냅스 기능장애가 존재함을 보여주었다.
    중요하게도, 뇌 투과성이 높은 마그네슘 제제인 magnesium-L-threonate (MgT)를 장기간 투여했을 때 TUSC3 결손세포와 결손 마우스 모두에서 ER Mg²⁺ 농도 회복, ER 스트레스 완화, 시냅스 단백질 발현 정상화, 행동 결손 개선이 관찰되었다. 특히 성장 초기 (4주령)부터 MgT를 보충한 경우, 성체 시점에서도 인지기능과 적응행동이 유지되는 등 예방적 효과도 확인하였다. 구조 및 분자적 회복은 해마와 선조체에서 두드러졌으며, 전전두엽에서는 상대적으로 제한적이었다.

    Intellectual disability (ID) is a heterogeneous neurodevelopmental condition with substantial genetic contribution, yet the molecular pathways underlying many autosomal recessive forms (ARID) remain poorly understood. Among the reported ARID genes, Tumor Suppressor Candidate 3 (TUSC3) has been implicated in diverse cellular processes, including N-glycosylation and magnesium (Mg²⁺) homeostasis; however, its precise function in the nervous system and its relevance to cognitive impairment have remained unresolved. In this thesis, I investigate the cellular, molecular, and behavioral consequences of TUSC3 deficiency and reveal a previously uncharacterized mechanism linking ER Mg²⁺ regulation to neuronal function and ARID pathogenesis.
    Using a combination of live-cell Mg²⁺ imaging, molecular assays, and biochemical approaches, I demonstrate that TUSC3 plays an essential role in ER Mg²⁺ uptake by acting as a positive regulatory partner of ER magnesium transporter A (ERMA), a recently identified ER-localized P-type ATPase. Loss of TUSC3 markedly reduces basal ER Mg²⁺ levels and impairs ER Mg²⁺ reuptake following depletion. This defect is conserved across multiple cell types, including SH-SY5Y neuroblastoma cells, primary cortical neurons, mouse embryonic fibroblasts, and fibroblasts derived from ARID patients carrying biallelic TUSC3 mutations. Co-IP and domain-mapping analyses further reveal that TUSC3 interacts with ERMA through its TM1 region, forming an ER-resident Mg²⁺ transport module required for maintaining ER Mg²⁺ homeostasis.
    To confirm the impact of TUSC3 loss in vivo, I generated a TUSC3 knockout (KO) mouse model. TUSC3 KO mice exhibited partial embryonic lethality, reduced body weight, and progressive behavioral abnormalities. Although early developmental assessments revealed only subtle changes, three-month-old TUSC3 KO mice showed clear deficits in spatial working memory, recognition memory, stress-coping behavior, and social interaction—core domains typically affected in ARID. Molecular analyses of brain tissues further supported these findings, revealing reduced expression of synaptic markers and decreased CREB activation, particularly in the hippocampus and striatum, regions that are critically involved in learning, memory, and adaptive behaviors.
    At the cellular and circuit levels, ER Mg²⁺ deficiency in TUSC3-deficient cells triggered a selective unfolded protein response characterized by prominent activation of the PERK–eIF2α axis, reduced global translation, and upregulation of the ER stress markers GRP78 and CHOP. In neurons, this stress state led to diminished expression of key synaptic proteins (PSD-93, PSD-95, GluA1), impaired CREB signaling, and structural abnormalities, including reduced dendritic branching and spine density. Electrophysiological recordings revealed region-specific alterations in hippocampal circuits: CA1 neurons exhibited increased spontaneous EPSC amplitude and reduced evoked EPSCs, whereas CA3 neurons showed decreased spontaneous EPSC frequency, indicating a combination of pre- and postsynaptic dysfunction. Collectively, these findings link ER Mg²⁺ imbalance in TUSC3 deficiency to impaired synaptic maturation, altered neurotransmission, and the in vivo behavioral phenotypes observed in TUSC3 KO mice.
    Notably, I demonstrate that restoring Mg²⁺ availability through chronic administration of magnesium-L-threonate (MgT), a brain-penetrant magnesium compound, rescues ER Mg²⁺ levels, alleviates ER stress, restores synaptic protein expression, and significantly improves cognitive and behavioral deficits in TUSC3 KO mice. Preventive supplementation beginning at juvenile stages also preserves cognitive function and adaptive behavior into adulthood, indicating a window of therapeutic opportunity. Molecular rescue is region-specific: hippocampal and striatal deficits respond robustly to MgT, whereas the prefrontal cortex remains largely unchanged, suggesting differential vulnerability among neuronal circuits.
    In conclusion, these findings identify TUSC3 as a critical regulator of ER Mg²⁺ homeostasis through its interaction with ERMA and establish ER Mg²⁺ deficiency as a mechanistic driver of ER stress, impaired protein synthesis, synaptic dysfunction, and ARID-like phenotypes. This work provides the first direct evidence that ER Mg²⁺ stress contributes to cognitive impairment and highlights magnesium supplementation as a potential therapeutic strategy for TUSC3-related ARID. More broadly, these results suggest that dysregulation of ER Mg²⁺ may underlie additional neurodevelopmental disorders and emphasize the importance of organelle-specific ion regulation in neuronal function and brain development.
