    Myeloid cell leukemia-1 (MCL-1)—an anti-apoptotic protein of the B-cell lymphoma 2 family—is commonly overexpressed in human cancers, promoting tumorigenesis and chemoresistance. Upregulated MCL-1 in human hepatocellular carcinoma (HCC) has been demonstrated in numerous studies, and therapeutic agents targeting this protein have been assessed. However, its prognostic significance in canine HCC remains unclear. The objective of this study was to detect MCL-1 protein in canine normal liver tissue and compare its expression level with that in HCC tissue using western blotting. Immunohistochemistry (IHC) was used to quantify MCL-1 intensity levels in normal, non-neoplastic hepatic diseases, and HCC tissues, and the differences were assessed. Additionally, the relevance of MCL-1 immunostaining to various clinical and pathological parameters was evaluated. MCL-1 expression was markedly elevated in HCC tissues relative to normal liver tissues (P = 0.029). Additionally, all 10 normal liver tissues exhibited low IHC expression, which significantly increased as the malignancy progressed (P < 0.001). In the HCC samples, high MCL-1 immunostaining was substantially correlated with metastatic status (P = 0.034) and tumor size (P = 0.046). Moreover, survival curve analysis revealed a significant relationship between upregulated MCL-1 and lower disease-free survival and overall survival rate (P = 0.006 and P = 0.031, respectively). MCL-1 expression is increased in canine HCC, and its overexpression significantly correlates to worse clinical outcomes. Therefore, MCL-1 is considered to be a promising prognostic marker.

    Myeloid cell leukemia-1 (MCL-1)은 B-cell lymphoma 2(BCL-2) 계열에 속하는 항세포사멸 단백질로, 사람의 다양한 종양에서 과발현되어 종양의 발생과 항암제 내성에 관여하는 것으로 알려져 있다. 사람의 간세포성 암종(hepatocellular carcinoma, HCC)에서는 MCL-1의 발현 증가가 여러 연구에서 보고되었으며, 이를 표적으로 하는 치료제의 개발도 진행 중이다. 그러나 개에서의 MCL-1 발현 및 그 예후적 의의는 아직 명확히 규명되지 않았다. 본 연구에서는 정상 개 간조직과 간세포성 암종 조직에서의 MCL-1 단백질 발현을 Western blotting으로 비교하고, 면역조직화학염색을 통해 정상 간, 비종양성 간질병, 간세포성 암종 조직 간의 발현 정도를 정량적으로 분석하였다.
    그 결과, 간세포성 암종 조직에서 MCL-1 발현은 정상 간조직에 비해 유의하게 증가하였으며(P = 0.029), 면역조직화학염색 분석에서도 악성도가 높아질수록 발현이 뚜렷하게 증가하였다(P < 0.001). 특히 높은 MCL-1 발현은 전이 여부(P = 0.034)와 종양 크기(P = 0.046)와 유의한 연관성을 보였고, 생존 곡선 분석에서는 과발현된 경우 무병생존기간(disease-free survival)과 전체 생존기간(overall survival)이 유의하게 낮았다(P = 0.006 및 P = 0.031).
    이러한 결과는 MCL-1이 개의 간세포성 암종에서 발현이 증가함을 보여주며, 그 과발현이 불량한 예후와 밀접하게 연관되어 있음을 시사한다. 따라서 MCL-1은 개 간세포성 암종에서 유망한 예후 지표로 활용될 수 있을 것으로 판단된다.
