    내측전전두엽(medial prefrontal cortex)은 다양한 뇌 영역으로부터 정보를 받아들이고 통합하여 다시 다른 피질, 하위 영역으로 전달하는 중앙 제어 허브로서의 역할을 수행한다. 이러한 특성을 바탕으로 내측전전두엽은 학습, 기억, 인지 기능, 그리고 정서 상태 조절 등 다양한 행동 양상을 형성하는 데 기여한다. 특히 정서 상태 조절은 내측전전두엽이 수행하는 인지 과정과 밀접하게 상호 연관되어 있어, 특정 상황에서 내측전전두엽이 정서를 어떻게 조절하는지에 대한 기전적 이해가 중요하게 부각되고 있다.

    The medial prefrontal cortex (mPFC) functions as a central control hub, integrating inputs from diverse brain regions and relaying information to various cortical and subcortical areas. Owing to these characteristics, the mPFC contributes to learning, memory, cognitive function, and the regulation of emotional states. Of these functions, affective state regulation is particularly important, as mPFC-dependent cognitive processes are tightly linked to emotional state. This underscores the need to understand the mechanisms by which affective states are modulated in the mPFC under specific conditions.
    While neuromodulatory systems have been extensively studied as key regulators of mPFC activity, glial modulation has emerged as an equally important but less understood component. Glia are a core components of the central nervous system (CNS), comprising astrocytes, microglia, and oligodendrocytes. Glial cells play diverse roles, including supporting neurons, maintaining CNS homeostasis, and regulating neuronal activity and synaptic transmission. Although glia share certain modulatory properties with neuromodulators, they also exhibit unique regulatory aspects that complement neuromodulatory systems. Therefore, elucidating the mechanisms that regulate glial activity and the functional outcomes shaped by glial modulation is essential for understanding mPFC function and developing therapeutic strategies. Nevertheless, a comprehensive understanding of glial roles and functions remains incomplete.
    Microglia are non-neuronal innate immune cells in the CNS and generally play neuroprotective roles. Upon immune challenge, microglia become activated and secrete pro-inflammatory cytokines, leading to neuroinflammation. Extensive studies have focused on suppressing neuroinflammation, particularly in the mPFC, since affective disorders such as depression and anxiety stem from dysregulation of the mPFC. Nonetheless, the specific microglial mechanisms underlying these effects remain unclear. In Chapter 1, I aimed to investigate how microglia in the prelimbic cortex (PL), one of the subregions of the mPFC, are regulated in the context of neuroinflammation, and how they contribute to affective disorders. To this end, I employed CX3CR1GFP+/- transgenic mice to investigate microglial electrophysiological properties while preserving intact synaptic activity. In particular, I found that the voltage-gated potassium channel Kv1.3 is markedly upregulated in microglia following inflammation. Moreover, comprehensive analyses using histology, western blot, and ELISA demonstrated that microglial Kv1.3 plays a critical role in amplifying neuroinflammatory responses. Lastly, a battery of behavior assays further demonstrated that neuroinflammation induces anxio-depressive behaviors. Notably, I showed that chlorpromazine (CPZ), a typical antipsychotic, inhibits microglial Kv1.3, thereby attenuating neuroinflammation as well as the associated anxiety- and depression-like phenotypes. These findings highlight microglial Kv1.3 as a key regulator of neuroinflammation and reveal a novel therapeutic application of CPZ for inflammation-induced affective disorders.
    Astrocytes regulate a wide range of neural activities including K+ buffering, neurotransmitter clearance, synaptic formation and elimination, synaptic modulation, and neuronal synchronization. Moreover, astrocytes modulate neurons through multiple mechanisms under diverse pathological conditions. Among various pathological conditions, astrocytes have been implicated in pain modulation. Pain constitutes a major public and personal health burden, but is difficult to address since it is often accompanied by comorbid anxiety. Extensive studies have demonstrated how pain-induced anxiety emerges and which brain regions are profoundly involved in this symptom. However, there is still no clear evidence regarding the precise relationship between pain and anxiety. In Chapter 2, I examined whether astrocytes in the anterior cingulate cortex (ACC), another subregion of mPFC, modulate both pain and comorbid anxiety. To achieve this, I established a pain model using complete Freund’s adjuvant (CFA) to induce peripheral inflammation and successfully reproduced comorbid symptoms of pain and anxiety. I employed fiber photometry and chemogenetics, and found that astrocytes in the ACC exhibit robust activation under pain conditions, with their activity was both necessary and sufficient for generating pain hypersensitivity and anxiety-like behavior. Moreover, viral tracing revealed that the ACC contains two distinct neural populations: one projecting to the dorsomedial striatum (DMS), strongly contributing to pain sensitivity, and another projecting to the basolateral amygdala (BLA), which appears to be insensitive to both pain and anxiety. Electrophysiological analyses showed that astrocytes modulate these circuits through two distinct mechanisms. Notably, the pain-sensitive circuit, ACC-DMS pathway is significantly regulated by mGluR1. These findings uncover a mechanism by which multiple circuits across the mPFC subregions can be differentially regulated by astrocytes and provide new therapeutic insights into treating comorbid pain and anxiety.
    In conclusion, these studies highlight that glial cells in the mPFC significantly shape affective states, particularly under pathological conditions. I revealed previously unrecognized mechanisms, showing that CPZ inhibits microglial Kv1.3 to attenuate neuroinflammatory responses, and that astrocytes engage distinct circuits to regulate pain and comorbid affective disturbances. Collectively, these studies broaden our understanding of glial modulators and regulatory mechanisms, establishing glial modulation as a critical determinant of affective states governed by the mPFC.
