소뇌 회로는 의도된 동작과 감각 피드백을 조화시켜 정확한 운동 수행을 가능하게 한다. 전통적 이론은 등반섬유(CF)가 퍼킨지세포(PC)에 운동 오차를 부호화하여 병렬섬유-퍼킨지세포(parallel fiber-PC) 시냅스에서 장기적인 시냅스 가소성을 유도하고, 학습된 기억이 피질에서 전정핵으로 이전되어 영구 저장된다고 설명한다. 이러한 시냅스 기반 프레임워크는 전정안구반사(VOR)와 눈운동반사(OKR) 등 여러 적응적 행동을 잘 설명해 왔다. 그러나 최근 연구들은 소뇌 기억의 안정성과 지속성을 단순한 시냅스 가소성만으로 충분히 설명하기 어렵다는 점을 지적하고 있다. 실제로 소뇌에서는 다양한 형태의 가소성이 관찰되며, 특히 비시냅스성 가소성인 내재적 흥분성(intrinsic excitability, IE)의 변화가 기억 공고화에 핵심적 역할을 할 가능성이 제기된다. 더욱이 학습에서 필수 정보를 전달하는 CF 신호가 습득과 공고화에 각각 어떠한 기능적 기여를 하는지, 장기 기억 형성을 위해 소뇌 피질에서 핵으로의 기억 전이가 어떠한 기준또는 메커니즘에 의해 결정되는지, 그리고 PC의 흥분성 변화가 시스템 수준의 장기 저장을 어떻게 조절하는지는 여전히 명확하게 규명되지 않았다.
본 논문에서는 위 질문들에 답하기 위해 수행한 세 편의 상보적 연구를 보고한다. 첫째, OKR 학습의 서로 다른 시기에 CF 신호 전달을 시간특이적으로 광유전학적 억제함으로써 CF 신호가 초기 운동기억 획득에 미치는 영향을 규명하였다. CF 입력은 PC에서 복합스파이크(complex spike)를 유발하며, 이는 시냅스 변화를 지시하는 신호로 여겨져 왔다. 학습 중 오차 신호가 두드러지는 50분동안의 훈련 구간에서 CF 활성을 억제하면 OKR 이득(gain)의 학습 유도가 완전히 소실되었으나, 동일한 억제를 공고화 기간 또는 기억 회상 시기에 적용해도 장기기억의 생성,유지 및 기억 회수에는 유의한 영향이 없었다. 이는 CF-induced complex spike가 기억 획득에 필수적이나 기억 안정화에는 필요하지 않음을 직접적으로 보여주며, CF 의존적 학습과 CF 비의존적 공고화가 기전적으로 분리되어 있음을 시사한다.
둘째, 고용량의 소뇌 피질과 상대적으로 저장 용량이 작은 전정핵 사이의 기억 전이를 지배하는 계산적 편향-분산(bias-variance) 절충 모델을 제시하고 실험적으로 검증하였다. 계산 시뮬레이션은 오차 분산으로 정의되는 과제 난이도가 새로 형성된 기억이 전정핵으로 전이되는지 혹은 피질에 잔존하는지를 결정한다고 예측하였다. 다양한 소뇌 의존 과제에서 수행한 행동 실험은 모델 예측과 일치하여, 난이도가 낮은 단순 과제에서는 초기 공고화 동안 전정핵으로의 전이가 잘 일어나는 반면, 난이도가 높은 과제에서는 기억이 피질에 더 오래 머무르는 경향을 보였다. 이 규범적 원리는 기존의 기억 전이 이론을 보완하며 단계별 공고화를 정량적으로 설명을 가능하게 하는 새로운 해석 틀을 제시한다.
셋째, PC-IE의 동역학이 시스템 수준 공고화를 매게하는 핵심 기전임을 밝혔다. 학습 후 약 90분에 걸쳐 퍼킨지세포의 흥분성이 일시적으로 저하되는 현상을 확인하였으며, 이 자연스러운 흥분성 감소를 해당 기간에 광유전학적으로 억제(즉, 흥분성 인위적 증강)하면 장기기억 형성이 차단되었다. 반면 동일한 조작은 기저 상태의 비학습적 안구 수행에는 영향을 미치지 않아, PC의 내재적 흥분성이 적응적 공고화 단계에서 특이적인 기능을 수행함을 시사한다. 또한 PC-IE의 인위적 상승은 장기기억 저장에 중요한 내측 전정핵(MVN)의 소뇌 소엽 (cerebellar flocculus)-표적 신경원(FTNs)에서 관찰되는 학습 관련 흥분성 증가를 억제하였다. 종합하면, PC-IE의 정확한 시간적 동역학이 피질에서 핵으로의 정보 흐름을 조절함으로써 운동기억의 안정적 공고화를 가능하게 한다.
결론적으로, 본 연구들은 소뇌 학습·기억에 대한 통합적 3단계 모델을 제시한다. 시기적으로 정밀한 CF 신호가 오차 기반 획득을 주도하고, 과제 난이도에 따른 배분 메커니즘이 소뇌 피질과 핵 간의 기억 할당을 결정하며, 마지막으로, PC-IE의 시간적 조절이 시스템 수준의 공고화를 매개한다. 이 프레임워크는 세포 수준의 시냅스 및 내재적 가소성을 시스템 수준의 메커니즘과 연결하여 소뇌 기능 향상을 위한 구체적 표적을 제시한다.

Cerebellar circuits coordinate intended actions with sensory feedback to ensure accurate motor execution. Classic theories propose that climbing fiber (CF) inputs to Purkinje cells (PCs) encode motor errors, which facilitate long-term synaptic modifications at parallel fiber to PC (PF-PC) synapses and ultimately transfer learned memories to the vestibular nuclei for permanent storage. Although this synaptic plasticity framework explains numerous adaptive behaviors, such as the fine-tuning of reflex pathways like the vestibulo-ocular reflex (VOR) and the optokinetic reflex (OKR), thereby enabling rapid adjustments to sensory disruptions, recent studies indicate that it alone does not fully account for the stability and durability of cerebellar memories. Instead, various forms of plasticity are observed within the cerebellum. In particular, changes in intrinsic excitability (IE), a form of non-synaptic plasticity, may play a vital role in memory consolidation. Nevertheless, several critical questions remain unresolved: how CF signals differentially contribute to the processes of memory acquisition and consolidation, the mechanisms and criteria governing the transfer of memory traces from the cerebellar cortex to the nuclei, and how dynamic shifts in PC excitability regulate long-term storage at the system level.
This thesis presents three complementary studies that address these questions. First, I investigate how CF signaling contributes to the early acquisition of motor memory by applying temporally specific optogenetic suppression of CF transmission during different phases of OKR learning. CF input evokes complex spikes in PCs, which have long been proposed to act as instructive signals for synaptic modification. The suppression of CF activity during the 50-minute training session, when error signals are most prominent, completely abolishes the learning-induced increase in OKR gain. In contrast, identical suppression applied either during the consolidation period or during memory recall has no measurable effect on long-term retention or gain retrieval. These findings provide direct causal evidence that CF-evoked complex spikes are indispensable for memory acquisition but not required for memory stabilization, thus establishing a mechanistic dissociation between CF-dependent learning and CF-independent consolidation.
Second, I introduce and empirically validate a normative bias–variance trade-off model that governs memory transfer between the high-capacity cerebellar cortex and the lower-capacity vestibular nuclei. The computational simulations indicate that task difficulty, measured as error variance, influences whether newly formed memories consolidate in the nuclei or are retained in the cortex. Behavioral experiments with various cerebellum-dependent tasks confirm that simpler tasks are more likely to have their memory transferred to the nuclei during early memory consolidation, while relatively difficult tasks generally keep memory in the cortex for a more extended duration, aligning with my model predictions. This normative principle enhances classical theories of memory transfer and provides a quantitative framework for phase-dependent consolidation.
Third, I reveal that the dynamics of intrinsic excitability in PCs act as a crucial gating mechanism for systems consolidation. I found that PCs experience a temporary decrease in excitability during a significant 90-minute post-learning period. Preventing this natural decline by optogenetically enhancing PC-IE during this time blocked long-term memory formation, whereas similar interventions did not impact innate, basal ocular performance, underscoring a specific function for PC-IE during the adaptive phase of memory consolidation rather than a non-adaptive, general state of ocular reflex. Additionally, elevated PC-IE eliminated the learning-associated rise in intrinsic excitability that is typically observed in flocculus-targeting neurons (FTNs) of the medial vestibular nuclei (MVN), a critical region for storing long-term memories. Collectively, these findings illustrate that the exact timing of PC-IE dynamics is vital for cerebellar systems consolidation. By regulating the flow of information from the cortex to the nuclei, PC-IE ensures the robust stabilization of motor memories. This study emphasizes intrinsic plasticity as a delicate yet essential mechanism in memory consolidation.
In summary, these studies support a cohesive three-stage cerebellar learning and memory model. Precisely timed CF signals drive error-driven acquisition, a task-dependent mechanism allocates memory between cerebellar cortex and nuclei, and temporal regulation of PC-IE gates systems consolidation. This framework unifies synaptic and intrinsic plasticity mechanisms across cellular and systems levels and suggests concrete targets for enhancing cerebellar function.
