현대 사회에서 우울 및 불안과 같은 정서적 이상은 스트레스 노출뿐 아니라 장내 미생물총의 변화, 면역 조절의 붕괴, 생활양식 교란 등 다양한 생리적·환경적 요인과 밀접하게 상호작용하는 것으로 알려져 있다. 특히 장–면역–뇌 축(gut–immune–brain axis)을 매개로 장내 미생물의 변화가 정서 행동에 영향을 미칠 수 있다는 근거가 축적되면서, 특정 미생물 구성 성분이나 프로바이오틱스가 정신건강에 기여할 가능성이 주목받고 있다. 이러한 배경에서, 장내 미생물 교란과 면역 변화가 어떻게 행동 변화를 유발하는지에 대한 체계적인 비교 모델 연구가 필요하다. 본 연구에서는 만성 스트레스와 장내 미생물총 교란이 우울 유사 행동과 장관 면역 반응에 미치는 영향을 규명하고, Lactobacillus reuteri 보충이 이러한 변화를 완화할 수 있는지를 평가하였다. 먼저 만성 예측불가능 경미 스트레스(Chronic Unpredictable Mild Stress, CUMS) 모델을 구축한 결과, CUMS 처리군은 EPM에서의 불안 유사 행동 및 TST, FST에서의 우울 유사 행동 증가를 나타냈으며, 장관 IL-2, IL-6, IL-17A가 전반적으로 감소하였다. V3–V4 region 기반 미생물총 분석에서는 CUMS에 의해 장내 미생물 다양성이 변화하고 L. reuteri의 상대적 풍부도가 현저히 감소하여, 스트레스가 미생물 구성과 면역 환경을 동시에 변화시킨다는 점을 확인하였다.
이러한 변화를 재현하기 위해 항생제 기반(Abx) 모델을 구축하고자 하였고, 스트레스가 없는 환경에서 Amp 처리만으로 L. reuteri가 효과적으로 제거됨을 확인하였다. 또한 EPM의 불안 유사 행동 증가와 LH의 탈출 행동 감소 등 부분적인 우울 유사 행동이 관찰되었다. Abx 모델은 장관 IL-2, IL-6, IL-10, IL-17A 발현이 일관되게 감소하여, 장내 미생물 교란이 스트레스 없이도 면역 억제와 행동 변화를 유발할 수 있음을 시사하였다. 두 모델에서 공통적으로 L. reuteri 감소와 면역 억제가 동반된 점은, 장내 특정 균주의 변동이 전신 면역 및 행동에 영향을 줄 수 있는 가능성을 보여준다. 또한 BV-2 미세아교세포를 이용한 in vitro 스크리닝에서는, L. reuteri G1 균주가 LPS 유도 NO 생성과 LDH 분비를 가장 효과적으로 억제하여 뚜렷한 신경면역 조절 효과를 보였다. 이를 바탕으로 G1을 Abx 모델에 보충한 결과, 개별 행동 지표에서는 통계적 유의성이 제한적이었으나 PCA, PERMANOVA 기반 통합 분석에서는 Abx 군과 유의하게 분리되며 NC 방향으로 이동하는 회복 경향이 관찰되었다. 면역 지표에서도 G1 보충은 IL-6, IL-10, IL-17A 발현을 부분적으로 회복시켜 장관 면역 기능을 개선하는 데 기여하였다. 이러한 결과는 G1 균주가 미생물총 교란 이후의 면역·행동 변화에 복합적인 영향을 미칠 수 있음을 암시한다.
종합하면, 본 연구는 장내 미생물총 교란, 특히 L. reuteri의 소실이 스트레스와 독립적으로도 우울 유사 행동과 장관 면역 억제를 유발할 수 있음을 제시하며, L. reuteri 보충이 이러한 변화를 완화할 수 있는 psychobiotic 후보임을 확인하였다. 본 연구는 장–면역–뇌 축의 교란과 행동 변화 간의 연계를 모델 간 비교를 통해 정량적으로 제시하였다는 점에서 의의가 있으며, 향후 미생물 기반 기분 장애 관리 전략 개발의 기초 자료로 활용될 수 있다.

Mood disorders such as depression and anxiety in modern society are known to arise through close interactions not only with stress exposure but also with various physiological and environmental factors, including changes in the gut microbiota, impaired immune regulation, and lifestyle disruption. In particular, as evidence has accumulated that alterations in the gut microbiota can affect emotional behavior through the gut–immune–brain axis, the potential contribution of specific microbial components or probiotics to mental health has gained increasing attention. In this context, model-based studies are needed to understand how gut microbial disruption and immune changes lead to behavioral alterations.
In this study, we examined how chronic stress and gut microbiota disruption affect depression-like behavior and intestinal immune responses, and we evaluated whether supplementation with Lactobacillus reuteri could alleviate these changes. In the chronic unpredictable mild stress (CUMS) model, the CUMS group showed increased anxiety-like behavior in the elevated plus maze (EPM) and increased depression-like behavior in the tail suspension test (TST) and forced swim test (FST). Intestinal IL-2, IL-6, and IL-17A were consistently reduced. Microbiome analysis based on the V3–V4 region showed that CUMS altered gut microbial diversity and markedly decreased the relative abundance of L. reuteri, confirming that stress affects both microbial composition and the intestinal immune environment.
To reproduce these changes, we sought to establish an antibiotic-based (Abx) model, and we confirmed that ampicillin treatment alone effectively eliminated L. reuteri in the absence of stress. In addition, partial depression-like behaviors, including increased anxiety-like behavior in the EPM and reduced escape behavior in the learned helplessness test, were observed. The Abx model also showed consistent reductions in intestinal IL-2, IL-6, IL-10, and IL-17A, suggesting that gut microbiota disruption can induce immune suppression and behavioral changes without stress. The observation that both models showed reduced L. reuteri together with immune suppression indicates that changes in specific gut bacteria can affect systemic immunity and behavior.
We also performed in vitro screening using BV-2 microglial cells, and the L. reuteri G1 strain showed the strongest neuroimmune regulatory effect by suppressing LPS-induced NO production and LDH release. Based on these findings, we supplemented G1 in the Abx model, and although the statistical significance of individual behavioral indicators was limited, PCA- and PERMANOVA-based integrative analyses showed clear separation from the Abx group and a shift toward the NC group. G1 supplementation also restored the expression of IL-6, IL-10, and IL-17A, which contributed to improved intestinal immune function. These results suggest that the G1 strain may influence both immune and behavioral changes following microbiota disruption.
Taken together, this study shows that gut microbiota disruption, especially the loss of L. reuteri, can induce depression-like behavior and intestinal immune suppression independently of stress, and confirms that L. reuteri supplementation is a psychobiotic candidate capable of alleviating these changes. This study provides supportive evidence by quantitatively comparing multiple models linking gut– immune–brain axis disruption with behavioral changes, and it offers foundational insight for the development of microbiota-based strategies for managing mood disorders.
