    The elimination of major xenoantigens, including the Galα1–3Gal (α–Gal) epitope and the nonhuman sialic acid, N-glycolylneuraminic acid (Neu5Gc), is crucial to reduce xenogeneic immunogenicity and optimize recellularization for cardiovascular xenografts. In this study, we evaluated a dual-enzyme strategy using α–galactosidase to remove α–Gal xenoantigens and peptide N-glycosidase F (PNGase-F) to remove non–α–Gal xenoantigens, combined with optimized decellularization. Biomechanical testing and lectin staining confirmed that α-galactosidase and PNGase-F treatment markedly reduced lectin fluorescence while maintaining tissue integrity, demonstrating that the dual-enzyme treatment effectively and synergistically eliminated xenoantigens.
    Decellularized xenografts are promising scaffolds for cardiac valves; however, their clinical application remains limited owing to residual immunogenicity, poor regenerative capacity, and a tendency to degenerate. Several of these limitations could be overcome through optimized dual-enzyme treatment and subsequent xenograft tissue recellularization with allogenic mesenchymal stem cells (MSCs).
    For in vitro recellularization, human adipose-derived stem cells (hADSCs) were cocultured with human umbilical vein endothelial cells (HUVECs). The dual-enzyme–treated porcine pericardial scaffolds were further modified with fibrin-heparin-vascular endothelial growth factor (VEGF) coatings and then seeded with hADSCs and HUVECs, which accelerated recellularization. Immunofluorescence analysis of vimentin, fibronectin, calponin, CD31, and vWF demonstrated cell infiltration into the tissue, and progressive differentiation into endothelial and interstitial cells.
    Similarly, to address a clinically applicable recellularization strategy, we conducted recellularization using hADSCs monoculture. Although the progression was slower than in coculture, greater expression of vimentin, calponin and fibronectin indicated interstitial–like cell differentiation of MSCs and extracellular matrix (ECM) that facilitated cell adhesion and migration. Simultaneously, the expression of CD31 and vWF colocalized with phalloidin, indicating endothelial differentiation and cytoskeletal organization, thereby confirming successful recellularization.
    To our knowledge, this is the first describing the successful generation of major xenoantigen-free scaffolds through the synergistic effects of α-galactosidase and PNGase-F coenzyme treatment, validated for biocompatibility, reduced immunogenicity, and enhanced by hADSCs–based endothelial cell coculture and monoculture recellularization. This dual-enzyme and stem cell–based recellularization strategy effectively promotes functional repopulation, endothelial differentiation, offering a practical and scalable pathway toward next–generation tissue–engineered heart valves.

    시험관 내 재세포화를 위해 인간 지방유래 중간엽 줄기세포 (hADSCs)와 인간 제대정맥 내피세포(HUVECs)를 공배양하였다. 이중 효소 처리된 돼지 심낭 스캐폴드는 피브린–헤파린–VEGF 코팅으로 추가 개질한 후, hADSCs와 HUVECs를 파종한 결과 재세포화가 가속화되었다. 면역형광 염색(vimentin, fibronectin, calponin, CD31, vWF)를 통해 내피세포 및 간질세포 유사 표현형으로의 점진적인 분화를 확인하였다.
    유사하게 임상 적용 가능한 재세포화 전략을 확립하기 위해, 동일한 조건에서 hADSCs 단일배양을 통한 재세포화도 수행하였다. 공배양에 비해 진행속도는 느렸으나, 강한 vimentin과 fibronectin 발현이 관찰되어 간질세포 유사 분화와 ECM의 재구성이 이루어졌음을 확인하였다. 또한 CD31 및 vWF 발현이 phalloidin 공발현과 함께 관찰되어 내피세포 분화와 세포골격 형성됨을 입증 하였다.
